Methylmalonic acidemia and homocysteinemia cblX type (cblX) (MIM#309541) is a rare, X-linked recessive disorder caused by a mutation in the HCFC1 gene. This disorder is characterized by multiple congenital anomalies which include intellectual disability, brain malformations, intractable epilepsy, microcephaly, and facial dysmorphia. The most severe symptom of cblX is intractable epilepsy which is 100% penetrant. Our lab developed a zebrafish model with a mutation in the zebrafish hcfc1a ortholog to study the mechanisms underlying seizure phenotypes in cblX. Host Cell Factor C1 (HCFC1) is a transcriptional co-activator factor that regulates the expression of over 5000 different downstream target genes in human cells. Missense mutations in the HCFC1 gene cause methylmalonic acidemia homocysteinemia cblX type (cblX). cblX, a multiple congenital anomaly syndrome (MCA) characterized by abnormal brain development, craniofacial abnormalities, metabolic deficits, and intractable epilepsy. Published studies suggest that HCFC1 regulates neural precursor proliferation, number, and differentiation. However, these results were derived from studies that were performed with nonsense alleles or transient knockdown approaches. Methylmalonic aciduria and homocystinuria, CblC type (cblC) syndrome (MIM 277400) is a genetic disorder resulting from a mutation in the MMACHC gene. This gene plays a crucial role as a chaperone in the conversion of vitamin B12 into its active form, which is essential for proper cellular metabolism. cblC syndrome is heterogenous by nature, primarily attributed to the extensive damage it causes across multiple bodily systems. The GABRA1 gene encodes for the alpha-1 (α1) subunit of the Gamma-Aminobutyric acid type A receptor (GABAAR), which are the primary modulators of synaptic inhibition in the central nervous system (CNS). Alpha-1 subunits are essential for maintaining the normal function of native receptors and contribute to over 60% of all GABAARs in the CNS. Remarkably, a broad spectrum of neurodevelopmental and epilepsy-associated disorders have been linked with mutations in the GABRA1 gene. Neural precursor cells (NPCs) are the stem-like cells of the developing brain. These cells differentiate into the differentiated cell types of the central nervous system. If disruption occurs in the number of total NPCs formed or their survival, various disorders including learning disabilities, behavioral problems, or epilepsy can occur. This dissertation describes how the HCFC1 gene controls the proliferation and differentiation of NPCs. HCFC1 encodes for a transcriptional co-factor that regulates the growth and metabolism of stem cells, including NPCs. We present a patient diagnosed with a multiple congenital anomaly syndrome characterized by scoliosis, intellectual disabilities, and craniofacial defects. Trio-based whole exome sequencing identified compound heterozygous variants in the HSPG2 gene: a maternally inherited c.5998-7A>G variant in the promoter region and a paternally inherited c.4916C>T variant in the coding sequence. HSPG2 encodes for perlecan, a large proteoglycan that plays an important role in cartilage formation, cell adhesion, and basement membrane stability. Mutations in HSPG2 have been associated with Schwartz-Jampel syndrome and Dyssegmental Dysplasia Silverman-Handmaker Type, two disorders characterized by skeletal abnormalities (1). The African river frog genus Amietia is found near rivers and other lentic water sources throughout central, eastern and southern Africa. Because the genus includes multiple morphologically conservative species, taxonomic studies of river frogs have been limited. We sampled 49 individuals of Amietia from multiple localities in and near the Albertine Rift (AR) of Burundi, Democratic Republic of the Congo and Uganda. We utilized single-gene (16S) and concatenated (12S, 16S, cytochrome b and RAG1) gene-tree analyses and coalescent species-tree analyses to construct phylogenetic trees. African snake-eyed skinks are relatively small lizards of the genera Panaspis and Afroablepharus. The allocation of these genera was frequently rearranged in the 20th century based on morphology, ecology and biogeography. Members of these genera occur primarily in savanna habitats throughout sub-Saharan Africa and include species that have highly conserved morphology, which poses a challenge for taxonomic studies. We sequenced two mitochondrial (16S and cyt b) and two nuclear genes (PDC and RAG1) from 91 Panaspis and Afroablepharus samples from various localities in eastern, central and southern Africa. 1. Cryptic speciation in zooplankton is a phenomenon that has been recently gaining much attention. This is due in part to advances in molecular techniques which help in the identification of morphologically indistinct species. Comparison Of Ectoparasitic Mite Loads Between Gonochoristic (Aspidoscelis Marmorata) And Parthenogenic (A. Tesselata) Syntopic Whiptail Lizards (Teiidae) From The Northern Chihuahuan Desert Of Trans-Pecos, Texas, William Dawson Lukefahr

The objective of this study was to compare the ectoparasitic loads between two syntopic whiptail lizard species (Teiidae) that differ in their reproductive modes; Aspidoscelis marmorata (bisexual) and A. tesselata (unisexual). The study site was located on Indio Mountains Research Station (IMRS) in a Chihuahuan Desert Scrub landscape in Hudspeth County, Texas. Previous studies conducted at IMRS showed the two species having similar diets, morphology, and reproductive characteristics, but fluctuated in nematode loads. No comparative study of ecotoparasitic mites (Eutrombicula alfreddugesi) between the two species has ever been done.

Leptopelis, a genus of Central African treefrogs, includes 51 species that live in tropical forests and savannas. Currently, only two species of Leptopelis are known from the poorly explored Itombwe Plateau in eastern Democratic Republic of the Congo (DRC). Itombwe is renowned among conservationists for its rich and endemic amphibian fauna, including: Xenopus itombwensis, Chrysobatrachus cupreonitens, Laurentophryne parkeri, Hyperolius leleupi and at least three species of Arthroleptis. Evolutionary relationships of Itombwe Leptopelis were examined by sequencing two mitochondrial genes (16S: 557 bp [base pairs], cyt b: 620 bp) and one nuclear gene (RAG1: 761 bp).
