ในปัจจุบันมีทางเลือกที่สามารถทดแทนการใช้ปุ๋ยเคมีและสารกำจัดศัตรูพืชที่หลากหลาย โดยมีความปลอดภัยและเป็นมิตรต่อสิ่งแวดล้อม หนึ่งในทางเลือกที่ใช้คือการใช้ปุ๋ยชีวภาพที่ประกอบด้วยราอาบัสคูลาร์ไมคอร์ไรซาและแบคทีเรียที่มีประโยชน์ต่อพื้นที่เกษตรกรรม ในการศึกษาครั้งนี้มีวัตถุประสงค์เพื่อแยกและระบุชนิดของราอาบัสคูลาร์ไมคอร์ไรซาและแบคทีเรียที่ช่วยส่งเสริมความสัมพันธ์แบบไมคอร์ไรซา รวมไปถึงผลของการอยู่ร่วมกันของราอาบัสคูลาร์ไมคอร์ไรซาและแบคทีเรียต่อการเจริญเติบโตของต้นอ้อยพบ สปอร์ราอาบัสคูลาร์ไมคอร์ไรซา 18 ลักษณะที่แตกต่างกันที่ได้จากการแยกดินรอบรากอ้อยของไร่อ้อยในประเทศไทย โดยลักษณะสัณฐานวิทยาของสปอร์สามารถจำแนกแบ่งออกได้เป็น 3 วงศ์คือ Glomeraceae, Acaulosporaceae และ Gigasporaceae โดยวงศ์ Glomeraceae เป็นวงศ์ที่พบมากที่สุดในการศึกษาครั้งนี้ ราอาบัสคูลาร์ไมคอร์ไรซา AY01 และAT04 เป็นราอาบัสคูลาร์ไมคอร์ไรซาที่สามารถเพิ่มจำนวนสปอร์และมีเปอร์เซ็นต์การติดเชื้อในรากข้าวฟ่างสูงที่สุด เมื่อระบุชนิดด้วยเทคนิคทางอณูวิทยาและสัณฐานวิทยาของราอาบัสคูลาร์ไมคอร์ไรซา AY01 และAT04 พบว่าอยู่ในสกุล Diversispora นอกจากนี้สามารถแยกแบคทีเรียจากสปอร์ราอาบัสคูลาร์ไมคอร์ไรซาได้จำนวน 122 ไอโซเลท โดยพบ 20 ไอโซเลทมีลักษณะความสามารถในการส่งเสริมการเจริญเติบโตของพืช ได้แก่ลักษณะการสร้าง IAA สารในกลุ่ม siderophore แอมโมเนีย การสร้างไบโอฟิล์ม และมีความสามารถในการละลายฟอสเฟต โดยพบเพียงแบคทีเรีย ATR16 เท่านั้นที่สามารถกระตุ้นการงอกของสปอร์ราอาบัสคูลาร์ไมคอร์ไรซา AY01 และAT04 ได้เมื่อทำการระบุชนิดด้วยเทคนิคอณูวิทยาสามารถระบุได้ว่า ATR16 คือ Acinetobacter baumannii ในการทดลองเพื่อศึกษาการอยู่ร่วมกันของราอาบัสคูลาร์ไมคอร์ไรซา AY01 และAT04 กับแบคทีเรีย ATR16 โดยเก็บค่าความสูง มวลชีวภาพเหนือดินและใต้ดิน และเปอร์เซ็นต์การติดเชื้อในรากอ้อย จากชุดการทดลองที่มีการใช้ราอาบัสคูลาร์ไมคอร์ไรซา AY01 ร่วมกับแบคทีเรีย ATR16 พบว่า มีค่าต่าง ๆ สูงกว่าชุดควบคุมและมีเปอร์เซ็นต์การติดเชื้อสูงสุด (69.59%) ผลการทดลองเหล่านี้ชี้ให้เห็นว่าการใช้ราอาบัสคูลาร์ไมคอร์ไรซาร่วมกับแบคทีเรียที่มีลักษณะของการส่งเสริมการเจริญเติบโตของพืชสามารถส่งเสริมการเจริญเติบโตของอ้อยได้ ซึ่งการศึกษาครั้งนี้สามารถนำไปพัฒนาเป็นหัวเชื้อชีวภาพเพื่อนำไปใช้ในการเกษตรแบบยั่งยืน

ในการศึกษานี้มีการใช้ไซแลนที่สกัดได้จากวัสดุเหลือทิ้งทางการเกษตร (ซังข้าวโพด, ฟางข้าว, แกลบ, ชานอ้อย, และเปลือกถั่วลิสง) ในการผลิตฟิล์มและนำฟิล์มที่ได้ไปประยุกต์ในการต้านอนุมูลอิสระในเนื้อหมูแผ่นแห้ง โดยพบว่าซังข้าวโพดมีปริมาณเฮมิเซลลูโลสสูงสุดที่ร้อยละ 43.42±1.21 โดยน้ำหนัก ในขณะที่แกลบมีปริมาณไซแลนที่สกัดได้สูงสุด (ร้อยละ 12.00 ± 0.006 โดยน้ำหนัก) นำไซแลนที่สกัดได้มาดัดแปรด้วยกรดซิตริกที่อัตราส่วนแตกต่างกัน ได้แก่ 1:1 1:3 และ 1:5 รวมทั้งทำการวิเคราะห์โครงสร้างด้วยฟูเรียร์ทรานส์ฟอร์มอินฟราเรดสเปกโทรสโกปี ไซแลนที่สกัดได้และไซแลนที่ผ่านการดัดแปรจากวัสดุเหลือทิ้งทางการเกษตรถูกนำมาหล่อเป็นฟิล์มด้วยซีเอ็มซีร้อยละ 3 โดยน้ำหนักต่อปริมาตร และใช้กลีเซอรอลที่ความเข้มข้นแตกต่างกัน ได้แก่ ร้อยละ 0.5 1.0 และ 1.5 โดยปริมาตร พบว่า ความหนาของฟิล์มทุกชนิดไม่มีความแตกต่างกันอย่างมีนัยสำคัญ ในขณะที่ฟิล์มที่ถูกหล่อขึ้นจากไซแลนจากแกลบที่ถูกดัดแปรด้วยกรดซิตริก (M-XRH) ทุกอัตราส่วนร่วมกับกลีเซอรอลร้อยละ 0.5 (โดยปริมาตร) มีค่าความทนต่อแรงดึงสูงกว่าอย่างมีนัยสำคัญ (54.71-58.84 เมกะปาสคาล) เมื่อเปรียบเทียบกับไซแลนที่ผ่านการดัดแปรชนิดอื่น ดังนั้น ฟิล์ม M-XRH จึงถูกคัดเลือกเพื่อนำมาวิเคราะห์สมบัติทางกายภาพต่าง ๆ พบว่า M-XRH ที่ถูกดัดแปรด้วยกรดซิตริกที่อัตราส่วน 1:5 มีระดับการแทนที่ที่เกิดจากปฏิกิริยาเอสเทอร์ริฟิเคชันสูงสุดที่ 0.84±0.03 ในขณะที่ M-XRH ที่ถูกดัดแปรด้วยกรดซิตริกอัตราส่วน 1:3 มีค่าการซึมผ่านของออกซิเจน (41.6 ลูกบาศก์เซนติเมตรต่อตารางเมตรต่อวันต่อบาร์) และค่าการซึมผ่านของไอน้ำ (4.17±0.01 (x10-6) กรัมต่อเมตรต่อวินาทีต่อปาสคาล) สูงสุด เมื่อฟิล์ม M-XRH ที่ถูกดัดแปรด้วยกรดซิตริกทุกอัตราส่วนนำมาใช้กับเนื้อหมูแผ่นแห้ง พบว่า ค่าเปอร์ออกไซด์มีแนวโน้มการเกิดออกซิเดชันของไขมันช้าลง เมื่อเปรียบเทียบกับเนื้อหมูแผ่นแห้งชุดควบคุมที่ไม่มีฟิล์ม M-XRH ห่อหุ้ม ดังนั้นฟิล์ม M-XRH จึงเหมาะสมกับการนำมาประยุกต์ใช้กับผลิตภัณฑ์เนื้อสัตว์

Bee pollen is one of bee products. It is the mixture of flower pollen, nectar, and bee secretion. It is produced by workers in the genus of Apis. The major chemical compositions of bee pollen are protein, essential amino acids, sugar, fat, nucleic acid, and fiber. Minor chemical compositions are minerals, vitamins, saturated/unsaturated fatty acids, polyphenol, and flavonoid. These compounds make bee pollen exhibit a wide range of bioactivities including anti-inflammatory, antioxidant, antibacterial, and antifungal activities. In this dissertation, immunoassays on paper-based microfluidic devices have been developed using different types of label for various applications in agriculture safety and medical diagnosis, which can be divided into three parts. In the first part, the conjugation method of ractopamine (RAC) and protein carrier is developed for fabricating paper-based lateral flow strip test of RAC using gold nanoparticles (AuNPs) as a label. The developed conjugation method provided a simple and one-step method with the use of less chemicals. The strain MAL 1HM19, a nitrate reducing and sulfur oxidizing bacteria (NR-SOB) was successfully isolated from Mae Um Long Luang hot spring from Mae Hong Son province (Thailand) using hydrogen sulfide (H2S) and nitrate (NO3-) as an electron donor and acceptor, respectively. Among the isolates of NR-SOB from different sources, the strain MAL 1HM19 was the most promising novel strain of NR-SOB because of its ability to yield ~100% H2S removal, under anoxic conditions, within 5.5 h at an initial H2S concentration of 650 ppmv. New naphthalene derivatives (1 and 2) and a new isomer of ventilagolin (3), together with three known anthraquinones, including chrysophanol (4), physcin or emodin 3-methyl ether (5), and emodin (6), were isolated from vines of Ventilago denticulata. The isolated compounds exhibited cytotoxic activity against cancer cell lines (HuCCA-1, A549, HepG-2, MOLT-3) with IC50 values of 1.15-40.54 µg/mL. Compounds 1-3 exhibited weak antibacterial activity (MIC 200.0-400.0 µg/mL), while emodin (6) displayed moderate antibacterial activity with MIC 25.0 µg/mL. UV-A and -B blocking agents are essential components for the formulation of topical cream for psoriasis phototherapy. In this research, many natural compounds were discovered as potent UV-A and UV-B blocking agents, but small number could absorb narrowband UV-B. Quercetin hydrate, oxyresveratrol and curcumin were good UV-A blocking agents, while catechin, hesperetin, lutein and naringenin were proper for UV-B blocking agents. Berberine chloride hydrate, demethoxycurcumin and mansonone G were UV-A and -B blocking agents. โรคกระดูกพรุนเป็นปัญหาสุขภาพที่สำคัญอย่างหนึ่งของสังคมผู้สูงอายุ สาเหตุหลักของภาวะดังกล่าวเกิดจากความไม่สมดุลของเซลล์ที่ทำหน้าทีสร้างกระดูก (osteoblast) และเซลล์ที่ทำหน้าที่สลายกระดูก (osteoclast) การลดลงของมวลกระดูกในผู้สูงอายุนำมาซึ่งความเสี่ยงในการแตกหักของกระดูก จุดประสงค์ของการศึกษาครั้งนี้ คือ การคัดกรองสารออกฤทธิ์ทางชีวภาพที่มีฤทธิ์ในการต้านการเปลี่ยนสภาพของเซลล์สลายกระดูกและกลไกการออกฤทธิ์ของสารนั้นๆ ในกลุ่มสารทดสอบอนุพันธ์สาร (3S)-1-(3,4-dihydroxyphenyl)-3-hydroxy-7-phenyl-(6E)-6-heptene (DHPH) และ กรดไซเปอเรโนอิก (Cyperenoic acid) ซึ่งสกัดได้จากส่วนเหง้า Curcuma comosa Rox. และราก Croton crassifolius Geiseler. ตามลำดับ มีฤทธิ์แรงในการต้านการเกิดเซลล์สลายกระดูก ในระดับ in vitro ใช้เซลล์จากไขกระดูกที่ถูกกระตุ้นด้วย receptor activator of nuclear factor-κB ligand (RANKL) DHPH มีฤทธิ์ในการยับยั้งการเปลี่ยนสภาพไปเป็นเซลล์สลายกระดูก โดยยับยั้งการเกิด TRAP-positive multinucleated cells (TRAP+ MNCs) ที่ระดับความเข้มข้นยับยั้งกึ่งหนึ่ง (IC50) 325±1.37 นาโนโมลาร์ การยับยั้งดังกล่าวสอดคล้องกับการลดต่ำลงของสัญญาณภายใต้การกระตุ้นของ RANKL โดยรวมถึงวิถี Mitogen-activated protein kinases (MAPKs) เช่น ERK (p44/42) และ p38 แต่ไม่มีผลต่อวิถี NF-κB โดยผลของสารดังกล่าวยังส่งผลกระทบต่อระดับของ transcription factor เช่น nuclear factor of activation T cells (NFATc1) และ c-Fos ซึ่งมีความสำคัญมากในการเปลี่ยนสภาพเป็นเซลล์สลายกระดูก DHPH ลดการกัดกร่อนกระดูก (bone resorption) บนแผ่นกระดูก (bone slices) นอกจากนั้นสาร DHPH ยังสามารถเพิ่มกิจกรรมของเอนไซม์ alkaline phosphatase (ALP) ที่เกี่ยวข้องกับการเปลี่ยนสภาพของเซลล์สร้างกระดูกในเซลล์ชนิด MC3T3-E1 ในกรณีของกรดไซเปอเรโนอิกนั้น พบว่าสามารถลดการเกิด TRAP+ MNCs ที่ระดับความเข้มข้น IC50 ที่ 36.69±1.02 …

Using microalgae as a biofuel feedstock has played more attention over a decade. The cultivation of microalgae requires high water use and high initial investment that can make the process still not economically appealing. This study attempted to examine the potential for using brackish shrimp aquaculture effluents as culture medium for algal biomass production. วัตถุประสงค์ของงานวิจัยนี้คือเพื่อหาภาวะที่เหมาะสมในการผลิตบีตากลูแคนจาก Aureobasidium thailandense NRRL 58543 และศึกษาผลกระทบของน้ำหนักโมเลกุลบีตากลูแคนต่อคุณสมบัติพรีไบโอติก และประยุกต์ใช้ในการผลิตเยลลี ปริมาณการผลิตบีตากลูแคนสูงสุด (37.73±0.15 กรัมต่อลิตร) กับความขาวที่ยอมรับได้ (ดัชนีความขาว ≥ 48) ได้รับหลังจากที่เลี้ยง A. thailandense NRRL 58543 ในอาหารสูตร production medium โดยมีโซเดียมไนเตรท ร้อยละ 0.06 (โดยน้ำหนักต่อปริมาตร) เป็นแหล่งไนโตรเจน เสริมด้วยน้ำมันดอกทานตะวันร้อยละ 8.17 (โดยปริมาตรต่อปริมาตร) มีค่า pH เริ่มต้น 6.5 ภายใต้สภาวะการเขย่าที่ 150 รอบต่อนาที อุณหภูมิ 25 องศาเซลเซียส เป็นเวลา 4 วัน บีตากลูแคนที่ได้ถูกนำมาย่อยด้วยเอนไซม์บีตากลูแคนเนสทางการค้า ได้เป็นบีตากลูแคนไฮโดรไลเซทซึ่งประกอบด้วยน้ำตาลกลูโคสและโอลิโกแซ็กคาไรด์ มีน้ำหนักโมเลกุลเฉลี่ย 77,720 ดาลตัน ผลได้ของการย่อยบีตากลูแคนไฮโดรไลเซทด้วยน้ำย่อยเทียมเป็นเวลา 4 ชั่วโมง คือร้อยละ 26.51 ขณะที่บีตากลูแคนดั้งเดิมทนต่อการย่อยได้ นอกจากนั้นยังพบว่าบีตากลูแคนดั้งเดิมที่ 2 มิลลิกรัม คาร์บอน/มิลลิลิตร สามารถกระตุ้นการเติบโตของ Lactobacillus casei และ L. brevis ได้มากกว่า 1.8 เท่า เมื่อเทียบกับบีตากลูแคนไฮโดรไลเซท ภายหลังจากที่บ่มเป็นเวลา 48 ชั่วโมง บีตากลูแคนดั้งเดิมถูกนำมาใช้เป็นส่วนผสมสำหรับการผลิตเยลลี เมื่อนำบีตากลูแคนดั้งเดิมที่ความเข้มข้นสูงสุดร้อยละ 0.5 (โดยน้ำหนักต่อปริมาตร) ผสมร่วมกับเยลลี พบว่าเป็นที่ยอมรับทางประสาทสัมผัสที่ประกอบด้วย สี ความยืดหยุ่น ความเหนียว และความชอบโดยรวม จากผู้บริโภคจำนวน 30 คน เยลลีพรีไบโอติกที่ผลิตได้นี้สามารถเก็บรักษาที่อุณหภูมิห้องได้นาน 7 วัน

ความสม่ำเสมอของขนาดซากไก่เป็นปัจจัยหนึ่งของการเกิดปัญหาไก่ไส้แตก ซึ่งปัญหานี้มักพบในขั้นตอนการนำเอาอวัยวะภายในออก เมื่อเกิดปัญหาไก่ไส้แตกพนักงานในโรงงานผลิตไก่ต้องเสียเวลาทำความสะอาด และเสียเวลา จึงทำให้ต้นทุนการผลิตเนื้อไก่สูงขึ้น โดยปรกติขนาดของซากไก่ถูกควบคุม ด้วยน้ำหนักและอายุของไก่ แต่ในความเป็นจริง ขนาดไก่ มีความสัมพันธ์กับน้ำหนักและอายุ เพียงเล็กน้อย งานวิจัยนี้จึงนำเสนอวิธีการตรวจสอบความสม่ำเสมอของซากไก่ ด้วยวิธีการประมวลผลภาพดิจิทัล โดยงานวิจัยนี้แบ่งออกเป็นสองส่วน ได้แก่ ส่วนแรกนำเสนอวิธีการตัดแบ่งส่วนซากไก่ซึ่งเกิดจากการประมวลผลร่วมกันสามวิธีการ ได้แก่ การใช้แบ่งส่วนภาพด้วยสีการใช้ฟิลเตอร์โครงสร้างส่วนประกอบ และการใช้สัณฐานวิทยาของภาพ โดย งานวิจัยนี้ใช้ 5 ตัวชี้วัด เพื่อเปรียบเทียบประสิทธิภาพของกระบวนการ และงานวิจัยส่วนที่สองเป็นการนำเสนอวิธีการตรวจและสังเกตความสม่ำเสมอของขนาดซากไก่ หลังจากที่ภาพไก่ถูกตัดแยกด้วยกระบวนการแยกภาพซากไก่จากการวิจัยส่วนแรก ซึ่งงานวิจัยนี้ได้ทดสอบวิธีการนำเสอน 4 วิธีการ ซึ่งพบว่า วิธีการที่อยู่บนพื้นฐานของลายเซ็นภาพดิจิทัลทีมีการกำหนด ช่วงขอบเขตการยอมรับ เป็นวิธีการที่เหมาะสมที่สุด สำหรับการตรวจสอบความสม่ำเสมอของภาพซากไก่

โปรตีนสกัดเข้มข้นในใบ (leaf protein concentrate, LPC) เป็นโปรตีนสำคัญที่ใช้เป็นส่วนผสมของอาหารมังสวิรัติ และอาหารเสริม ทั้งในมนุษย์และสัตว์ โดยส่วนใหญ่ LPC ผลิตจากผักและธัญพืช เช่น ถั่วลิสง มันสำปะหลัง และผักบุ้ง ขณะที่ปริมาณรายงานการผลิตโปรตีนจากพืชน้ำมีน้อยโดยเฉพาะจากแหน (duckweed) แหนเป็นพืชน้ำที่โตเร็วและมีปริมาณไนโตรเจนอินทรีย์สูง ซึ่งงานวิจัยครั้งนี้ศีกษาปริมาณโปรตีนทั้งหมด (total protein content, TPC) ในแหนที่พบในประเทศไทย 4 ชนิด ได้แก่ ไข่น้ำ (Wolffia globosa) แหนเป็ดเล็ก (Lemna minor) แหนเป็ดใหญ่ (Spirodela polyrrhiza) และแหนแดง (Azolla pinnata) ด้วยวิธี Kjeldahl พบว่าไข่น้ำมีปริมาณโปรตีนสูงที่สุด ซึ่งเท่ากับร้อยละ 27.45±0.02 (โดยน้ำหนัก) จึงคัดเลือกมาศึกษาสภาวะที่เหมาะสมในการสกัดโปรตีนเข้มข้นจากไข่น้ำโดยการปรับ pH ระหว่าง 3-6 และอุณหภูมิระหว่าง 37-80 องศาเซลเซียส ซึ่งสภาวะที่เหมาะสมในการผลิตโปรตีนเข้มข้นจากไข่น้ำมากที่สุดที่ pH 4 และอุณหภูมิ 80 องศาเซลเซียส โดยผลิตโปรตีนได้สูงสุดร้อยละ 69.96±0.10 (โดยน้ำหนัก) จากนั้นวิเคราะห์องค์ประกอบทางเคมีของโปรตีนเข้มข้นจากไข่น้ำพบว่ามีปริมาณโปรตีนร้อยละ 69.96 (โดยน้ำหนัก), ไขมันร้อยละ 8.96 (โดยน้ำหนัก), คาร์โบไฮเดรตร้อยละ 8.84 (โดยน้ำหนัก), ความชื้นร้อยละ 5.66 (โดยน้ำหนัก), เส้นใยร้อยละ 4.06 (โดยน้ำหนัก) และเถ้าร้อยละ 2.49 (โดยน้ำหนัก) เมื่อวิเคราะห์โลหะหนักในตัวอย่างด้วยวิธี Air-C2H2 Flame Atomic Absorption Spectrometry พบว่า LPC ที่ผลิตได้มีปริมาณ แมงกานีส 2.37 มิลลิกรัม/ลิตร, เหล็ก 2.29 มิลลิกรัม/ลิตร และสังกะสี 2.16 มิลลิกรัม/ลิตร จากการศึกษาชนิดและปริมาณกรดอะมิโนที่ในตะกอนโปรตีน พบว่ามีปริมาณกรดกลูตามิคสูงที่สุดคือร้อยละ 8.07 …

Epstein-Barr Nuclear Antigen 1 (EBNA1), a crucial protein encoded by the Epstein-Barr virus (EBV), plays an important role in the replication and transcriptional regulator of EBV virus latent gene expression. Therefore, it is associated with several diseases especially human malignancies. The previous studies have demonstrated that chalcone derivatives can potentially inhibit the EBV virus. In this study, a series of designed chalcones were in silico screened toward EBNA1 using molecular docking. Oxidized carbon nanospheres (OCSs) and graphene oxide sheets (GOShs) were prepared by oxidation of graphite. Oxidized carbon nanotubes (OCTs) was synthesized from single-walled carbon nanotubes. All three shapes of oxidized carbon nanoparticles (OCNs) were used for investigation of their intracellular trafficking in human embryonic kidney cells (HEK293T), mouse macrophage cells (RAW264.7) and human liver cancer cells (HepG2). The results revealed all three shapes of OCNs were internalized into through HEK293T and RAW264.7 cells without endosomal trapping, including localization of all the three OCNs in HEK293T's lysosome. In this study, isolation, screening, fermentation optimization, and determination of the expression of key enzymes involving in D-lactic acid production were conducted for selection of the novel D-lactic acid isolate with the high yield, productivity, and optical purity. From the screening experiment, the novel isolate, NK26-11T, was obtained. The isolate NK26-11T was a Gram-stain-positive, catalase-positive, facultatively anaerobic, spore-forming, rod-shaped bacterium isolated from soil sample in Thailand. This isolate homofermentatively fermented glucose for D-lactic acid and grew at the wide range of temperature between 20 and 45 °C and the pH of 5-8.5. Brackish water reverse osmosis (BWRO) and seawater reverse osmosis (SWRO) membranes were used in the 2-stage reverse osmosis (RO) unit to recover, pre-purify, and pre-concentrate lactic acid. Calcium lactate (CaLAC), sodium lactate (NaLAC), and ammonium lactate (NH4LAC) were used as model feed solutions. The operating pressure showed a pronounced effect on lactate passage through the first BWRO unit, and the Donnan exclusion effect and hydrogen bonding were responsible for cation rejection. A free radicals can promote denaturing reactions in many cellular components, which create oxidative stress and it results to damage in body tissue. The free radicals can causing many diseases including heart diseases, Alzheimer, inflammation and cancer. This research aimed to investigate the effect of bioactive peptides derived from Synechococcus sp. cultured for 21 days on antioxidant, anti-inflammation and anticancer activities. The Synechococcus sp. protein hydrolysate was prepared by trypsin digest and purified by ultrafiltration with molecular weight cut off membranes of 10, 5 and 3 kDa. The MW <3 kDa fraction showed the highest NO, ABTS and DPPH radical scavenging activities with IC50 values of 34.51±9.8 µg protein/mL, IC50 11.54±0.3 µg protein/mL and IC50 13.63±0.15 µg protein/mL, respectively. The F2 fraction from gel filtration chromatography showed the strongest NO and ABTS radical scavenging activities with IC50 values of 7.68±0.64 µg protein/mL and 9.74±0.2 µg protein/mL, respectively. The F2 was purified by RP-HPLC to yield four fractions. The 30 – 40 min sub-fraction (F2-4) was selected for further analysis by mass spectroscopy. Five isolated peptides with amino acid sequences of AILQSYSAGKTK; 1,265.69 Da, ALNKTHLIQTK; 1,265.74 Da, LLVHAPVK; 875.55 Da, IPDAHPVK; 875.48 Da and VVVLRDGAVQQLGTPR; 1,706.97 Da were identified. The F2-4 had higher DPPH and NO radical scavenging activity compared to the synthetic peptide. Moreover, AILQSYSAGKTK had the highest ABTS radical scavenging activity. Furthermore, the F2 fraction protected oxidation-induced DNA damage in pBR322, pKS and pUC19 cells. F2 fraction was selected to study anti-inflammatory and anticancer properties. Anti-inflammatory effect, F2 fraction showed no cytotoxicity toward RAW264.7 macrophage cells. RT-PCR and qRT-PCR results showed that F2 reduced gene expression of pro-inflammatory cytokines iNOS, TNF-α, COX-1, COX-2 and IL-6. For anticancer activity, F2 fraction showed high anticancer activities in the human colon cancer cells (SW620) according to cytotoxic activity (MTT assay), with IC50 values of 106.58±21.46 µg protein/mL. The F2 fraction activated the apoptotic pathway in SW620 cells after treatment for 24, 48 and 72 hours. The highest activities of caspases 3, 8 and 9 were observed after treatment for 72 hours. These findings suggested that unicellular cyanobacterium Synechococcus sp. may be used to develop for natural anti-inflammation and natural anticancer drugs.

Six different parts of Manilkara zapota which consisted of barks, flowers, fruits, leaves, roots and seeds were investigated for total phenolic content, total flavonoid content, antioxidant and antityrosinase activities. Methanol crude extract of flowers showed the highest total phenolic content (368.73 ± 0.65 mg GAE/g), while methanol crude extracts of seeds and roots showed high total flavonoid content (90.21 ± 0.57 and 89.03 ± 1.00 mg QE/g, respectively). น้ำนมวัวเป็นแหล่งโภชนาการที่สำคัญของมนุษย์ ประกอบไปด้วย โปรตีน วิตามิน แร่ธาตุ และแคลเซียม อย่างไรก็ตามเคซีนซึ่งเป็นโปรตีนหลักที่อยู่ในน้ำนมวัวนั้น ก่อให้เกิดการแพ้ในผู้บริโภคบางรายได้ ซึ่งมีอาการตั้งแต่เบาจนถึงหนัก เช่น หายใจไม่ออก อาเจียน เป็นลมพิษ หรือตาย เป็นต้น ดังนั้นในอาหารที่มีส่วนประกอบของน้ำนมวัว จะต้องมีการระบุแจ้งบนฉลากตามที่กฎหมายได้กำหนดไว้ แต่ในอาหารหลายชนิดไม่มีการระบุดังกล่าว ดังนั้นวิธีตรวจเคซีนอย่างง่ายสำหรับผู้บริโภคที่แพ้ต่อเคซีนจึงเป็นสิ่งจำเป็น ในการศึกษาครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาการตรวจด้วยอิมมูโนวิทยา แบบการไหลในแนวระนาบ (lateral flow immunoassay) ในรูปแบบแถบทดสอบสำหรับการตรวจวัด เคซีนจากวัว โดยในการเตรียมแถบทดสอบจะใช้วิธีการเติมสารต่างๆ ลงบนแถบทดสอบ 2 วิธีคือ การหยดสารละลายลงบนขอบกระจกสไลด์แล้วประทับลงบนเมมเบรน และการจ่ายสารละลายด้วยเครื่องดูดจ่ายสารละลายแบบอัตโนมัติ จากการหาภาวะที่เหมาะสมในการเตรียมแถบทดสอบโดยใช้วิธีการประทับด้วยกระจกสไลด์ในการเติมสารนั้น พบว่า ต้องตรึงเคซีนที่เส้นทดสอบและตรึง IgG-Fc ที่เส้นควบคุมด้วยความเข้มข้น 1 และ 0.25 มิลลิกรัมต่อมิลลิลิตร ตามลำดับ โดยมีค่าคัดออก (cut-off value) อยู่ที่ 20 ไมโครกรัมต่อมิลลิลิตร และพบว่าความเข้มของสีที่ปรากฏบนแถบทดสอบมีความไม่แน่นอน ในการหาภาวะที่เหมาะสมในการเตรียมแถบทดสอบโดยใช้เครื่องดูดจ่ายสารละลายแบบอัตโนมัติในการเติมสาร พบว่าต้องใช้ เคซีน 0.2 ไมโครกรัม และ IgG-Fc 0.0125 ไมโครกรัม ตามลำดับ และใช้อนุภาคทองระดับนาโนที่ติดกับแอนติบอดีที่ความเข้มข้น 300 ไมโครกรัมต่อมิลลิลิตร เติมลงบนส่วนแผ่นคอนจูเกต โดยมีค่าคัดออกอยู่ที่ 20 ไมโครกรัมต่อมิลลิลิตร พบว่าความเข้มของสีที่ปรากฏบนแถบทดสอบมีความแน่นอนมากกว่าวิธีแรก เมื่อนำแถบทดสอบที่เตรียมได้ไปตรวจหาเคซีนจากวัวที่เติมลงในตัวอย่างน้ำนมควาย น้ำนมแพะ และน้ำนมถั่วเหลือง พบว่ามีค่าคัดออกอยู่ที่ 22.5, 9 และ15 มิลลิกรัมต่อมิลลิลิตรตามลำดับ จากการทดลองทั้งหมดสรุปได้ว่า สามารถนำแถบทดสอบไปใช้ตรวจในตัวอย่าง นมควาย นมแพะ และนมถั่วเหลือง ที่มีการปนเปื้อนเคซีนของนมวัวได้
