The Influence Of Myocardin Effector Genes On The Tgf-Beta Smad3 Signaling Pathway In Human Pleural Mesothelial Cells, Hema Chandra Hrushi Dusharla

Pleural Fibrosis (PF) involves excessive extracellular matrix (ECM) deposition, mainly by myofibroblasts, leading to pleural thickening and impaired lung function. Pleural mesothelial cells (PMCs) contribute via mesothelial and mesenchymal transition (MesoMT), triggered by cytokines like TGF-β. Our lab identified Myocardin (MyoCD), a transcriptional co-activator of cardiac and smooth muscle, is a master regulator of pleural fibrosis through interactions with serum response factor (SRF) and Smad2/3 transcription factors which initiate fibrosis signaling. RNA sequencing and qPCR analysis revealed that silencing MyoCD decreased the expression of CTNNB1, MDFI, and Tspan2. MDFI emerged as a potent candidate, as this gene affecting the expression …

Despite this, there is limited evidence regarding the prevalence and associated factors of mental health problems among university students in Iraq. Therefore, this study aimed to assess the prevalence and severity of depression, anxiety, and stress in this population and to examine their associations with sociodemographic characteristics, insomnia, internet use, and physical activity. This study aims to investigate, through in vitro methods, the molecular, immunofluorescent, and immunohistochemical effects of resveratrol and IL-6 on CXCL1 gene/protein expression in human breast cancer cells (MCF-7). MCF-7 and MCF-10A cell lines were cultured and divided into four groups: control, IL-6-treated, resveratrol-treated, and a combination-treated group. CXCL1 gene expression was assessed by real-time PCR (RT-PCR), while CXCL1 protein levels were analyzed using immunofluorescence and immunohistochemistry. Cell viability was quantified by MTS assay using ELISA. Breast cancer is the second leading cause of mortality among women worldwide, highlighting the need for efficient histopathology-based screening methods for early diagnosis. This study introduces HistoDX, a deep learning framework to classify Invasive Ductal Carcinoma (IDC) using 277,524 histopathology image patches (50x50 pixels) from Paul Mooney’s IDC dataset on Kaggle, comprising No Cancer and IDC(+) classes. HistoDX employs a preprocessing pipeline with normalization, data augmentation, and class balancing via oversampling and weighted loss to address the class imbalance. Idiopathic pulmonary fibrosis (IPF) is an incurable interstitial lung disease defined by the progressive, irreversible scarring of the tissues between the lung’s alveoli. Schiff bases exhibit a wide range of biological activities, including antibacterial, antifungal, anticancer, and anti-inflammatory properties. The aim of study was to synthesized 2-amino-benzothiazole derivatives and showed their novel structure leads to potent antimicrobial activity. The Schiff base compounds derived from 6-bromo-4-fluoro-2-aminobenzothazol was prepared from the reaction of 6-bromo-4-fluoro-2-aminobenzothazol compound (W1) with potassium thiocyanate in the presence of bromine. Background: Understanding the knowledge, attitudes, and practices (KAP) regarding refractive errors and spectacle use is essential for promoting eye health and effective vision correction, especially in children.

Objective: This study assessed the demographic characteristics, knowledge, attitudes, and practices of participants concerning refractive errors and spectacle use, focusing on correlations influencing these factors.

Methods: A cross-sectional study of 228 participants analyzed demographic data, duration of glasses use, and responses to a structured questionnaire on KAP related to refractive errors and spectacles.

Cerium oxide nanoparticles (CNPs) are recognized for their ability to scavenge reactive oxygen species (ROS), making them promising candidates for mitigating oxidative damage in biomedical applications. However, many conventional synthesis methods limit the formation of Ce³⁺ states, which are critical for this antioxidant function. Building upon previous findings that glucose can promote Ce³⁺ formation, this study investigates the broader impact of both reducing (glucose, fructose, galactose) and non-reducing (dextran, sucrose, cyclodextrin) natural sugars on CNP synthesis. The thoracic dystrophies are inherited skeletal conditions where abnormal embryonic development of the thoracic skeleton results in a narrow chest, pulmonary hypoplasia, and respiratory insufficiency, which can be severe or lethal. The majority of thoracic dystrophies are due to biallelic alterations in genes needed for normal ciliary function. However, despite the identification of over 20 genes as causal for the thoracic dystrophy phenotype, around 20% of patients remain without a molecular diagnosis. We present two unrelated families with a clinical diagnosis of thoracic dystrophy with associated respiratory insufficiency without a molecular diagnosis on previous genetic testing. Equine Herpesvirus-1 (EHV 1) is a worldwide significant pathogen that causes respiratory illness, abortion, and neurological disorders in equine. Current vaccines, including live attenuated, inactivated, and subunit platforms, do not prevent viral latency, mucosal shedding, or cross-strain immunity, requiring alternative approaches. To address these gaps, this project explores the design, synthesis, and in vitro testing of an mRNA vaccine targeting immunogenic EHV 1 glycoprotein (gB, gC, gD, gG, and gM). The monkeypox virus (MPXV), a DNA virus causing zoonotic disease, poses major global public health challenges, with mortality rates between 3%–6%. Although smallpox vaccines provide partial cross-protection, there is a critical need for a dedicated, effective monkeypox (mpox) vaccine. This study aimed to design a multi-epitope peptide-based vaccine specifically adapted to the HLA allele profiles common in Southeast Asian populations, where MPXV cases are rising. The growing concern over antibiotic resistance has driven global efforts to explore innovative solutions, including the use of Penicillin G acylase (PGA) to produce semisynthetic β-lactam antibiotics. This study screened four potential in-tracellular PGA-producing bacteria: Alcaligenes faecalis InaCC B444 (AfPGA), Kluyvera cryocrescens InaCC B850 (KcPGA), Providencia rettgeri InaCC B25 (Pr25PGA), and P. rettgeri InaCC B466 (Pr466PGA). Penicillin G Acylase encoding genes (pgas) were isolated from them using a Degenerate Oligonucleotide Primed-PCR (DOP-PCR) approach and sequenced. Background: High-risk strains of HPV are known to cause cervical cancer. Multiple clinical studies have emphasized that smoking and drinking are critical risk factors for cervical cancer and its high-grade precursors. In this study, we investigated if smoking and/or drinking augment the molecular mechanisms of the cervical carcinogenesis and defined a potential therapeutic approach for their attenuation.

This study evaluates bacterial adhesion, biofilm formation, and Staphylococcus aureus (S. aureus) (ATCC 25923) colonization on medical and cosmetic contact lenses made from Hilafilcon B and Omafilcon B under simulated ocular conditions. Non-incubated lenses served as controls, and experiments were repeated three times with a variability margin of ±5%, ensuring accuracy and reliability. Results revealed that biofilm growth and bacterial adhesion were influenced by lens material, type, and incubation duration. Cosmetic lenses made from Hilafilcon B exhibited the highest biofilm growth (1.95%) after one day, highlighting their susceptibility to microbial colonization. Pancreatic cancer (PanCa) is characterized by a lack of early diagnosis, poor response to available therapeutic modalities, and chemoresistance. Gemcitabine (GEM) is currently considered the most effective therapy for PanCa; however, it shows only a marginal survival benefit of 6 months. This poor drug response has been attributed to desmoplasia, causes suboptimal drug delivery, alters tumor microenvironment (TME), which includes tumor surrounding blood vessels, fibroblasts, immune cells, extracellular matrix, and other signaling molecules and induces chemo-resistance in tumors. This research utilized a carbonization procedure via microwave irradiation assisted by H3PO4 to generate a cost-effective adsorbent (CWDAG) from wood dust (WD) and algal (AG) biomass. The resulting CWDAG adsorbent was characterized for its methylene blue (MB) dye adsorption properties. The activation process employs 800 W microwave radiation for 15 min under a nitrogen gas (99.99%) atmosphere. However, their use carries a higher risk of ocular infections, particularly when proper hygiene practices are neglected. Herein, biohybrid biosorbent material was produced from grafting chitosan benzaldehyde/Lactobacillus casei (CHS-BZ/LAC) for the removal of anionic dye acid red 88 (AR88). Background: Migration and Invasion Enhancer 1 (MIEN1) is a critical oncogene that drives cancer cell migration, invasion, and epithelial-to-mesenchymal transition (EMT) in various cancers. MIEN1 overexpression is linked to aggressive tumor progression and poor prognosis. Despite its established role in cancer biology, targeted therapeutic options remain unexplored. This study aimed to identify and characterize first-in-class therapeutic molecules targeting the MIEN1 signaling pathway to inhibit cancer progression.

The insulin receptor (INSR) has been shown to be hyperactive in hepatic stellate cells (HSCs) in humans and rodents with liver fibrosis. To explore HSC cellular mechanisms that INSR regulates during pro-fibrotic stimulation, we used CRISPR-Cas9 technology. We knocked out a portion of the INSR gene in human LX2 HSC cells (INSRe5- 8 KO) that regulates insulin responsiveness but not the insulin-like growth factor (IGF) or transforming growth factor-β (TGFβ) signaling. The INSRe5- 8 KO HSCs had significantly higher cell growth, BrdU incorporation, and lower TP53 expression that suppresses growth, and they also exhibited increased migration compared to the Scramble …

Background and Objective: The early diagnosis of neurodegenerative diseases, such as Parkinson's disease (PD), is particularly challenging because symptoms appear only after significant neuronal damage has already occurred. This study is utilizing variant call format (VCF) analysis to identify genetic variants and novel genes that could serve as early prognostic markers for prodromal PD. Materials and Methods: Data were sourced from the Parkinson's Progression Markers Initiative (PPMI), focusing on prodromal patients with gVCF data from the 2021 cohort.
