The raffinose family of oligosaccharides (RFOs) serve as transport carbohydrates in the phloem, storage compounds in sink tissues, and putative biological agents to combat both abiotic and biotic stress in several plant species. To investigate further the functional roles of this class of compounds in trees, two cDNAs encoding galactinol synthase (GolS, EC 2.4.1.123), which catalyses the first step in the biosynthesis of RFOs, were identified and cloned from hybrid poplar (Populus alba3grandidentata). Phylogenetic analyses of the Populus GolS isoforms with other known GolS proteins suggested a putative role for these enzymes during biotic or abiotic stress in hybrid poplar. …

Effectiveness And Utility Of Acoustic Recordings For Surveying Tropical Birds, Antonio Celis-Murillo, Jill L. Deppe, Michael P. Ward
Faculty Research & Creative Activity
Although acoustic recordings have recently gained popularity as an alternative to point counts for surveying birds, little is known about the relative performance of the two methods for detecting tropical bird species across multiple vegetation types. During June and July 2008, we collected species detection/nondetection data to compare the performance of a quadraphonic acoustic recording system and point counts for estimating species richness and composition and detection probabilities of 15 rare, moderately common, and common tropical bird species across six structurally distinct vegetation types (coastal dune scrub, mangrove, low-stature deciduous thorn forest, early and late successional medium-stature semievergreen forest, and …

Since the dawn of abstract thinking, humans have wondered about the seemingly unnecessary elaborate ornamentations of birds. Gaudy colours, cumbersome tails, complex vocalizations and bizarre displays are found in bird species from all corners of the globe. Darwin (1871) provided an elegant explanation for the existence of these non-utilitarian traits: they increase mating success, and although they may impair survival, the costs of producing and bearing elaborate ornaments can be repaid in the currency of additional offspring. Darwin’s model still serves as the foundation for our concept of sexual selection but great strides have been made in our understanding of …

Carotenoid-Based Ornaments Of Female And Male American Goldfinches (Spinus Tristis) Show Sex-Specific Correlations With Immune Function And Metabolic Rate, R. J. Kelly, Troy G. Murphy, K. A. Tarvin, G. Burness
Biology Faculty Research
Conspicuous ornamentation has been linked to immunological and physiological condition in males of many species. In species where both sexes are ornamented, it is unclear whether the signal content of ornaments differs between males and females. We examined the immunological and physiological correlates of carotenoid-based bill and plumage ornamentation in American goldfinches Spinus tristis, a species in which bright orange bills are sexually monomorphic but yellow plumage is sexually dimorphic during the breeding season. Because bill color is dynamic over short periods while plumage color is static over longer time frames, we tested whether these signals have the potential to …

To effectively balance investment in predator defenses versus other traits, organisms must accurately assess predation risk. Chemical cues caused by predation events are indicators of risk for prey in a wide variety of systems, but the relationship between how prey perceive risk in relation to the amount of prey consumed by predators is poorly understood. While per capita predation rate is often used as the metric of relative risk, studies aimed at quantifying predator-induced defenses commonly control biomass of prey consumed as the metric of risk. However, biomass consumed can change by altering either the number or size of prey …
Dopamine-Beta-Hydroxylase And Norepinephrine Transporter Immunoreactivity In Rat Taste Buds, Jenna Ashley Walton
Dopamine-Beta-Hydroxylase And Norepinephrine Transporter Immunoreactivity In Rat Taste Buds, Jenna Ashley Walton
Electronic Theses and Dissertations
Of the handful of neurotransmitters modulating taste response, norepinephrine (NE) may be the least understood. Although the physiological role of NE has been tested, it is still unclear whether the rat taste bud may serve as an endogenous source of neurotransmitter or if it must be taken up from outside the taste bud. Immunocytochemical analysis of DBH has shown that DBH–LIR is present in taste cells of the rat circumvallate papillae. Specifically, DBH–LIR is present in a subset of Type II taste cells. Nearly all (98%) DBH–LIR cells express PLCβ2–LIR, and about half (41%) of PLCβ2–LIR cells express DBH–LIR. DBH–LIR …
Estradiol Modulation Of Calcium Dynamics In Pituitary Mmq Lactotroph Cells, Monika Pauckova
Estradiol Modulation Of Calcium Dynamics In Pituitary Mmq Lactotroph Cells, Monika Pauckova
Electronic Theses and Dissertations
Pituitary lactotrophs are excitable cells that exhibit spontaneous, calcium influx triggering prolactin (PRL) secretion to stimulate lactation. Lactotrophs express estrogen receptors (ER) and are a well established estrogen-responsive cell system. 17B-estradiol (E2) is known to directly affect lactotrophs by increasing PRL transcription and biosynthesis, intracellular Ca2+ concentration ([Ca2+]i) and PRL secretion. This study demonstrates that the MMQ clonal cell line, isolated from the 7315a rat pituitary tumor, is a model lactotroph cell line that is E2-responsive. Spontaneous and evoked Ca2+ transients were especially sensitive to L-type channel block, but not affected by block of omega-conotoxin-GVIA-sensitive CaV channels or TTX-sensitive voltage-gated …

Pomc Is Expressed In Pancreatic Alpha Cells, Samia M.M Farrara
Electronic Theses and Dissertations
Pancreatic alpha cells secrete glucagon to increase blood glucose during hypoglycemia. Currently, the mechanisms that initiate glucagon secretion are not well understood. This study investigates the expression of pro-opiomelanocortin (POMC), a potent regulator of glucagon secretion, in alpha cells.
Insulin tolerance tests in mice lacking the POMC gene showed a correlation between alpha MSH, and glucagon secretion from alpha cells (Hochgeschwender et al. 2003). Results from a previous study showed that the POMC product, melanocortin á-MSH, acts through a melanocortin receptor 4 (MC4R) at the surface of the alpha cells to trigger the release of glucagon [Angleson, unpublished data, Lumsden …

Although several lichen inventories exist for European ultramafic sites, only four surveys of serpentine lichens for North America have been published to date. Of those, only one has been conducted in California. We conducted a survey of saxicolous lichens from ultramafic rocks (including nephrite, partially serpentinized peridotite, and serpentinite) and non-ultramafic rocks (including silica-carbonate, shale, and sandstone) at the New Idria serpentinite mass, San Benito County, California. X-ray Fluorescence Analysis of the rocks from which the lichens were collected revealed significant elemental differences between the ultramafic and non-ultramafic rocks for 26 of the 32 major and trace elements analyzed. We …
The Discovery Of Insulin: A Case Study Of Scientific Methodology, William D. Stansfield
The Discovery Of Insulin: A Case Study Of Scientific Methodology, William D. Stansfield
Biological Sciences
The nature of scientific research sometimes involves a trial-and-error procedure. Popular reviews of successful results from this approach often sanitize the story by omitting unsuccessful trials, thus painting the rosy impression that research simply follows a direct route from hypothesis to experiment to scientific discovery. The discovery of insulin is a classical case study in this genre that begs for an explanation to our students because it is so often ignored or misrepresented even in biology and physiology textbooks.

Bioluminescence In The High Arctic During The Polar Night, J. Berge, A.S. Båtnes, G. Johnsen, S.M. Blackwell, Mark A. Moline
Biological Sciences
This study examines the composition and activity of the planktonic community during the polar night in the high Arctic Kongsfjord, Svalbard. Our results are the first published evidence of bioluminescence among zooplankton during the Arctic polar night. The observations were collected by a bathyphotometer detecting bioluminescence, integrated into an autonomous underwater vehicle, to determine the concentration and intensity of bioluminescent flashes as a function of time of day and depth. To further understand community dynamics and composition, plankton nets were used to collect organisms passing through the bathyphotometer along with traditional vertical net tows. Additionally, using a moored bathyphotometer closed …
Acute Alcohol Ingestion And Sympathetic Neural Responses During Orthostatic Stress In Humans, Sarah F. Stream
Acute Alcohol Ingestion And Sympathetic Neural Responses During Orthostatic Stress In Humans, Sarah F. Stream
Dissertations, Master's Theses and Master's Reports - Open
Acute alcohol consumption has been reported to decrease mean arterial pressure (MAP) during orthostatic challenge, a response that may contribute to alcohol-mediated hypotension and eventually syncope. Muscle sympathetic nerve activity (MSNA) increases during orthostatic stress to help maintain MAP, yet the influence of alcohol on MSNA during orthostatic stress has not been determined. We hypothesized that alcohol ingestion would blunt arterial blood pressure and MSNA responses to progressive lower body negative pressure (LBNP). MAP, MSNA, and heart rate (HR) were recorded during progressive LBNP (-5, -10, -15, -20, -30, and -40 mmHg; 3 min/stage) in 30 subjects(age 24 ± 1 …

Dissertations, Master's Theses and Master's Reports - Open
Hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) is a nitramine compound that has been used heavily by the military as an explosive. Manufacturing, use, and disposal of RDX have led to several contamination sites across the United States. RDX is both persistent in the environment and a threat to human health, making its remediation vital. The use of plants to extract RDX from the soil and metabolize it once it is in the plant tissue, is being considered as a possible solution. In the present study, the tropical grass Chrysopogon zizanioides was grown hydroponically in the presence RDX at 3 different concentration levels: 0.3, 1.1, …

Hummingbirds represent an end point for small body size and water flux in vertebrates. We explored the role evaporative water loss (EWL) plays in management of their large water pool and its use in dissipating metabolic heat. We measured respiratory evaporative water loss (REWL) in hovering hummingbirds in the field (6 species) and over a range of speeds in a wind tunnel (1 species) using an open-circuit mask respirometry system. Hovering REWL during the active period was positively correlated with operative temperature (Te) likely due to some combination of an increase in the vapor-pressure deficit, increase in lung ventilation rate, …

Prostate cancer is diagnosed in over 186,000 men in the United States and accounts for more than 28,000 deaths annually. The epidemiology of prostate cancer is identified by risk factors including genetics, diet, smoking alcohol consumption, chronic inflammation, hypertension and age. Angiotensen II (AngII) is a major effector peptide of the rennin-angiotensin system (RAS), and is an important factor in hypertension. RAS interacts with the autocrine/paracrine system, and relates to cell growth and differentiation in some organs. Investigators have reported that exogenous application of Ang II to LNCaP cells results in the activation of Akt signaling, increased activity of SOD, …

The Molecular And Behavioral Characterization Of Cav1.3 Over-Expressing Mice, Jamie Nicole Slater
Master's Theses and Doctoral Dissertations
Age-related cognitive decline refers to the memory impairment and difficulty learning new tasks that occurs during the normal process of aging. There are many possible changes that occur at the neuronal level that could account for age-related cognitive decline. One hypothesis suggests that a dysregulation of neuronal intracellular calcium concentration contributes to age-related cognitive deficits. Previous research suggests that an increase in neuronal L-type voltage-gated calcium channels (LVGCCs) occurs in the brain during aging. This increase could account for altered intracellular calcium concentration and lead to agerelated cognitive decline. A line of transgenic mice that over-express the LVGCC CaV …

Amphibians are considered one of the most threatened vertebrate groups. Although numerous studies have addressed the many causes of amphibian population decline, little is known about effects of invasive plants. Chinese tallow (Triadica sebifera) is an exotic deciduous tree that has invaded the southeastern United States. Amphibian larvae in environments invaded by T. sebifera may be impacted negatively as autumn leaf litter decomposes in natal areas. We compared effects of leaf litter decomposition from T. sebifera and two native tree species on survival and development of four species of anuran larvae from eastern Texas. Larvae from Pseudacris fouquettei, Lithobates (Rana) …

Physaria ludoviciana (Nuttal) O'Kane & Al-Shehbaz (silvery bladderpod) is a state endangered species in Illinois, Minnesota and Wisconsin, the eastern edge of its range. Our goal was to investigate its seed biology (production, dispersal, seed bank longevity, afterripening, seed longevity after storage using germination and emergence, and seedling establishment). This information will be useful for conservation efforts of this endangered species. Over a ten-year period, various seed biology parameters were measured. Infructescences were counted to estimate seed production with 110–744 seeds per reproductive plant. Soil samples were collected at various distances to estimate dispersal where seeds dispersed less than a …
