The Role Of Tumor Stromal Discoidin Domain Receptor 2 (Ddr2) In Breast Cancer Metastasis., Samantha Van Hove Bayer May 2019

The Role Of Tumor Stromal Discoidin Domain Receptor 2 (Ddr2) In Breast Cancer Metastasis., Samantha Van Hove Bayer

Arts & Sciences Graduate Student Theses and Dissertations

Characteristics of breast tumor stroma, including altered collagen architecture and increased stiffness, are known to contribute to tumor invasion and metastasis. However, the cellular and molecular mechanisms by which these changes occur are not fully understood. To address this question, we used a mouse genetic model to delete Discoidin Domain Receptor 2 (DDR2) from mouse tumor stromal cells and interrogated breast cancer associated fibroblasts (CAFs) to determine the molecular events downstream of DDR2 action that may lead to changes in the tumor extracellular matrix (ECM). Our work revealed that the action of DDR2 in breast stromal cells is required for …

The Role Of Ifrd1 In The Recruitment And Function Of Reserve Stem Cells In Regeneration And Cancer, Mark Anthony Lewis May 2019

The Role Of Ifrd1 In The Recruitment And Function Of Reserve Stem Cells In Regeneration And Cancer, Mark Anthony Lewis

Arts & Sciences Graduate Student Theses and Dissertations

Mature cells can reprogram into a proliferative, progenitor-like state to repair tissue following injury and inflammation. Differentiated cells in diverse tissues can become proliferative via a dedicated, evolutionarily conserved program we termed paligenosis. We detailed how paligenosis occurs, in both gastric chief and pancreatic acinar cells, in a step-wise manner that involves: 1) autodegradation of mature cell components; 2) re-expression of progenitor genes; 3) re-entry into the cell cycle. This process is governed by mTORC1, a fundamental cellular energy sensor and regulator of protein translation. Blocking mTORC1 permitted autophagy and metaplastic gene induction but blocked cell cycle re-entry at S-phase. …

The Role Of Reactive Oxygen Species In Crotalus Atrox Venom-Induced Cell Death, Lindsay Brown May 2019

The Role Of Reactive Oxygen Species In Crotalus Atrox Venom-Induced Cell Death, Lindsay Brown

Master of Science in Integrative Biology Theses

Venomous snake bites impact humans all around the world. Anti-venom treatments mitigate systemic effects such as vascular hemorrhage, platelet aggregation inhibition, and the activation of inflammatory mediators. However, hemorrhagic snake venom also causes a loss of cellular adhesion to extracellular matrix components resulting in massive local tissue damage. To better understand the mechanism in which venom induces local tissue damage, human embryonic kidney cells (HEKS) were grown on PEI then stimulated with 500μg/ml Crotalus atrox (CA) venom for 4 and 10 hours. Alamar Blue assays were used to measure cell viability and results suggest a 15±8.6% (p<0.05) and 59±10.7% (p<0.05) reduction in cell viability at 4 and 10 hours, respectively. Cells stimulated with 500μg/ml venom for 10 hours stained 98±2.2% (p<0.05) positive for Trypan blue, suggesting the venom reduces membrane integrity. Identical treatment in the presence of 200 units PEG-catalase (PC) increased viability by 37±5.7% (p>0.001) compared to …

An Investigation Of Possible Reservoirs Of Lymphoproliferative Disease Virus Of Turkeys In Mississippi, Walker Hyche

Honors Theses

Emerging infectious diseases are a problem among wildlife and domestic populations. It is critical for these diseases to be studied. Lymphoproliferative disease virus is a retrovirus affecting turkeys and chickens. Prior research originally identified the virus in the United Kingdom and Israel. The first case of LPDV in the United States was found in 2009 in Arkansas. This study tests different reservoirs for the virus. DNA was extracted from samples of different origins and preservation types. Through PCR and gel electrophoresis analysis, different sample types from turkey populations were tested for LPDV. This study provides a method for submitting blood …

Dna Methods To Understand Genetic Variation Of The Mola Mola Populations In And Near Cape Cod, Jessica Barros

Honors Program Theses and Projects

The ocean sunfish Mola mola is a frequent summer visitor to the waters off of Southeastern Massachusetts and Cape Cod. In late fall, water temperatures drop and some animals strand on area beaches. Stranded animals provide an easily available source of DNA to study the population genetics of this species with the specific goal to learn about genetic variation within and between individuals in this population. This study will continue to identify and characterize microsatellite repeat regions that will be used to study genetic variation. Four new microsatellite regions have been identified and amplified using polymerase chain reaction conditions that …

The Effects Of Meloidogyne Incognita On Jaz1, Jaz12, And Jaz13 Gene Expression In Arabidopsis, Emily Green May 2019

The Effects Of Meloidogyne Incognita On Jaz1, Jaz12, And Jaz13 Gene Expression In Arabidopsis, Emily Green

Honors Program Theses and Projects

Meloidogyne incognita, a root-knot nematode (RKN), poses a threat to the well-being of the global agricultural industry. These endoparasites rely on plant roots and drain nutrients from crops intended for human or livestock consumption. Root knot nematodes cost the agricultural industry billions of dollars in lost crops each year and threaten efforts to meet the growing demand for food. Though plants are susceptible to dangers, such as these parasites, many have evolved defense and/or repair mechanisms to compensate for this vulnerability. The goal of this research is to study one gene family involved in such defenses. Specifically, I studied the …

Microdamage As A Bone Quality Component: Practical Guidelines For The Two‐Dimensional Analysis Of Linear Microcracks In Human Cortical Bone, Victoria M. Dominguez, Amanda M. Agnew May 2019

Microdamage As A Bone Quality Component: Practical Guidelines For The Two‐Dimensional Analysis Of Linear Microcracks In Human Cortical Bone, Victoria M. Dominguez, Amanda M. Agnew

Publications and Research

Microdamage is a component of bone quality believed to play an integral role in bone health. However, comparability between existing studies is fraught with issues due to highly variable methods of sample preparation and poorly defined quantification criteria. To address these issues, this article has two aims. First, detailed methods for preparation and analysis of linear microcracks in human ribs, specifically addressing troubleshooting issues cited in previous studies, are laid out. Second, new, partially validated criteria are proposed in an effort to reduce subjective differences in microcrack counts and measures, ensuring more comparable results between studies. Revised definitions based on …

Transformative Learning Gains In Undergraduate Learning Assistants, Caitlin Gold May 2019

Transformative Learning Gains In Undergraduate Learning Assistants, Caitlin Gold

Honors Projects

The Learning Assistant (LA) program is utilized at Bowling Green State University (BGSU) to assist in shifting classrooms from lecture based to more interactive and groupwork based learning. LAs are employed in courses with greater than 80 students enrolled that have historically high drop, withdrawal, and failure rates. Not only are students in courses with LAs having positive experiences, but the LAs themselves are as well. Interest has grown in the development of LAs during their time in the program; this can be measured in the form of transformation. Transformative learning is defined as “learning that results in transformative changes …

Investigation Of Origin And Growth Of Fungi In Coastal Beach Sand, Ruth L. Wright May 2019

Investigation Of Origin And Growth Of Fungi In Coastal Beach Sand, Ruth L. Wright

Honors Theses

In a previous study from this lab (Stevens, Evans, & Aguirre, 2012) yeast colonies were isolated and grown grown from sand collection at pristine, medium use and high use beaches along the grand strand. In that study greater yeast abundance and greater diversity both correlated with higher census of human use, suggesting that some yeast growth of sites was anthropogenic. However the sand matrix itself also varied from very fine, uniform, dark sand at the pristine beach to coarser, color varying, less tightly packed sand at residential and commercial beaches. This suggested an alternative hypothesis to differential colonization and growth …

Warming Up: Climate Change Related Shifts Of Mycorrhizal Fungal Communities In High Latitude Ecosystems, Megan Rae Devan May 2019

Warming Up: Climate Change Related Shifts Of Mycorrhizal Fungal Communities In High Latitude Ecosystems, Megan Rae Devan

Biology ETDs

This dissertation examines how climate change affects mycorrhizal fungal communities in boreal and arctic ecosystems. In chapter one, I revealed that increases in fire severity and related increases in deciduous tree dominance result in greater Ascomycota relative abundance (RA) and subsequent declines in Basidiomycota RA. In chapter two I analyzed the effects of post-fire mycorrhizal fungal communites on host growth. There were trends at the fungal genus level that were largely reflected at the guild level across all hosts; however, there were some fungal genera that had the opposite effect on different host species. In chapter three, I found host …

Effects Of The Pathogen Mycoplasma Gallisepticum Transmission On Growth And Condition Of Eastern Bluebirds Nestlings (Sialia Sialis), Gurshan Kaur May 2019

Effects Of The Pathogen Mycoplasma Gallisepticum Transmission On Growth And Condition Of Eastern Bluebirds Nestlings (Sialia Sialis), Gurshan Kaur

Honors Theses

A critical aspect to understanding disease dynamics is to understand how transmission occurs between hosts. There are two basic modes of transmission: vertical and horizontal. Vertical transmission is the passage of a pathogen from mother to offspring before or during birth, whereas horizontal transmission occurs between an infected host to a susceptible host. Mycoplasma gallisepticum (MG) is a common bacterial pathogen of poultry that causes respiratory disease. Infection of MG often results in weight loss, slowed growth rate, and increased mortality. Both vertical and horizontal transmission of MG have been found to occur in poultry. In the mid-90s, MG underwent …

Screening For Mental Health Concerns Across Pregnancy : Factors Associated With Depression And Anxiety Symptons In A Large Obgyn Clinic, Addie Keller May 2019

Screening For Mental Health Concerns Across Pregnancy : Factors Associated With Depression And Anxiety Symptons In A Large Obgyn Clinic, Addie Keller

Honors Theses

Depression and anxiety are prevalent among pregnant women, however, mental health continues to be a portion of care neglected in the obstetric setting. Depression is of-ten thought of as a disorder that leads to a persistent feeling of sadness. Anxiety is charac-terized by excessive fear and worry. Because of the many changes occurring throughout pregnancy, both physically and emotionally, it is not uncommon for pregnant women to struggle with depression, anxiety, or both. Previous studies have found that there are many factors associated with depression and anxiety throughout pregnancy. This study focuses on the relationship between age, gravida, and social …

Histone Deacetylase Inhibitors And Breast Cancer Metastasis: A Review And Exploration Of Hdac(I)S And Other Chemotherapeutic Agents, William E. Sistrunk May 2019

Histone Deacetylase Inhibitors And Breast Cancer Metastasis: A Review And Exploration Of Hdac(I)S And Other Chemotherapeutic Agents, William E. Sistrunk

Honors Theses

The traditional perspective of Histone Deacetylase enzymes is focused on their inherent epigenetic modification characteristics. While it is true that the histone modification these enzymes exhibit play a role in cancer and related diseases, Histone Deacetylase has a variety of non-histone targets. The non-histone targets include microtubules and are of specific interest because of the microtubules’ role in cell line differentiation, replication, apoptosis, and cancer metastasis. Using a variety of Histone Deacetylase Inhibitors (HDACi) and other chemotherapeutic compounds, our research group explored the HDACi effect on breast cancer cell lines. Our goal was to indicate the presence of HDACi cell-line …

Effects Of Mycoplasma Gallisepticum On Experimentally Infected Eastern Bluebirds (Sialia Sialis), Meredith A. Fry May 2019

Effects Of Mycoplasma Gallisepticum On Experimentally Infected Eastern Bluebirds (Sialia Sialis), Meredith A. Fry

Honors Theses

The bacterium Mycoplasma gallisepticum is an avian respiratory pathogen that causes inflammation and swelling of conjunctiva in domestic poultry and wild finches. In the past, severity of conjunctivitis symptoms has been used to quantify infection severity and host resistance. However, many songbirds function as hosts to M. gallisepticum but do not develop this symptom at all. The absence of conjunctivitis, as well as other clinical signs, hinders our understanding of M. gallisepticum infection in the songbird community because some species are responding very differently than others. One such species is the Eastern Bluebird (Sialia sialis), a common songbird of the …

Impacts Of Residential Garden Practice On Bird And Butterfly Communities In Southeast Kansas, Katie Mcmurry May 2019

Impacts Of Residential Garden Practice On Bird And Butterfly Communities In Southeast Kansas, Katie Mcmurry

Electronic Theses & Dissertations

Ecological research indicates that global species richness is declining due to a combination of urbanization and human population growth that fragments and simplifies the landscape. As these rates continue to grow, the value of urban greenspaces increases. If managed properly, residential greenspaces can help to maintain species richness and mitigate landscape simplifications. However, residential greenspaces are often the result of a number of individual management decisions that are vastly different from historically native vegetation. Additionally, management decisions are closely related to a number of sociodemographic factors such as age, education, home ownership, and income that influences management behaviors. I surveyed …

Fungi In A Hot, Dry, Changing World, Miriam I. Hutchinson May 2019

Fungi In A Hot, Dry, Changing World, Miriam I. Hutchinson

Biology ETDs

My doctoral work focused on understanding the reciprocal relationship between fungi and their environment, namely how fungi respond to environmental flux, as well as how fungi can modify and structure their habitats, especially in the context of climate change. As such, I aimed my research on fungi with distinct adaptations to their environmental niches: endophytic fungi that inhabit plant tissue and thermophilic fungi that are capable of growing at the upper temperature limit for eukaryotic life. My research consisted of three studies. First, I investigated the thermophilic species Myceliophthora heterothallica to demonstrate its use as a model organism for efficient …

Developing Sensory Behavioral Assays For Zebrafish Autism Model, Shannon Wagner May 2019

Developing Sensory Behavioral Assays For Zebrafish Autism Model, Shannon Wagner

Honors College Theses

Individuals of all ages can suffer from a wide variety of symptoms and disabilities that could be diagnosed as Autism Spectrum Disorder (ASD). Due to new methods and technology, individuals are now being diagnosed in the first two years of their life, which is when the signs of ASD are initially exhibited. Individuals diagnosed with ASD share many similar disabilities and symptoms such as hyperactivity to social, visual, and auditory stimuli, as well as hyposensitivity to olfactory stimuli. Neural circuit-based alterations are widely considered as a cause for these behavioral aberrations. We have created behavioral assays using zebrafish larvae to …

Oil Hydrocarbon Degradation By Caspian Sea Microbial Communities, John Miller, Stephen Techtmann, Julian Fortney, Nagissa Mahmoudi, Dominique Joyner, Jiang Liu, Et. Al. May 2019

Oil Hydrocarbon Degradation By Caspian Sea Microbial Communities, John Miller, Stephen Techtmann, Julian Fortney, Nagissa Mahmoudi, Dominique Joyner, Jiang Liu, Et. Al.

Department of Biological Sciences Publications

The Caspian Sea, which is the largest landlocked body of water on the planet, receives substantial annual hydrocarbon input from anthropogenic sources (e.g., industry, agriculture, oil exploration, and extraction) and natural sources (e.g., mud volcanoes and oil seeps). The Caspian Sea also receives substantial amounts of runoff from agricultural and municipal sources, containing nutrients that have caused eutrophication and subsequent hypoxia in the deep, cold waters. The effect of decreasing oxygen saturation and cold temperatures on oil hydrocarbon biodegradation by a microbial community is not well characterized. The purpose of this study was to investigate the effect of oxic and …

O-Glcnacylation Of Hif-1Α Protein In Human Breast Cancer Cells, Joshua D. Dolsen May 2019

O-Glcnacylation Of Hif-1Α Protein In Human Breast Cancer Cells, Joshua D. Dolsen

Honors Theses

Cancer is an insidious disease that, if not caught in time, can cause devastating effects on the human body, especially if it metastasizes. It is important to note that each cancer behaves and develops differently based on its location in the body and the cellular characteristics constitute the instance of the disease. However, the development of cancer can universally be characterized by the acquisition of different hallmark abilities that promote disease progression. Proteins known as hypoxia-inducible factors (HIFs) can influence progression through these hallmarks, but primarily provides cancer cells with the ability to alter its metabolism to survive and further …

Microplastics In The Digestive System Of The Atlantic Sharpnose Shark (Rhizoprionodon Terraenovae) In Winyah Bay, Sc, Elise Virginia Pullen May 2019

Microplastics In The Digestive System Of The Atlantic Sharpnose Shark (Rhizoprionodon Terraenovae) In Winyah Bay, Sc, Elise Virginia Pullen

Electronic Theses and Dissertations

The digestive tracts and livers of adult male Atlantic sharpnose sharks (N=16), Rhizoprionodon terraenovae, from Winyah Bay, South Carolina were examined for ingested microplastics. R. terraenovae is a small, locally abundant, coastal mesopredatory elasmobranch belonging to the family Carcharhinidae. Microfibers comprised the largest categories of plastics (94% of the total), and were found in 100% of sharks examined. The number of micro- and other plastics ranged from 34 to 75 per individual and totaled 927. The majority of plastics (40%) were blue in coloration, and 55% were <1 mm in length. Microplastics were observed on both the interior and exterior of the organs examined, and three microfibers were embedded within the stomach lining, an observation not previously reported in marine vertebrates and one which represents a potential pathway for the translocation of ingested microplastics.

Bacteriophages Of The Lower Urinary Tract, Andrea Garretto, Taylor Miller-Ensminger, Alan J. Wolfe, Catherine Putonti May 2019

Bacteriophages Of The Lower Urinary Tract, Andrea Garretto, Taylor Miller-Ensminger, Alan J. Wolfe, Catherine Putonti

Bioinformatics Faculty Publications

Phages are vital members of the microbiota, having critical roles in shaping bacterial metabolism and community structure. Although phages have been discovered in the urinary tract, such as phages that infect Escherichia coli, sampling them is challenging owing to low biomass, possible contamination when using non-invasive methods and the invasiveness of methods that reduce the potential for contamination. Phages could influence bladder health, but an understanding of the association between phage communities, bacterial populations and bladder health is in its infancy. However, evidence suggests that phages can defend the host against pathogenic bacteria and, therefore, modulation of the microbiome …

Biogeographical Implications Of Climate Change For An Alpine Mammal, The American Pika, Marie Louise Westover May 2019

Biogeographical Implications Of Climate Change For An Alpine Mammal, The American Pika, Marie Louise Westover

Biology ETDs

Anthropogenic climate change has already impacted a majority of species globally. The aim of this dissertation is to understand how climate and climate change influences animal ecology and evolution across space and time, using the American pika (Ochotona. princeps) as a model system. I investigate how pika body size, diet, and occupancy are influenced by different aspects of climate over space and time. Body size in O. princeps populations best correlates to precipitation and vegetation, rather than temperature. Our findings suggest that pika body size may be more related to vegetation and food availability than the direct effects …

Rising Rural Body-Mass Index Is The Main Driver Of The Global Obesity Epidemic In Adults, Con Burns, Tara Coppinger, Janette Walton, Et Al

Publications

Body-mass index (BMI) has increased steadily in most countries in parallel with a rise in the proportion of the population who live in cities1,2. This has led to a widely reported view that urbanization is one of the most important drivers of the global rise in obesity3,4,5,6. Here we use 2,009 population-based studies, with measurements of height and weight in more than 112 million adults, to report national, regional and global trends in mean BMI segregated by place of residence (a rural or urban area) from 1985 to …

Amino-Terminal Sequencing Of Ssv1 Major Capsid Protein Vp1 Mutants, Nichole D. Procter, Kenneth M. Stedman, Ignacio De La Higuera, George Kasun, Ajay Sapre

Student Research Symposium

Unique lemon-shaped viruses, the Fuselloviridae, or Sulfolobus Spindle-Shaped Viruses (SSVs) of hyperthermophilic archaea are characterized by both their unusual structure and extreme thermal tolerance. The most well characterized of the SSVs is SSV1. Wild-type SSV1 tolerates multiple mutations to its genome while still remaining infectious, making this virus ideal to study molecular processes in extreme environments. The major capsid protein, VP1 forms the unique structure of the virus and provides protection for the genome. The amino terminus of the VP1 protein purified from wild-type SSV1 is glutamate 66 (E66) indicating proteolytic cleavage. E66 is universally conserved in all SSVs. Interestingly, …

Molecular Systematics Of Odonata, Kristin Anderson, Susan Masta May 2019

Molecular Systematics Of Odonata, Kristin Anderson, Susan Masta

Student Research Symposium

Dragonflies and damselflies together compose the order Odonata. Odonates are insectivorous, filling a crucial niche in aquatic communities. When Odonates are present in communities, it's a sign of ecosystem health. Because of their comparatively large size and bright colors, they are often popular insects with the public and can serve as a flagship species in conservation initiatives. Odonate systematics has been greatly debated in the literature over last hundred years, and there are still many unanswered questions to ponder. This study tests paraphyletic families and the deepest nodes of evolutionary divergence within the order Odonata. Sequences accessed from GenBank NCBI …

Neuroprotection With Novel Anti-Oxidant And Cannabinoid-Based Small Molecules, Jacqueline R. Mcgrath May 2019

Neuroprotection With Novel Anti-Oxidant And Cannabinoid-Based Small Molecules, Jacqueline R. Mcgrath

Honors Theses

Neurodegenerative diseases are caused by various underlying mechanisms within the nervous system. Excitotoxicity is a hallmark of many neurodegenerative diseases, and is produced by over excitation of glutamate receptors on neurons, ultimately leading to cell death. Protein aggregates and oxidative stress have also induced neuronal cell death that lead to many neurodegenerative diseases as well. Many compounds isolated from plants have shown protective ability against these underlying toxicity mechanisms. In this study, we tested several cannabinoid and anthyocanin-based small molecules protective abilities against excitotoxicity. We found several compounds that protected against glutamate-induced excitotoxicity in a concentration-dependent manner. We also tested …

Honors Theses

Health professionals consider improper posture as a risk factor for musculoskeletal disorders, chronic pain, and additional health issues. The chief aim of my study was to investigate links between poor posture and health, particularly in college students. To collect data regarding postural health, I distributed an exploratory survey to Human Anatomy and Physiology II (BISC 207) students at the University of Mississippi in Spring 2018. Students reported significant concern with quality of personal posture and significant experience with negative side effects of poor posture. Results demonstrated a need for postural education, leading to phase 2 of my experiment, the evaluation …

Coastal Ecosystem Investigations With Lidar (Light Detection And Ranging) And Bottom Reflectance: Lake Superior Reef Threatened By Migrating Tailings, Charlie Kerfoot, Martin M. Hobmeier, Sarah Green, Foad Yousef, Colin Brooks, Robert Shuchman, Michael Sayers, Et Al. May 2019

Coastal Ecosystem Investigations With Lidar (Light Detection And Ranging) And Bottom Reflectance: Lake Superior Reef Threatened By Migrating Tailings, Charlie Kerfoot, Martin M. Hobmeier, Sarah Green, Foad Yousef, Colin Brooks, Robert Shuchman, Michael Sayers, Et Al.

Michigan Tech Publications, Part 1

Where light penetration is excellent, the combination of LiDAR (Light Detection And Ranging) and passive bottom reflectance (multispectral, hyperspectral) greatly aids environmental studies. Over a century ago, two stamp mills (Mohawk and Wolverine) released 22.7 million metric tons of copper-rich tailings into Grand Traverse Bay (Lake Superior). The tailings are crushed basalt, with low albedo and spectral signatures different from natural bedrock (Jacobsville Sandstone) and bedrock-derived quartz sands. Multiple Lidar (CHARTS and CZMIL) over-flights between 2008–2016—complemented by ground-truth (Ponar sediment sampling, ROV photography) and passive bottom reflectance studies (3-band NAIP; 13-band Sentinal-2 orbital satellite; 48 and 288-band CASI)—clarified shoreline and …

Effects Of Treatment On Melanoma With Checkpoint Inhibitor, Pembrolizumab (Keytruda ®), Anti-Cd47 Tti-621 (Trillium), And Anti-Sema4d Pepinemab Combination Treatment, Lauren E. Fitzgerald May 2019

Effects Of Treatment On Melanoma With Checkpoint Inhibitor, Pembrolizumab (Keytruda ®), Anti-Cd47 Tti-621 (Trillium), And Anti-Sema4d Pepinemab Combination Treatment, Lauren E. Fitzgerald

Seton Hall University Dissertations and Theses (ETDs)

Traditionally, cancer therapies are generally non-specific treatments meant to eradicate cancer cells. This can result in death of healthy tissue, which can significantly affect the overall well-being of the patient during and after treatment. Immunotherapy offers a more targeted approach by using immune cells to specifically identify and kill cancer cells through various neoantigens presented on the surface of the cancer cell. Drugs that target and block PD-(L)1 and CTLA-4, known as checkpoint inhibitors, act to “cut the breaks” on the immune response and keep it actively seeking out and killing cancer cells. Many patients fail on these treatments due …
