Altered patterns of methylation have been shown in both ribosomal and transfer RNA isolated from cancer cells. In contrast to the elevated activity of the methyltransferases responsible for this modification, hypomethylation appears as a general characteristic. The endogenous levels of S-adenosylmethionine and S-adenosylhomocysteine are important in relationship to their role as substrate and product of the transmethylation reaction. The levels of S-adenosylmethionine and S-adenosylhomocysteine were determined using phosphocellulose cation exchange or high pressure liquid chromatography. It has been previously demonstrated that 7,12-dimethylbenz(a) anthracene (DMBA) is an initiator of ovarian granulosa cell tumors. It is also known that DMBA leads to destruction of primordial oocytes, and that tumor induction may involve the interaction of pituitary gonadotropins. However, the effects of DMBA on ovarian steroidogenesis and uterine growth in relation to the tumorigenic process have not been investigated. Therefore, the present study was conducted to examine these parameters using adult C57Bl/6N mice treated in proestrus and diestrus II with 0.1 ml corn oil with or without 1.5 mg DMBA and killed 44 hours later. 5.8S rRNA is a low molecular weight ribosomal RNA which is found hydrogen bonded to 28S rRNA in the eucaryotic cell. There are two nucleotides which have 2'-0-methylations; a guanine residue at position 77 is fully methylated and a uridine residue at position 14 which is partially methylated. This partial 2'-0-methylation of the uridine residue has been found to vary with the tissue source, with the highest level in normal tissue and the lowest level in neoplastic tissue. In order to study the significance of this site-specific methylation, a reliable and convenient method of analysis was needed.

A two-dimensional polyacrylamide gel electrophoretic technique was developed for the detection of unique tumor-associated proteins in urine of patients with transitional cell carcinoma (TCC) of the urinary bladder. This technique yielded high resolution of polypeptides in the urine of cancer patients. Urine was chosen as a source of tumor-associated components because the location of the tum or should release already solublized tum or substances in to the urine.
Urinary proteins were concentrated and then analyzed by the two - dimensional electrophoretic technique using isoelectric focusing in the first dimension and sodium dodecylsulfate - polyacrylamide gel electrophoresis in the second dimension. …

Neurospora produces a phase specific, cationic mucopolysaccharide composed primarily of galactosamine (GAG-MP) which becomes part of the cell wall and is later secreted into the media. Its appearance coincides with the onset of the restricted phase of growth and causes efflux of small molecular weight metabolites when incubated with conidial cells. This activity may be a product of electrostatic interactions of the GAG-MP with the conidial plasma membrane. The activity is blocked by acetylation of the primary amines on the molecule, digestion by enzymes that hydrolyze carbohydrate linkages and the inclusion of NaCl (lM) in the GAG-MP/conidia reaction mixture. Sand G2 phase cells were exposed to 150 ergs mm⁻² UV and their ability to photoreactivate the induced cell killing (loss of colony forming ability) and chromosomal aberrations was determined as a function of time following the UV exposure. In S phase cells, the lesions leading to cell death and those leading to aberrations were both converted to a non-photoreactivable state shortly after the UV exposure. A significant fraction of the lesions induced in G2 cells, that led to cell death, were converted to a non-photoreactivable state before the progeny of the exposed cells reached the next succeeding S phase. …

Variously aged (C57BL X C3H)F1 hybrid mice were used as recipients in experiments to determine if spleen or bone marrow cells from normal C57BL or C3H donors, spleen or bone marrow cells from C57BL donors previously treated with cortisone, or C57BL bone marrow cells previously incubated with serum from X-irradiated C57BL mice could induce a graft-versus-host reaction (GVHR).
The mice were assayed for GVHR by mortality or splenomegaly as detected by the discriminant spleen assay developed by Simonsen. Results show that spleen cells from normal C57BL, normal C3H, or cortisone-treated C57BL mice are capable of inducing a GVHR in …

The ampullariid Pomacea urceus uses its ctenidium and lung in respiration. The snails reach the surface with their siphon and ventilate their lung by withdrawing their head-foot in a pumping action. The lung gas also serves to change the buoyancy of the snails. After a ventilation a snail does not necessarily return to the same buoyancy level. Nemerteans are common invertebrates along coastlines in both temperate and arctic climates throughout the world. Amphiphorus imparispinosus is a carnivorous member of this phylum that is found abundantly in the intertidal areas of the Oregon coast. As in many other invertebrate phyla, endocrine integration within the nemerteans is thought to be accomplished by neurosecretory mechanisms. In Amphiphorus the presence of these neurosecretory systems has not been previously shown. The dispersal and dispersion patterns of Hydra fusca were examined. Hydra were placed in petri dishes at various densities. The water in the dishes was swirled forcing the animals to the center. The location of each animal was marked at time intervals thereafter. Analyses of the dispersal rates and dispersion patterns were made. Hydra dispersed from a central release point at a non-random rate. There was rapid movement from the center followed by a minimal daily movement. This eventually resulted in a uniform dispersion pattern at high densities in a limited environment. There was a relationship between the ratio of …

Research dealing with the expression of human karyotype is of prime interest; but, closely rivaling that is the genetic study of domestic animals which are of economic importance to man.

Blastospore and mycelial cytoplasmic extracts from four strains of Candida albicans, varying in virulence, were compared by sodium dodecyl sulfate (SDS) polyacrylamide disc gel electrophoresis Cytoplasmic extracts were made by disrupting the cultures in a Braun homogenizer followed by ultracentrifugation. The supernatant fractions were then resolved on polyacrylamide gels containing SDS. Each of the preparations yielded a distinct pattern. There were mycelial specific and blastospore specific protein bands. There were more components resolved from the blastospore phase cytoplasmic extracts than from the mycelial phase cytoplasmic extracts. There were bands common to both phases. The external surfaces of pear sclereids commonly are illustrated as covered with apertures. This SEAA investigation of the surface features has shown the surface to have few or no apertures. When the primary wall layer was removed the typical ramiform canal system was obvious. This observation confirms the often-ignored fact that the pitapertures of the secondary wall are not continuous with the primary wall. Hence, they do not show on the surfaces of the intact cell.

The ultrastructure of abnormal meiotic cytokinesis in an autosomal male sterile mutant of Drosophila melanogaster is described. In the mutant ms(2)3R mitotic cytokinesis proceeds normally, but abnormal meiotic divisions give rise to four spermatids developing in one cytoplasmic mass. The contractile ring which is responsible for cell constriction during cytokinesis appears to form and function normally during meiosis. Ring canals which form intercellular bridges between synchronously developing spermatids are also apparently normal. However, there is an apparent adhesion and subsequent disintegration of the furrow membranes after division is completed. In addition to double membrane fragments in areas of open …

The mitotic apparatus in dividing cells of the ring-legged earwig is composed of the cell center or pole, chromosomal fibers, continuous fibers, and chromosomal and background structure. Parts from cellular elements such as Golgi membranes, endoplasmic reticulum, and nuclear envelope as well as whole mitochondria associate with the spindle microtubules to produce the diffraction pattern of the spindle fibers as seen with the light microscope. The microtubules of the chromosomal fibers attach independently and demonstrate the diffuse nature of the centromere or polycentric condition of the chromosomes.

Jackson and Patton stated that the key to understanding mechanisms of chromosomal evolution lies in studies of chromosomal variation within populations that can hybridize. Hybrids between cytotypes contain the answer to problems of meiotic pairing and reduced fertility, as well as information concerning the nature of zones of contact. However, before detailed studies can be initiated, chromosomal races and hybrid zones must be located. Few chromosomal races have been described for bats. Within the family Vespertilionidae, five species are known to have such races and races have been reported in two species of the family Phyllostomatidae.

The ry+ gene of Drosophila melanogaster and its associated enzyme, xanthine dehydrogenase, were employed in an analysis of the relationship between regulation and the location of a gene in the genome. Enzyme assays as an indication of gene activity were performed on genotypes containing zero, one and two doses of ry+ genes. Xanthine dehydrogenase activity of rosy genes in normal and relocated positions was determined during development.
The results indicate: 1) the gene ry+ is differentially active during ontogeny; 2) the activity during ontogeny is proportional to the number of ry+ genes present in the genome; …

Two bacteriophages which infect the myxobacterium Cytophaga succinicans, strains 14 and 16, were isolated from fish hatchery effluent. Each was submitted to a detailed study. These phages have a limited host range, even within the genus Cytophaga. The turbid plaques formed by these phages are similar in appearance to those produced by coliphage Lambda, and average 0.85 mm in diameter. Both phages have a latent period of 195 minutes and a rise period of an additional 165 minutes. Phage 0CS-16 has a burst size of 137 and phage 0CS-14 has a burst of 45. Fourteen recessive, male-sterile, autosomal mutants of Drosophila melanogaster were induced using ethyl methanesulfonate. The techniques employed restricted the recovery of mutants to those located on the second chromosome. Eleven of the mutants were male-sterile at 23°C, and the remaining three, though fertile at 23°C, were male-sterile at 28°C. Complementation tests involving all possible pairs of mutations showed each to be located in a unique cistron.
Spermiogenesis in each mutant was studied by light microscopy to determine the stage at which cellular differentiation deviates from normal and to characterize the associated gross abnormalities. Phase contrast microscopy showed that each mutant sterile …

An ultrastructural study of sperm differentiation was made on two male-sterile mutants of Drosophila melanogaster. The mutations were induced on the third chromosome with the monoalkylating agent ethyl methanesulfonate and were maintained in stock cultures over a balancer chromosome using a system of multiple inversions. Lipid is associated with all of the major serum protein fractions of both the mallard duck and man. however, whereas the ρ lipoproteins predominate in man with little lipo-albumin evident, according to available literature, in ducks the lipo-albumin constitutes more than half of the serum lipoproteins, and a substantial lipo-prealbumin fraction is evident. Most studies of lipo-proteins are carried out on human serum primarily for the purpose of diagnosis of abnormalities. Induction of Entner-Duodoroff pathway enzymes in Pseudomonas fluorescens was investigated to study the role of gluconate as a possible inducer. Glucose oxidase-deficient mutants were isolated and characterized. One of these mutants, gox-7, was deficient in particulate glucose oxidase; another mutant, gox-17, was deficient in particulate glucose and gluconate oxidase activities. Gluconate, but not glucose, induced synthesis of gluconokinase and 6-phosphogluconate dehydratase in both mutants. High constitute levels of 2-keto-3-deoxy-6-phosphogluconate aldolase were found when both mutants were grown on glucose. Growth of parent and both mutant strains on glycerol also resulted in high levels of Entner-Doudoroff pathway enzymes. It was concluded …

A cell has been designed and built that allows for rapid measurement of volume moved in a definite time by electroosmosis. The cell is simple to use and is not very elaborate. Using a water jacket, the cell temperature can be controlled to ± 0.1° C. Measurements are presented for acetonitrile, dimethylformamide, and nitrobenzene at 25° C for applied voltages of 25, 50, 75, and 100 volts.
