The phytohormone auxin is a critical signal molecule, regulating fundamental processes in plant growth and development, such as shaping the root and shoot architecture, organ patterning, and nodulation. Auxin regulates plant gene expression mainly through auxin response factors (ARFs), which bind to auxin response elements in the promoter, upstream of auxin-activated genes. Here we examine and assess the function and expression pattern of a gene described as an auxin response factor, containing a DNA-binding pseudobarrel and B3 DNA-binding domains, from Medicago truncatula (MtARF-B3). For the model legume species M. truncatula, stable transgenic plants with MtARF-B3 overexpression, downregulation, and transcriptional reporters …

Cloning, Expression, And Characterization Of Human Brain Acetylcholinesterase In Escherichia Coli Using A Sumo Fusion Tag, Hami̇d Ceylan, Orhan Erdoğan
Turkish Journal of Biology
The molecular structure of acetylcholinesterase (AChE) attracts interest because of its versatility and significant role in the cholinergic system. The main purpose of the present study was to clone a full-length cDNA sequence of human brain acetylcholinesterase (hAChE) into pET SUMO vector and express it successfully. The integrity of the constructed plasmid was confirmed by cross PCR. This recombinant construct was expressed in Escherichia coli BL21 (DE-3). In this work, we produced hexahistidine (6xHis) tagged fusion protein by isopropyl β-D-1-thiogalactopyranoside (IPTG) induction and purified using nickel (Ni2+) affinity chromatography. Using anti-His antibody, we detected ~90 kDa fusion protein. The expression …

Human mesenchymal stromal cells (MSCs) from different parts of the body (i.e. bone marrow, BM) have distinct cellular and molecular features including global gene expression profiles. Quantitative polymerase chain reaction is a reliable method used in the quantification of gene expression. Correct assessment of target gene expression mostly depends on the reference gene (RG) of choice. Herein, expression levels of RGs (n = 19) in adipo-/osteogenic-differentiated and control (uninduced) BM-MSCs from donors were measured using the RealTime ready Human Reference Gene Panel. Characterization of BM-MSCs was assessed using in vitro differentiation by histochemical staining and immunophenotyping of cells by flow …

Plant defensins are multifunctional small cysteine-rich proteins. They are active against fungi, bacteria, and many viruses, and they inhibit trypsin and α-amylase activities. In this study, we expressed the maize defensin gene (ZmDEF1) in tobacco seeds in order to establish the basis for generating transgenic maize plants resistant to weevils. The ZmDEF1 gene was isolated from Maison, a Vietnamese local maize cultivar, which is well known for having the highest resistance to weevils among other local cultivars. The ZmDEF1 gene was cloned into a binary vector, pBetaPhaso-dest, which carries phaseolin, a seed-specific promoter, to direct defensin expression in tobacco seeds. …

ING1 has regulatory roles in the expression of genes associated with proliferation, apoptosis, and senescence. p33(ING1b) is the most widely expressed isoform of the gene. Downregulation of its nuclear expression is involved in differentiation and pathogenesis in invasive breast carcinoma. Yet the mechanism(s) by which p33 nuclear targeting is regulated remains unknown. In this study, we analyzed human invasive breast carcinoma tissue samples by immunostaining with p33 and correlating p33 location with the presence of ERα. Our findings show the expression of p33 protein in ERα-positive tumor samples was in the nucleus alone, while the expression was mainly in the …

The use of quercetin as a bioflavonoid is becoming increasingly common in food industries even though poor water solubility, instability, absorption, and permeability have limited its application. The oil-in-water single-emulsion solvent evaporation method to synthesize highly stable and soluble quercetin-encapsulated nanoparticles (NPs), in which the reaction yield, particle size, and polydispersity of the NPs are varied greatly within the process parameters of the synthesis method, has been optimized. NPs with different initial quercetin amounts were used to determine how the quercetin amount affected nanoparticle properties and antimicrobial eficiency. Listeria monocytogenes, Salmonella typhimurium, Escherichia coli, and Staphylococcus aureus were chosen as …

Turkish Journal of Biology
The use of transition metal complexes as appropriate analogues to cisplatin and similar anticancer drugs nowadays is of utmost importance. The inertness of Pt(IV), Pd(II), and Rh(III) complexes and the choosing of convenient ligands could offer a possible advantage in comparison to Pt(II) complexes. These advantages could be attributed to the better anticancer activity and fewer possible side effects. In this paper, we have chosen three complexes of different transition metal ions, [(TLtBu)PdIICl]ClO4, [RhIII(LH2tBu)Cl3], and [PtIV(LH2tBu)Cl3]Cl (where TLtBu is 2,6-bis[(1,3-di-tert-butylimidazolin-2-imino)methyl]pyridine and LH2tBu is 2,6-bis(5-tert-butyl-1H-pyrazol-3-yl)pyridine), for biological tests. All three complexes exerted strong prooxidative and cytotoxic effects on the human colorectal …

Sweet potato is one of the most important food crops with strong environmental adaptability. Protein phosphatase 2A (PP2A) is a major intracellular protein phosphatase that plays crucial roles in hormone signal transduction and abiotic stress response. Characterization of PP2A can elucidate the mechanisms of stress resistance in this crop. In this study, a total of 15 PP2A transcripts, including nine regulatory subunit-encoding sequences and six catalytic subunit-encoding sequences, were identified from sweet potato and found to be expressed at varying levels. Only one contained a complete open reading frame and encoded a B regulatory subunit (termed IbPP2A1). Ten polymorphic sites …

Somatic embryogenesis is a powerful tool for clonal propagation and plant breeding in many plant species, but its application is limited by the genotype response based on the interaction between gene expression and culture conditions. In this context, the recalcitrance of the nonembryogenic cherry rootstock Prunus cerasus cultivar CAB 6P was investigated via the analysis of the candidate genes PiABP19, Picdc2, and PiSERK3 in comparison with the embryogenic genotype Prunus incisa cultivar N°131. Semiquantitative RT-PCR was employed to assess the transcript level of these genes in leaf explants and derived calli of the two genotypes cultured on picloram-containing MS medium …

Lipocalin Gene Expression Is Varied In Developmental Stages By Larval Nutritional Stress In Drosophila, Nazli Ayhan, Pinar Güler, Banu Şebnem Önder
Turkish Journal of Biology
Lipocalin genes NLaz, GLaz, and Karl are evolutionarily conserved genes in Drosophila melanogaster. There are studies on lipocalin gene expression differences under diverse diet conditions, but these studies have focused mainly on age-dependent expression profiles of these genes. The main aim of our study is to determine lipocalin expression in the developmental period by nutritional manipulation with an isofemale-based design. Three larval developmental periods have been researched under normal and restricted diets. We found significant differences between lines during their developmental time-related lipocalin expression. Here, we demonstrate that upregulations in the early developmental stages of lipocalin genes under stressful conditions …

The vacuolar (H+)-ATPases that pump H+ from the cytoplasm to extracellular compartments can alter the pH of the tumor microenvironment. Esomeprazole can effectively inhibit vacuolar (H+)-ATPases and may increase the effectiveness of chemotherapeutics. Therefore, we used esomeprazole in combination with cisplatin, carboplatin, paclitaxel, docetaxel, gemcitabine, and vinorelbine on the A549 nonsmall-cell lung cancer cell line. Cisplatin and carboplatin combinations with esomeprazole exhibited superior cytotoxicity compared to the other selected chemotherapeutics. Low-dose combinations of esomeprazole with either cisplatin or carboplatin resulted in synergistic interaction. We examined cytotoxic activity of these combinations with the xCELLigence real-time cytotoxicity assay and detected that esomeprazole …

Microrna-34a Regulates Brain-Derived Neurotrophic Factorin An Intracerebral Hemorrhage Model, Jin Hee Kim, Jae-Sun Choi
Turkish Journal of Biology
Intracerebral hemorrhages (ICHs) are devastating neurological events frequently resulting in a serious negative prognosis. The exact physiological and disease processes involved in ICHs are complex, but are thought to involve microRNAs (miRNAs), 22 nucleotide small noncoding RNAs that control a variety of normal physiological and disease processes. In this study, we show that a miRNA, miR-34a, regulates BDNF in a model of ICH injury. In particular, we assessed the impact of AM34a, an inhibitor of miR-34a, on the toxicity of thrombin-induced apoptosis and on BDNF-mediated signaling. We investigated the increased expression of miR-34a after an ICH-induced thrombin toxicity injury using …

Obesity is one of the most widespread nutritional diseases in developed societies and it is considered a cardiovascular disease risk factor. The aim of the present work was to evaluate how a high-fat diet may influence the chronobiology of glucose and albumin circadian rhythms. Eighty-four male common Wistar rats were separated into two groups: a control group (n = 42) and a group fed a high-fat diet (n = 42); both for the control and the obesity-induced group we stablished 7 subgroups (6 rats per subgroup) to take blood samples at 0000, 0200, 0400, 1000, 1400, 1800, and 2200 hours. …

Turkish Journal of Biology
The main purpose of this study was to analyze the cytotoxic activity of an extract obtained from Centaurea fenzlii Reichardt, and the fractions eluted from this extract, in breast cancer cells. After isolation and structural analysis of the fractions were conducted, a meaningful cytotoxic effect was indicated. The goal of the analysis was to reveal the mechanism by which this effect occurs through researching the apoptotic side of these fractions and determining the amount of several proteins that are the products of the genes. Test substances were applied to breast cancer cells and the inhibitory concentration value 50 (IC50) that …

In view of the increasing demand for essential oils from medicinal and aromatic plants, the application of irradiated polysaccharides like chitosan is a promising option. A pot experiment was performed to study the influence of various concentrations of irradiated chitosan (ICN) on growth, physiological, and yield parameters of Mentha piperita L., a plant with considerable pharmaceutical importance. The treatments were foliar spray of deionized water only (control), foliar spray of unirradiated chitosan (40 mg L-1), and ICN at 40, 80, 120, and 160 mg L-1. Among these, 80 mg L-1 ICN (treatment 4) proved to be the best for most …

Turkish Journal of Biology
The collaboration between the Human Genome Project and the Sanger Institute, spurred on by their contemporary- Celera Genomics, in the race to sequence the human genome, opened the door to a generation of research concerned with the study of the nucleic manual. Following this monumental achievement, the genomic era was born, heralding the development of gene therapy, personalized medicine, pharmacogenomics, and CRISPR/Cas9 gene-editing systems. These 'omic' disciplines offer a wide range of applications in the treatment of disease and the betterment of the quality of human life. Their notability, however, manifests in the unique mix of sociological and scientific discourse …

The Role Of Predicted Spermidine Family Transporters In Stressresponse And Cell Cycle In Schizosaccharomyces Pombe, Aslihan Gevrekci̇
Turkish Journal of Biology
Fission yeast Schizosaccharomyces pombe has a variety of stress-signaling proteins that protect cells against environmental or intracellular stress. These proteins help the cells to respond to stress conditions and regulate intracellular functions such as cell division or gene expression. Polyamines (spermidine, spermine, and putrescine) are known to be important in the regulation of stress response and cell division. In this study, we tried to experimentally characterize novel S. pombe genes that are involved in the polyamine pathway and understand their potential roles. Sequence analysis revealed four genes that code for (predicted) spermidine family transporters in S. pombe. In an attempt …
