Production Of 2,3-Butanediol By Enterobacter Cloacae From Corncob-Derived Xylose, Cuiying Zhang, Wei Li, Dongsheng Wang, Xuewu Guo, Lijuan Ma, Dongguang Xiao
Turkish Journal of Biology
During the process of industrial production of biofuels and biochemicals from lignocellulosic biomass, large amounts of waste byproducts rich in xylose are generated, resulting in excessive wastage of natural resources and environmental pollution. In this work, xylose solution from corncob hydrolysate was utilized to produce 2,3-butanediol (2,3-BD), using the strain Enterobacter cloacae CICC 10011 to improve the utilization rate of hemicellulose and reduce environmental pollution. 2,3-BD fermentation conditions were subsequently developed. Xylose solution and (NH4)2HPO4, selected with the Plackett-Burman experiment, were determined as significant independent variables to conduct a response surface experiment. With the optimized medium, 50.02 g/L 2,3-BD production …

Germline Transformation Of The Olive Fruit Fly, Bactrocera Oleae(Rossi) (Diptera: Tephritidae), With A Piggybac Transposon Vector, Hani̇fe Genç, Marc F. Schetelig, Xavier Nirmala, Alfred M. Handler
Turkish Journal of Biology
The olive fruit fly, Bactrocera oleae, is a highly significant pest in olive growing countries, and controlling it may be enhanced by using genetically modified strains, especially for sterile insect technique programs. To improve and expand this technology, piggyBac-mediated germline transformation was achieved in a laboratory-adapted wild olive fruit fly strain. A piggyBac vector was used that is marked with both green (EGFP) and red (DsRed) fluorescent protein genes, with a duplicate piggyBac 5' terminal inverted repeat sequence inserted between the marker genes for subsequent immobilization of vectors integrated into the host genome. Five transformant G1 adults were selected based …

We investigated the implication of AMP-activated protein kinase (AMPK) in melatonin-induced protection in a renal ischemia/reperfusion (I/R) model. Animals were divided into four groups: rats of the sham group were not subjected to renal I/R. Rats in the I/R group received the vehicle 30 min before renal ischemia for 1 h followed by reperfusion for 6 h. Rats in the Mel+I/R group received melatonin (10 mg/kg) 30 min before the onset of I/R. Rats in the Mel+I/R+araA group received araA (AMPK inhibitor, 100 μg/kg per min for 10 min) just before melatonin administration. The results showed that melatonin treatment induced …

Turkish Journal of Biology
Sirtuins are NAD+-dependent deacetylases and ADP ribosyltransferases that are activated under stress conditions such as calorie restriction and starvation. Boron, which is mostly found in the form of boric acid (BA) or sodium borate (NaB), is known to bind NAD+. In this study, the effect of NaB on hepatoma cell line HEP3B was investigated by analyzing the proteins harvested from NaB-treated and serum-starved HEP3B cells. The NaB treatment (15 μg/mL) led to a decrease in the overall proteome acetylation and particularly in mitochondria, the synthesis rate of respiratory complexes, the amount of cellular reactive oxygen species, and the proliferation rate …

Sox6, a member of the Sox (Sry-related HMG box) family of transcription factors, plays an important role in embryonic muscle development and adult fast myofiber maintenance. Here we report the expression and purification of a His-tagged version of recombinant porcine Sox6 (pSox6) in Escherichia coli. The recombinant pSox6 was expressed as a C-terminal fusion protein with His6 tag in the E. Coli Rosetta (DE3) host strain. The protein was purified by Ni affinity chromatography, yielding approximately 5 μg/mL. The protein was identified by western blot analysis and confirmed by matrix-assisted laser desorption/ionization time-of-flight/time-of-flight (MALDI-TOF/TOF) mass spectrometer analysis. Doubled haploid (DH) plant production plays an important role in pepper (Capsicum annuum L.) breeding for development of new varieties. However, information is lacking for DH plant production in pepper genotypes carrying ornamental value. The major goal of this study was to produce DH ornamental pepper lines having the mentioned value. Androgenic responses of 48 genotypes at the F2 or F3 generation were compared in each of three androgenesis protocols to determine the most effective method. Of the three protocols tested, anthers were placed on two different semisolid culture media and on a double-layer medium also called shed-microspore culture medium. …

Physical exercise is beneficial for bone tissue health, yet its usage is limited for preventing osteoporosis. Even though natural for the bone tissue from development to homeostasis, mechanical loads present with a multitude of physical parameters, including amplitude, duration, frequency, and distribution. Utilizing the most beneficial parameters of mechanical loads may potentiate a nonpharmaceutical tool for biotechnology to prevent and treat bone loss related to aging, bedrest, sedentary lifestyles, weightlessness, and other diseases. Low intensity vibrations (LIVs) consist of mechanical loads with amplitudes smaller than loads prescribed by habitual activity, with a higher frequency. In this review, literature covering LIV …

Cellular And Molecular Basis Of Cardiac Regeneration, Justin Judd, Wanling Xuan, Guo N. Huang
Turkish Journal of Biology
Heart disease is one of the largest causes of death in humans throughout the developed world. Despite its critical role in maintaining physiologic homeostasis, the adult mammalian heart does not exhibit a significant capacity for regeneration. By contrast, several lower vertebrates exhibit a remarkable ability to regenerate the aging myocardium in response to severe injury. Additionally, recent evidence has shed light on a transient regenerative window in postnatal mammals. In this review, we discuss key findings that help to unravel the differences in cardiac regeneration across phylogeny and ontogeny. Furthermore, we highlight recent significant progress in the pursuit of adult …

Increased fragility of long bones and delay in fracture healing in adults as well as skeletal development disorders in children are thought to be associated with endochondral ossification problems. The extracellular matrix (ECM) of cartilage is absorbed during endochondral ossification. During the recovery process from damage, chondrocytes undergo hypertrophy and increase their cell volume; on the other hand, it is also necessary to digest the ECM elements around the cells. Recent research on the proteolytic enzymes in ECM assumed to be responsible for the digestion is connected to proteinases digesting type II collagen and aggrecan. ADAMTS1, 4, and 5 are …

Findings in the last decade suggest that there is a considerable amount of cardiomyocyte turnover in the human heart throughout life, albeit not sufficient for heart regeneration following myocardial infarctions. Only a few species are known to be remarkably efficient in cardiac regeneration. They restore lost cardiomyocytes via a process of cardiomyocyte dedifferentiation, which is followed by robust proliferation of cardiomyocytes and incorporation into the myocardium. Similarly, neonatal mice have been recently shown to regenerate their heart following myocardial injuries. Studies with a neonatal cardiac regeneration mouse model suggest that the major source of new cardiomyocytes is likely to be …

Turkish Journal of Biology
Cardiomyogenic differentiation from mesenchymal stem cells has emerged as a novel approach for repair of damaged myocardium. Cell transplantation through direct cell injection is not an optimal method due to the lack of cell-extracellular matrix interactions. In the present study, differentiation potential of human adipose-derived stem cells (ASCs) to cardiomyocytes has been investigated by growing them on hyaluronic acid/gelatin (HA/G) plasma gels and coverslips and supplementing the growth medium with chemical modifiers (activin-a, BMP-4, insulin, valproic acid, and 5-azacytidine) in various combinations. The HA/G plasma gels were produced from human blood plasma-derived fibrinogen, gelatin, and human umbilical cord-derived hyaluronic acid. …

A Matter Of Regeneration And Repair: Caspases As The Key Molecules, Şehnaz Bolkent, Füsün Öztay, Selda Oktayoğlu, Serap Sancar Baş, Ayşe Karatuğ
Turkish Journal of Biology
Researchers have been focused on understanding the pathogenesis of human diseases. They have been working to find the roles of caspases in the balance between apoptosis, autophagy, pyroptosis, and necroptosis, and also in the regeneration of damaged tissue. At this point, besides their death-inducing roles, new findings indicate the role of caspases in proliferation for maintaining the viability of cells in response to signals from apoptotic cells. Recently, determining cell fate has also been identified among other functions of caspases. Findings indicate that caspases direct cellular pathways to cell differentiation by suppressing stem cell self-renewal. The common opinion about the …

Collagen/Peo/Gold Nanofibrous Matrices For Skin Tissue Engineering, Ömer Aktürk, Di̇lek Keski̇n
Turkish Journal of Biology
As a novel approach in skin tissue engineering, gold nanoparticles (AuNPs) were synthesized and incorporated at different concentrations into collagen/PEO nanofibrous matrices in this study. The group containing 14.27 ppm AuNPs (CM-Au) had the best nanofibrous morphology. CM-Au was cross-linked with glutaraldehyde vapor (CM-AuX). All groups were disrupted in collagenase in 2 h, but cross-linked groups and Matriderm1397904493 resisted hydrolytic degradation for 7 and 14 days, respectively. Due to its small pores and dense structure, lower water swelling results (7.26 ± 2.62 g/g) were obtained for CM-AuX than Matriderm (17.51 ± 1.97 g/g). CM-Au and Matriderm had statistically similar tensile …

Mesenchymal stromal cells (MSCs) have been characterized as an important component of the hematopoietic niche that plays a role in regulating hematopoietic stem cell (HSC) quiescence, self-renewal, and lineage commitment. Macrophages, one kind of innate immune cells, are mainly developed from HSCs in niche. However, how MSCs and their subpopulations regulate macrophage commitment is still unknown. The current study compared the contribution of MSCs and Sca-1+MSCs to macrophage commitment and further modulation on the function of macrophages. We found that Sca-1+MSCs could promote macrophage commitment through the M-CSFR gene, and Sca-1+MSCs led macrophage polarization towards the M2 phenotype. Further functional …

Human corneal endothelium (HCE) is vital in maintaining corneal transparency and thickness, and damages to it may result in endotheliopathy and even corneal blindness, which can only be cured by keratoplasty. Due to the shortage of donor corneas, tissue-engineered HCE (TE-HCE) has become an equivalent to HCE. This study constructs a high endothelial cell density (ECD) TE-HCE and evaluates its functions by corneal transplantation in rabbits. A TE-HCE was constructed by culturing DiI-labeled nontransfected HCE cells on modified denuded amniotic membrane (mdAM) using collagen IV, fibronectin, and laminin, and rabbit corneas were characterized both in vivo and ex vivo after …

This study investigated the effect of transplantation of bone marrow mesenchymal stem cells (BM-MSCs) on functional deterioration and neuroinflammatory response in aged C57BL/6J mice. Mice were divided into 3 groups: normal aged group, saline control group, and BM-MSCs treatment group. Saline and BM-MSCs were administrated by intraventricular injection into the right ventricle. Spatial Y-maze (SYM) test, novel objective recognition (NOR) test, and locomotor activity test were used to evaluate cognitive and locomotor ability at 7 and 42 days after transplantation. Microglia and astrocyte expression in the hippocampus CA1 region and dentate gyrus (DG) were evaluated by immunohistochemical method. Furthermore, the …

Evidence indicates that reactive astrocytes play an important role in neurological functional amelioration after stroke. Bone marrow stromal cell (BMSC) transplantation-promoted astrocyte-derived neurotrophic factors' expressions resulted in decreasing the number of apoptotic neurons and improving neurogenesis and angiogenesis in the ischemic boundary zone. Recent studies found that simvastatin had neuroprotective activity in acute stroke. However, it is still ambiguous as to whether simvastatin combined with BMSCs can further activate astrocytes, which contribute to neurological functional recovery after ischemic stroke. In our present study, we established a middle cerebral artery occlusion model in rats and found that combination treatment of simvastatin …

The effects of NaCl on expression of some genes related to salinity tolerance in grape (Vitis L.) were investigated. According to our screening study on eighteen grape genotypes, H6 and Gharashani (tolerant) and Shirazi and GhezelUzum (sensitive) were selected for molecular analysis. Our tolerant genotypes showed a higher water potential and a lower MDA content compared to other genotypes. Plants were treated with 50 mM NaCl as a critical concentration that is not lethal for grape. The expression profile of VvNHL1 in leaves of all genotypes and in roots of tolerant genotypes was similar to that of VvEDS1; these genes …

Eichhornia crassipes is an abundant floating aquatic weed that has great potential for cadmium (Cd) remediation owing to its large biomass and relatively high tolerance and accumulation capabilities. This study was conducted with Eichhornia in 5, 10, 15, and 20 mg L-1 CdCl2 in a hydroponic system for 21 days, and the Cd concentrations in the roots, shoots, and leaves were estimated. The plant showed tolerance, but at high Cd concentrations declines in biomass, root length, and leaf area were observed. Leaves showed a progressive decline in chlorophyll, carotenoid, and soluble protein and a significant elevation in lipid peroxidation. Cd …

Glycation of proteins is a nonenzymatic process in which proteins react with reducing sugar molecules. This process takes place at ?-amino (?-NH+) groups of lysine or hydroxylysine residues as well as α-amino groups of N-terminal residues. In the present study glycation of ?-NH+ groups of lysines was computationally analyzed for 26 proteins based on their 3D structures. We determined the spatial relationship with acidic or basic residues and correlated them with the glycation prediction algorithm Netglycate 1.0 software, which employed primary structure exclusively for glycation site prediction. Of the lysines from 19 of the 20 proteins employed to build the …

The recent progress in DNA sequencing and computational algorithms dramatically heightened the value of single nucleotide polymorphism (SNP) databases. The experimental studies of mutation for the particular gene of interest are laborious and time-consuming. In this study, we used several computational algorithms to investigate the structural and functional consequences of SNPs on the glyceraldehyde 3-phosphate dehydrogenase (GAPDH) gene of zebrafish (Danio rerio). The GAPDH gene is involved in multiple cellular activities, in addition to its association with classical glycolytic pathway. It has most commonly been used as the housekeeping gene for differential gene expression profiling, including western blots. The computational …
