Flagellation in archaea is widespread and is involved in swimming motility. Here, we demonstrate that the
structural flagellin gene from the crenarchaeaon Sulfolobus solfataricus is highly expressed in stationary-phasegrown
cells and under unfavorable nutritional conditions. A mutant in a flagellar auxiliary gene, flaJ, was
found to be nonmotile. Electron microscopic imaging of the flagellum indicates that the filaments are composed
of right-handed helices.

Culture growth and recombinant protein yield of the Pichia pastoris GS115 methanol utilization positive system were studied in response to the types and levels of metals present in the growth medium and the supplemental salts typically used for these fermentations. Magnesium and zinc were both required to support cell growth but at significantly reduced levels compared to the control. However, supplementation with calcium, cobalt, iron, manganese, iodine, boron, and molybdenum were not required to sustain cell mass. When the medium was reformulated with only zinc and magnesium, the cells grew to 12–15 generations, which are expected for high cell density …

Asymmetric Localizations Of Lin-17/Fz And Mig-5/Dsh Are Involved In The Asymmetric B Cell Division In C. Elegans, Mingfu Wu, Michael A. Herman
School of Biological Sciences: Faculty Publications
LIN-44/Wnt and LIN-17/Frizzled (Fz) function in a planar cell polarity (PCP)-like pathway to regulate the asymmetric B cell division in Caenorhabditis elegans. We observed asymmetric localization of LIN-17/Frizzled (Fz) and MIG-5/Disheveled (Dsh) during the B cell division. LIN-17∷GFP was asymmetrically localized within the B cell prior to and after the B cell division and correlated with B cell polarity. Asymmetric localization of LIN-17∷GFP was dependent upon LIN-44/Wnt and MIG-5/Dsh function. The LIN-17 transmembrane domain and a portion of the cysteine-rich domain (CRD) were required for LIN-17 function and asymmetric distribution to the B cell daughters, while the conserved KTXXXW …
An Epidemiological Model Of Rift Valley Fever, Holly D. Gaff, David M. Hartley, Nicole P. Leahy
An Epidemiological Model Of Rift Valley Fever, Holly D. Gaff, David M. Hartley, Nicole P. Leahy
Biological Sciences Faculty Publications
We present and explore a novel mathematical model of the epidemiology of Rift Valley Fever (RVF). RVF is an Old World, mosquito-borne disease affecting both livestock and humans. The model is an ordinary differential equation model for two populations of mosquito species, those that can transmit vertically and those that cannot, and for one livestock population. We analyze the model to find the stability of the disease-free equlibrium and test which model parameters affect this stability most significantly. This model is the basis for future research into the predication of future outbreaks in the Old World and the assessment of …

Detection Of Mitochondrial Insertions In The Nucleus (Numts) Of Pleistocene And Modern Muskoxen, Sergios-Orestis Kolokotronis, Ross De Macphee, Alex D. Greenwood
Biological Sciences Faculty Publications
Background: Nuclear insertions of mitochondrial sequences (NuMts) have been identified in a wide variety of organisms. Trafficking of genetic material from the mitochondria to the nucleus has occurred frequently during mammalian evolution and can lead to the production of a large pool of sequences with varying degrees of homology to organellar mitochondrial DNA (mtDNA) sequences. This presents both opportunities and challenges for forensics, population genetics, evolutionary genetics, conservation biology and the study of DNA from ancient samples. Here we present a case in which difficulties in ascertaining the organellar mtDNA sequence from modern samples hindered their comparison to ancient DNA …
Life In The Absence Of Oxygen: Alternative Electron Acceptors For Anaerobic Microorganisms In A Petroleum Environment, Meli̇ke Balk
Life In The Absence Of Oxygen: Alternative Electron Acceptors For Anaerobic Microorganisms In A Petroleum Environment, Meli̇ke Balk
Turkish Journal of Biology
Anaerobic microorganisms derive energy by transferring electrons from an external source or donor to an external electron sink or terminal acceptor and often have the capacity to reduce 2 or more terminal electron acceptors. The well-known type of microbial respiration, in which oxygen serves as an electron acceptor for the oxidation of organic carbon and/or hydrogen, has been studied elsewhere in detail. Anaerobic microorganisms are widely distributed in oil-producing vents, hydrothermal vents, volcanic hot springs, non-volcanic geothermally heated subsurface aquifers, and soil. In this study, anaerobic, thermophilic, and fermenting microorganisms in a petroleum sample from the Adıyaman region of Turkey …

In Vitro Antibacterial Studies Of Ciprofloxacin-Imines And Their Complexes With Cu(Ii),Ni(Ii),Co(Ii), And Zn(Ii), Muhammad Imran, Javed Iqbal, Shahid Iqbal, Nazia Ijaz
Turkish Journal of Biology
Some new transition metal complexes of ciprofloxacin-imines derived from ciprofloxacin and p-substituted anilines were synthesized and characterized on the basis of physical properties, conductance measurements, elemental analysis, UV/Vis., infrared and nuclear magnetic resonance spectroscopy. These ligands as well as their metal complexes were also evaluated for their antibacterial activity against several bacterial strains, such as Staphylococcus aureus, Bacillus subtilus, Salmonella typhae, and E. coli. It was found that metal complexes are more antibacterial as compared to uncomplexed ligands. The present study was carried out in search of a target antibiotic instead of a broad spectrum one.

Molecular Characterization Of Borrelia Burgdorferi Linear Plasmids By Dna Hybridization, Pcr, Two-Dimensional Gel Electrophoresis, And Electron Microscopy, Ali̇ Karami, Seyed Javad Hosseyni, Yaser Kiarudi
Turkish Journal of Biology
Borrelia burgdorferi sensu stricto is the predominant cause of Lyme disease. B. burgdorferi strains carry several linear and circular plasmids and this may be also used as a distinguishing property of the bacteria. The presence of a mixed population of linear and circular plasmids makes separation and characterization of each kind very difficult. In this study, 7 clinical strains of skin and CSF isolates were determined by clonal selection and plasmid profile of B. burgdorferi by electron microscopy and 2-dimensional agarose gel electrophoresis. The results demonstrate that the skin isolate (DK1) has 2 different phenotypes A and B, but the …

The nif genes of Clostridium beijerinckii NRRL B593 occupy a region of about 16 kilobases. Besides the two glnB-like genes, five other genes are interspersed between the nifNB and the nifVw genes. An expression analysis of the nif genes in nitrogen-fixing and non-nitrogen-fixing cells with probes generated from various regions of the nif cluster by northern blot analysis revealed the presence of four different transcripts in nitrogen-fixing cells. Two of these transcripts had the predicted sizes spanning from nifH to nifK and from nifVw to nifVa, respectively. The other two transcripts did not have the expected sizes, but they suggested …
