Currently there are no site-specific bioavailability-based prediction models for assessing the impacts of nickel (Ni) in marine environments although there are indications that these may be warranted. The aim of this research was to characterize the complexation of Ni in relation to toxicity and speciation. Various complexing ligands were used, and it was predicted that the binding affinity (logKf) of ligands would be inversely correlated to toxicity based on dissolved Ni concentrations ([NiD]) but that on a free ion concentration ([Ni2+]) basis, toxicity would not vary. Gold mining is an important part of economic development in Northern Canada. A large portion of the gold that is found in the North is contained within arsenopyrite ores, therefore, arsenic contamination is of special concern. Little is known regarding the impacts of arsenic on plants and mycorrhizae in Northern Ecosystems. Arsenic has been shown to negatively impact plant growth and seed germination in some temperate species, while others are tolerant and can accumulate arsenic concentrations over 1000 mg/kg. Chromite and rare earth element development was identified in the 2015 Canadian Federal budget as a significant opportunity, however, key data gaps exist regarding the environmental concerns related to these resource developments. Chromium is essential in the production of stainless steel, and no suitable substitute is known. The fruit fly Drosophila melanogaster has a long history as a research model for studies on behaviour and group dynamics. When individuals are grouped in an environment and resources decrease, then they may behave co-operatively with one another or antagonistically compete. Hamilton’s Law states that if the benefit of helping a related individual out-weighs the cost to its direct fitness, then the individual’s indirect fitness will increase when they help their kin compared to helping an unrelated stranger. Virginia Mallow (Sida hermaphrodita) is a perennial herb of the Malvaceae family that is native to riparian habitats in northeastern North America. Throughout most of its geographical distribution however, it is considered threatened and only two populations are known from Canada. Rainbow trout is the most farmed fish in Ontario, and thus is economically important to the province. Despite this, there is a lack of understanding regarding fish innate immunity, specifically with regards to interferon-stimulated genes (ISGs) and their antiviral effector functions. ISGs are the workhorses of the innate antiviral response, operating together to limit each step of virus replication. The Viral Hemorrhagic Septicemia Virus (VHSV) induced gene (Vig)-3 is a newly identified ISG within many fish species and is homologous to ISG-15 in mammals. Invasive sea lamprey (Petromyzon marinus) in the Great Lakes are controlled using the pesticide (lampricide) 3-trifluoromethyl-4-nitrophenol (TFM), which is applied to nursery streams containing larval lamprey. The toxicity of TFM to lamprey is affected by various environmental and physiological factors, which can lead to residual lamprey that survive TFM treatment. The lampricide 3-trifluoromethyl-4-nitrophenol (TFM) is applied to tributaries of the Great Lakes to control invasive sea lampreys (Petromyzon marinus). Although TFM is selectively toxic to larval sea lampreys, non-target mortality can occur during lampricide treatments. This study focused on the environmental impacts of mining and potential techniques to mitigate them. The goals of this study were to (1) Quantify vegetation and fungal colonization at a remediated mine site in northern Canada, and to determine if there were effects from petroleum hydrocarbon contamination, (2) Determine the response of northern plants inoculated with mycorrhizal fungi to diesel contamination. Mycorrhizal colonization was observed in field collected roots and trends in colonization and vegetation cover suggested that these communities were no longer impacted by residual contamination. This study investigated ovarian development in Fundulus heteroclitus (mummichog), through molecular characterization of key mediators in the ovarian steroidogenic pathway; these included ovarian StAR (steroidogenic acute regulatory protein), cytochrome P450 enzymes, estrogen receptors (ERα, ERβ1 and ERβ2), and gonadotropin receptors (FSHr and LHr); additionally, hepatic vitellogenin (VTG1) and ERs, and brain P450 aromatase (P450arom; CYP19b) were studied. Lamprey (Petromyzontiformes) are a phylogenetically ancient group of jawless fishes that begin their lives as filter-feeding larvae (ammocoetes) before undergoing a complex metamorphosis into juvenile lamprey that involves major changes to their internal and external body plan. Some parasitic species, such as the sea lamprey (Petromyzon marinus), migrate to sea following metamorphosis, where they use their oral discs and rasping tongue to attach to and ingest vast quantities of blood from fishes. Accelerated climate warming in northern regions is causing permafrost degradation, leading to the reduction of the areal extent of permafrost. Permafrost is the foundation for many northern ecosystems and communities, so its thaw can have important ecological and societal consequences. Thaw of ice-rich permafrost can cause subsidence of the ground surface proportional to excess ice content. Terrain settlement can cause ponding or damage to infrastructure. Riparian wetlands are unique habitats facilitating all forms of life. The riverbanks of these environments provide ideal conditions for bacteria, plants, and higher organisms. Of particular interest to this research was the variation in microbial community structure at high, intermediate and poor water quality impacted areas. Assessing the capabilities of plants to retain microbial pathogens was identified. Root systems and corresponding soil are ideal locations for bacterial deposition, resulting in attachment at these areas. Application of 3-trifluoromethyl-4-nitrophenol (TFM) to control invasive sea lamprey (Petromyzon marinus) within the Laurentian Great Lakes seldom causes non-target mortality. However, under certain conditions, TFM can harm species such as the lake sturgeon (Acipenser fulvescens). Lake sturgeon less than 100mm in length are particularly vulnerable to TFM-induced mortality, and are more sensitive to TFM toxicity with increasing water alkalinity as compared to sea lamprey. Cuscuta (dodder) is a genus of roughly 200 species of obligate stem parasites with sub-cosmopolitan distribution. The fruit, generally regarded as a capsule, has a thin pericarp containing one to four seeds and opening at the base (circumscissile dehiscence; DE), or remaining closed (indehiscent; IN). IN has evolved multiple times in Cuscuta from DE, and is most common in the North American clades of subgenus Grammica. In addition, some species produce fruits that open irregularly. Giant Mine is an inactive gold mine located nine kilometers north of Yellowknife, Northwest Territories. Giant Mine has been the source of arsenic trioxide for the Baker Creek watershed since it opened over 60 years ago. Although arsenic levels in the creek are above the limits considered to be biologically relevant, there is no concrete evidence that plants and mycorrhizae have actually been affected. The Arctic has warmed by at least 3°C over the past 50 years and this rapid warming is expected to continue. Climate warming is driving the proliferation of shrubs across the tundra biome with implications for energy balance, climate, hydrology, nutrient cycling, and biodiversity. Changes in tundra plant water use attributable to shrub expansion are predicted to increase evapotranspirative water loss which may amplify local warming and reduce run-off. Increasing evidence indicates that plant community structure and therefore ecosystem function are mediated by below-ground fungal communities that form intracellular associations with plant roots called mycorrhizal associations. Arbuscular mycorrhizal fungi (AMF) are a type of mycorrhiza that colonize the plant host intracellularly but maintain hyphae outside the root cell for resource acquisition. The importance and function of AMF associations are well-documented in terrestrial ecosystems, but are less understood in aquatic or semi-aquatic systems. The photosynthetic electron transport (PET) chain relies on the chlorophyll and carotenoid biosynthetic pathways for pigments that harness light, and protect the photosynthetic apparatus from damage by light. The pigment pathways also connect to the PET chain through electron transfer where the plastoquinol (PQ) pool accepts electrons from phytoene desaturase (PDS) of the carotenoid pathway and the aerobic cyclase (ACS) of the chlorophyll biosynthetic pathway. In fishes, hyperammonemia may occur following feeding or exposure to abnormally high concentrations of environmental ammonia due to sewage effluents, agricultural run-off and in crowded aquaculture pens. Increased internal ammonia can result in hyperactivity, convulsions, coma and death. In mammals, it is also associated with potentially fatal brain edema, in which the accumulation of intracellular water results in swelling, increased intracranical pressure and herniation leading to death. The backbone of sea lamprey (Petromyzon marinus) control in the Great Lakes is the use of the piscicide, 3-trifluoromethyl-4-nitrophenol (TFM), which is applied to streams containing larval sea lampreys. However, TFM effectiveness can be influenced by abiotic factors such as pH and temperature, which fluctuate daily and seasonally. The objectives of this thesis were to evaluate the influence of pH, temperature, and season on the toxicity, uptake, and elimination of TFM by larval sea lamprey. Canada’s northern boreal forest has faced significant climate warming over the past century. This change can have direct and indirect effects on forest dynamics, including altering the disturbance patterns of herbivores and changing the abundance of natural enemies. Warmer air temperatures can impact the distribution and survival of arthropods, both novel species and resident natural enemies. Gall-inducing mites are a resident natural enemy in high latitude forests and have been shown to drive dramatic reductions in gas-exchange rates in infested plants. Industrially important metals, such as Cu and Ni, sometimes are present at elevated concentrations in lakes, including those in the Sudbury, ON region. Although they are essential metals, their divalent-cation state (Cu2+ and Ni2+) can be toxic at high concentrations in the water. The mechanisms behind metal-metal interactions in freshwater environments are currently not well understood. Freshwater environments consist of many different types of metals, from those naturally present such as copper (Cu) and those that originate from anthropogenic sources like silver (Ag). Both Cu and Ag use apical sodium (Na+) channels for uptake into the gills of freshwater fish. In the gills, the mechanisms of Cu2+ and Ag+ toxicity appear to be similar to one another, which is by inhibiting Na+ /potassium (K+)- adenosine triphosphatase (NKA) and carbonic anhydrase (CA).
