Investigating The Role Of Prostasin In Angiogenesis: A Chorioallantoic Membrane Model Approach, Zaima Aline

Prostasin, also known as PRSS8 or CAP1, is a glycosylphosphatidylinositol (GPI)-anchored extracellular serine protease involved in various physiological and pathophysiological processes. Prostasin expression is observed during key phases of trophoblast invasion and placental development, critical steps in embryo implantation in Rhesus monkeys. This suggests a role for prostasin in extracellular matrix remodeling and tissue invasion, both essential for a healthy pregnancy. Moreover, prostasin appears vital for development as studies in mice show that a germline knockout of the gene leads to significant placental defects and eventual embryonic death, highlighting its importance in vascular and structural development within the placenta.

Protein-based self-assembling biomaterials, known as Thermo-Responsive Assembled Proteins (TRAP), present meaningful potential as drug delivery carriers. These materials enable the controlled, slow release of poorly soluble chemotherapeutic agents, such as doxorubicin (Dox), while potentially reducing systemic off-target effects. In collaboration with multiple NYU labs, this study evaluated the efficacy of two TRAP variants—TRAP and F-TRAP—as drug delivery carriers in a xenograft mouse model of glioblastoma multiforme (GBM).

Inflammatory cytokines such as interleukin-6 (IL-6) and interleukin-12 (IL-12) are central regulators of immune signaling and key biomarkers of disease, yet existing assays for their detection remain slow, invasive, and lack multiplexing ability. This dissertation advances the development of single-walled carbon nanotube (SWCNT) optical nanosensors capable of real-time, multiplexed, and molecularly specific cytokine detection through innovative use of single-stranded DNA (ssDNA) interfaces.

First, an IL-6-specific DNA aptamer was employed as both a dispersing agent and recognition probe for SWCNT fluorescence sensing. Intradermal injections of silicone-based fillers for the face and body may cause chronic inflammation and fibrotic reactions in some individuals. Chickens from the UCD 200/206 White Leghorn (WL) line spontaneously develop a fibrotic autoimmune disease known as scleroderma/systemic sclerosis (SSc). Using the SSc-prone UCD and healthy WL chickens, a longitudinal study was conducted to examine the cellular inflammatory responses to intradermal (i.d.) injections of sterile SILIKON-1000. This thesis explores the development and application of a DNAzyme-based biosensor designed to detect labile metal species in environmental samples. Real time and on-site monitoring of labile metal fractions would make a valuable contribution to environmental management, as these fractions are the most bioavailable and pose significant toxicity risks to aquatic organisms. Conventional methods for detecting labile metals, while effective, are often burdened by high costs, complexity, and lengthy processing times, making them less ideal for rapid and widespread environmental assessments.

Cholesterol oxidase (CO) was successfully produced from Streptomyces sp. via the submerged fermentation method, and 69 U/mL enzyme activity was obtained. This study aimed to determine cholesterol oxidation kinetics and the production of CO as a catalyst. The enzyme was diluted to 0.15, 0.075, and 0.00375 U/mL for the oxidation reaction. The substrate was also prepared in three concentrations: 3.23, 6.46, and 12.93 mM. The optimization of conditions for enzymatic cholesterol oxidation was investigated through measurement of the effect of initial cholesterol and enzyme concentrations. The development of nanostructures for delivering multiple proteins to targeted antibodies has become a promising avenue in biotechnology, driven by advancements in synthetic polymers like poly(propylene sulfone), PPSU. However, the PPSU polymer faces a major limitation - its high insolubility. To address this challenge, this study investigates whether PPSU oligomers can enhance both the solubility and stability of lysozyme, a model protein that is cost-effective and widely accessible. Using dimer, trimer, tetramer, and hexamer configurations, we analyzed the interactions between PPSU oligomer and lysozyme under varying conditions. Oocyte cryopreservation is a crucial technique for preserving the genetic material of female cats, yet maintaining post-thaw oocyte developmental competence remains a significant challenge. This study investigated the impact of intra- and extracellular trehalose on the developmental competence of feline oocytes following cryopreservation. In Experiment 1, immature feline oocytes were matured in vitro (IVM) with varying concentrations (0, 1.5, and 3 mM) of intracellular trehalose hexaacetate (Tre-(OAc)6). Objective: This study aims to investigate the trend and awareness of e-cigarette use among students in a private college located in Sabah, Malaysia. Research Methods: The cross-sectional study was conducted in a private college in Sabah. A self-administered questionnaire was adapted and distributed to 300 randomly selected students regardless of their e-cigarettes vaping status. IBM SPSS Statistic 27 was utilized to analysed accumulated data. Results: A-245 students participated in the study with a response rate of 81.6%. This study investigated the role of long non-coding RNA (lncRNA) PCAT3 in arthritis, focusing on its impact on proinflammatory markers. Researchers analyzed both mRNA and protein levels of various proinflammatory markers (TNF-α, IL-1β, IL-4, IL-6, IL-8, IL-10, and IL-13) in arthritis patients and compared them to healthy controls. Additionally, PCAT3 expression was assessed in both patient groups and further investigated using siRNA knockdown in the human synovial cell line SW982. Results revealed distinct expression patterns of proinflammatory markers in arthritis patients, with TNF-α and IL-6 showing the highest levels. Advances In Biomedical Research And Treatments: What Is Acceptable?, Michael L. Clancy, Khalid M. Iskandarani, Mitchell C. Mccrea

This study investigated the technology acceptance (TA) of twenty-first century biomedical treatments by adults in the United States. A new TA instrument was created, using five distinct levels: (1) Healing and Prevention, (2) Replacement Organs, (3) Enhancements-Medical, (4) Enhancements-Discretionary, and (5) Transhumans. An on-line survey produced 353 usable responses, which showed distinct patterns for each of five biomedical treatment levels. There was clear support for Levels 1–3, but very strong opposition to Levels 4–5. The TA finding draws the line between which human interventions are acceptable versus others that should be prohibited through public policies and medical guidelines.

Nanomedicine is an interdisciplinary field within the developing nano medication delivery system. Ayurveda, a traditional form of healing, employs nanoparticles known as Bhasmas. Nanoparticles can be selectively designed to simultaneously deliver many medications. This innovative functionality enables meticulous regulation of drug release, therefore obviating the necessity for intricate dosing schedules. This enhances patient compliance with treatment. Interzone/cavitation are key steps in early stage joint formation that have not been successfully developed in vitro. Further, current models of endochondral ossification, an important step in early bone formation, lack key morphology morphological structures such as microcavities found during development in vivo. Background

Reactive oxygen species (ROS) play an important role in cell signaling and homeostasis. However, overproduction of ROS leads to oxidative stress, as well as other related diseases, including cancer, neurodegenerative diseases, and cardiovascular disorders. Conventional ROS detection methods are limited by their low sensitivity and low specificity in real-time measurements. Fluorescent probes can offer valuable tools to detect ROS in biological systems. This study is focused on the detection of ROS formation during hypoxia and reoxygenation.

A major challenge of developing collagen mimetic peptides (CMPs) by bacterial expression is to include hydroxyproline for biomedical applications. Coexpression of Prolyl-4-hydroxylase from A.mimivirus with CMPs was investigated. Additionally, four expression designs were created for potential applications in mammalian cells to utilize their natural ability of proline-hydroxylation by post-translational modification.

In the current work, rice husk (RH) biomass was pyrolyzed using tartaric acid as an activating agent to produce activated carbon (AC). For 60 min, the procedure was run at 700 °C and 5 L/min of nitrogen (N2) flow rate. The study used response surface methodology (RSM) and a 3-level Box-Behnken design (BBD) to optimize the extraction efficiency of rice husk activated carbon (RHAC) in eliminating reactive orange 16 (RO16) dye from an aqueous solution.
