Mesenchymal stem cells (MSCs) are widely used in regenerative medicine due to their self-renewal capacity, multilineage differentiation potential, and paracrine activity. Limitations associated with the sourcing and scalability of human MSCs have increased interest in xenogeneic alternatives, particularly sheep umbilical cord–derived MSCs (sUC-MSCs). These cells display high proliferative capacity, stable karyotypes, and a typical MSC immunophenotype (CD44, CD73, CD90, CD105), with low basal immunogenicity. sUC-MSCs exhibit osteogenic, chondrogenic, and adipogenic differentiation potential and secrete angiogenic and immunomodulatory factors, including VEGF, HGF, IGF-1, TGF-β, and IL-10. Owing to their biological similarities to human MSCs and accessibility from perinatal tissue, …

Development Of Self-Assembled Polymeric Polyplexes For Microrna Delivery: Toward Treatment Of Alcohol-Associated Liver Disease, Marjina Akter Kalpana
Theses & Dissertations
Self-assembled polymeric polyplexes have the potential to serve as an RNA interference (RNAi) delivery platform for the combined inhibition of CXCR4 and miR-155 in the treatment of alcohol-associated liver disease (AALD). A central challenge in RNA delivery is balancing the extracellular stability of polyplexes, cellular uptake, and intracellular release of RNA cargo. We developed polymeric polyplexes using the CXCR4-antagonist polymer PAMD, designed to target CXCR4 on activated hepatic stellate cells (HSCs) and deliver therapeutic miRNA to activated Kupffer cells (KCs) in fibrotic liver. We improved the polyplexes by cholesterol modification of PAMD and by PEGylation of PAMD-Ch. The cholesterol modification …

Cold atmospheric pressure plasma (CAP) is emerging as a clinically relevant therapy for dermatological conditions such as actinic keratosis, warts, and chronic wounds. However, these therapies lack strategies to monitor CAP delivery in situ and to ensure delivery of an effective CAP dose without unwanted toxicity. CAP acts as a therapeutic agent in these biomedical applications primarily (but not solely) through reactive oxygen and nitrogen species (RONS) generated at transiently high local concentrations. Here we demonstrate the use of bio-electrochemical sensors capable of real-time measurements of key CAP RONS: hydrogen peroxide and oxidation-reduction-potential (ORP). In in vitro scratch assays and …

AUIQ Complementary Biological System
Postoperative shivering is a common complication that can affect patient comfort and stability. Pethidine and tramadol are widely used to relieve this symptom, but their relative effectiveness and safety remain under investigation. In this randomized trial, 50 patients (aged 25–35 years) with postoperative shivering were divided into two groups: one group receiving intravenous tramadol (n = 25 patients) and one group receiving pethidine (n = 25 patients). Onset of drug action, vital signs, and side effects were systematically assessed. Pethidine had a significantly faster onset of action than tramadol (12.20 ± 0.35 min vs 7.16 ± 0.28 min, p < 0.001). No significant differences were observed between the two groups regarding body temperature, heart rate, oxygen saturation, or blood pressure (P < 0.05). Side effects, such as dizziness (16% vs. 8%), nausea and vomiting (20% vs. 8%), and nausea alone (48% vs. 20%), were more common and showed a statistically significant increase in the tramadol group. Both drugs were effective in suppressing postoperative tremor, but pethidine demonstrated superior efficacy, a faster onset of action, and a better safety profile. These results confirm pethidine as the recommended choice at an intravenous dose of 0.5 mg/kg.

This work investigates the efficacy of fenugreek seed powder (FSP), an agricultural product, as an economical and sustainable biosorbent for the extraction of crystal violet (CV) dye from aqueous environments. The biosorbent was analyzed using XRD, FTIR, and SEM-EDX, validating its amorphous structure, surface functional groups, and porous shape. Batch biosorption tests were adjusted utilizing a Box–Behnken design (BBD), incorporating three independent variables: FSP dose (0.02–0.1 g/100 mL), pH (4–10), and time (5–60 min). The model exhibited exceptional prediction accuracy (R² = 0.99, CV = 2.12%) and recognized all three variables as statistically significant (p < 0.05). The maximum CV removal attained was 91.8% efficiency at an FSP dose of 0.1 g/100 mL, pH 7, and a time of 60 minutes. FTIR examination indicated changes in the –OH, C=O, and C–O functional groups, signifying their participation in dye interaction, whilst SEM pictures validated dye deposition on the biosorbent surface. The biosorption process was ascribed to electrostatic forces, hydrogen bonding, π–π stacking, and pore filling. These findings underscore FSP as a viable, economical biosorbent for the removal of cationic dyes in wastewater treatment applications.

Monoamine oxidase A (MAO-A) has recently been identified as a candidate for cancer therapy due to its roles in tumor development and growth. Conventional approaches for the discovery of MAO-A inhibitors are both costly and time-consuming. The objective of this work was to apply in silico methods to predict natural isoquinoline alkaloids capable of acting as potent MAO-A inhibitors, and to establish this class of compounds as potential sources of a cost-effective anticancer therapy. Twenty-one natural isoquinoline alkaloids were subjected to a multistep computer-aided drug discovery protocol. The compounds were initially screened for pharmacokinetic (ADME), toxicity, and drug-likeliness properties by …

Background The increasing resistance of bacteria to conventional antibiotics poses a significant health challenge. Innovative strategies, such as combining antibiotics with agents like quorum sensing inhibitors (QSIs), have been developed to combat this issue. QSIs enhance antibiotic efficacy without inhibiting bacterial growth, minimizing the risk of resistance. Aims Evaluate the combined effect of Sitagliptin (STG) as a QSI with Meropenem (MER), fabricate drug-loaded Polyvinyl Alcohol (PVA) nanofibers, and investigate their antimicrobial activity against standard Pseudomonas aeruginosa (PAO1) and carbapenem-resistant P. aeruginosa (CRPA). Methods The combinatorial effect was assessed using a checkerboard assay. PVA/STG, PVA/MER, and PVA/STG/ MER nanofibers were fabricated …

Polysomnography (PSG) remains the gold standard for evaluating sleep-disordered breathing (SDB) and related conditions, yet its clinical potential is often limited by overreliance on the apnea-hypopnea index (AHI) and similar summary metrics. This narrative review offers a structured, physiology-informed approach to PSG interpretation that integrates sleep architecture, arousal burden, respiratory event morphology, oxygenation patterns, and CO2 trends. Recognizing phenotypic patterns-such as REM-related and positional OSA-through sleep stage and positional stratification is essential for directing targeted therapy. We outline common interpretive pitfalls, including automated scoring errors, overlooked signal artifacts, and the first night effect, emphasizing the need for careful manual …
