Due to its ability to transport hydrophobic materials through the bloodstream, human serum albumin has become an intriguing component for drug delivery systems. The most pressing concerns with current albumin-based systems are the risk of pathogenic transfer associated with human-sourced blood proteins, the negative effect on osmotic pressure due to administration of a large dose of albumins, the potential immune response and loss of biological activity due to heavy modification of these albumins, and the need for organic solvents for preparation of these products. Alternative splicing occurs throughout the human genome, leading to multiple proteins from a single gene. The resulting proteins can be nearly identical or vastly different in how they function. Alternative splicing within the perilipin family has been observed in perilipins 1 and 3, giving rise to proteins with varying functions. In the course of our studies on perilipin 5, an immunoblot signal was observed that corresponded to an approximately 35 kDa protein. The alternansucrase (ALT, EC 2.4.1.140) catalyses transferring of glucose from sucrose to produce a polymer with α-1,6 and α- 1,3 linkages, so-called "alternan". The ALT-encoding gene from Leuconostoc citreum ABK-1 (WTalt) was successfully cloned and expressed in Escherichia coli BL21 (DE3). The two versions of truncated SH3-like domains, ∆3SHALT and ∆7SHALT, were successfully constructed. The WTALT, ∆3SHALT and ∆7SHALT possessed optimum temperature of 40 °C, while optimum pH of WTALT was at 5.0 but ∆3SHALT's and ∆7SHALT's were shifted to pH 4.0. Alpha-glucosidase [EC 3.2.1.20; α-D-glucoside glucohydrolase] is an exohydrolase which catalyzes non-reducing end of substrates to release D-glucose. In addition, alpha-glucosidase also displays a transferase activity, which bring about the formation of α-glucosylated compounds such as soluble starch and glycogen. This research aims to express and characterize α-glucosidase from Weissella confusa BBK-1 (WcAG). WcAG gene was expressed in modified pET28b vector. Fisetin is a flavonoid which was widely exerted in pharmacological industries. However, fisetin has low water solubility and unstable in aqueous solution. So the uses of fisetin have been limited. The purpose of this study was to increase water solubility of fisetin by transglycosylation reaction using cyclodextrin glycosyltransferase (CGTase) from Paenibacillus sp. RB01. The reaction had to perform in co-organic solvent system and DMSO at 40% (v/v) was proved to be the most suitable co-organic solvent since it provided the high enzyme stability and high production yield. The OsCaM1-1 is a rice calmodulin protein which transduces Ca2+ signals to their target protein that is important to regulate cellular and physiological responses. Lung neuroendocrine tumors (NETs) incidences are increasing in recent years. Women with lung NETs have significantly better survival rates as compared to men suggesting the involvement of sex steroids and their receptors in the progression of lung NETs. Recent data suggested that progesterone receptor (PR) may play a role in the survival of lung NET patients. PR exists as two major isoforms, PRA and PRB. ในปัจจุบัน ออทิซึมสเปกตรัม (Autism spectrum disorder, ASD) ยังคงเป็นโรคที่ยังคงไม่ทราบกลไกระดับโมเลกุล แน่ชัด ยีนและโปรตีนที่เคยมีการรายงานการแสดงออกผิดปกติไปในผู้ป่วยออทิซึมสเปกตรัมมีความสามารถในการทำซ้ำ (Reproducibility) ต่ำหรืออาจไม่มีเลย ซึ่งน่าจะมาจากความหลากหลายของอาการแสดงของผู้ป่วย จากการศึกษาล่าสุด แสดงให้เห็นว่าการจัดกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกสามารถช่วยในการระบุยีนและกลไกที่ผิดปกติไปในกลุ่มย่อยของผู้ป่วย อย่างไรก็ตาม การแบ่งกลุ่มผู้ป่วยเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกใน ออทิซึมสเปกตรัมยังไม่มีการใช้กันอย่างแพร่หลายในการค้นหาโปรตีน Biomarker ที่เกี่ยวข้องกับออทิซึมสเปกตรัม ในการศึกษาครั้งนี้ ผู้วิจัยทำการวิเคราะห์โดยการแบ่งกลุ่มผู้ป่วยจำนวน 85 รายออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R และทำการวิเคราะห์รูปแบบการแสดงออกของยีนในออทิซึมสเปกตรัมในแต่ละกลุ่มย่อยเปรียบเทียบกับกลุ่มควบคุมคนปกติที่อายุใกล้เคียงกันและเพศเดียวกันกับผู้ป่วย (Sex/age-matched control) ซึ่งสามารถระบุยีนที่มีการแสดงออกผิดปกติ (Differentially expressed genes, DEGs) ในออทิซึมสเปกตรัม นอกจากนี้ ผู้วิจัยได้ทำการวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมจากเซลล์ไลน์เม็ดเลือดขาว (Lymphoblastoid cell lines,LCLs) ด้วยเทคนิค Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) ร่วมกับ Nano-liquid chromatography-tandem mass spectrometry (Nano-LC-MS/MS) รายชื่อของโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญที่ได้จากการวิเคราะห์ถูกนำมาเปรียบเทียบกับ DEGs และผลการวิเคราะห์ Meta-analysis ของโปรตีนและโปรตีโอมในการศึกษาก่อนหน้า หน้าที่ทางพยาธิสภาพที่มีความเกี่ยวข้องกับโปรตีนถูกวิเคราะห์โดยโปรแกรม Ingenuity Pathway Analysis (IPA) และได้ทำการคัดเลือกโปรตีนที่น่าสนใจมาทำการตรวจสอบโดยใช้ Western blot analysis จากการศึกษาครั้งนี้พบว่าผลการวิเคราะห์การแบ่งกลุ่มผู้ป่วยออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R สามารถแบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อย ซึ่งพบว่าหนึ่งในกลุ่มนั้น เป็นกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง จากผลวิเคราะห์รูปแบบ ทรานสคริปโตมของเซลล์ไลน์เม็ดเลือดขาว พบว่ามีจุดโปรตีนจำนวน 82 จุดที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง และได้ทำการคัดเลือก 18 จุดโปรตีนจากทั้งหมดมาวิเคราะห์ชนิดของโปรตีนโดยเทคนิค Nano-LC-MS/MS ซึ่งพบว่ามีความเกี่ยวข้องกับการทำงานของระบบประสาทและกระบวนการอักเสบ จากการศึกษานี้พบว่าจากโปรตีนทั้งหมด การแสดงออกของ Isocitrate dehydrogenase (IDH2) และ Diazepam binding inhibitor (DBI) ลดลงในออทิซึมสเปกตรัม ซึ่งโปรตีนเหล่านี้ยังพบการแสดงออกผิดปกติในการวิเคราะห์ Meta-analysis ของทรานสคริปโตมและโปรตีโอมร่วมด้วย การแบ่งกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยทางคลินิกตามด้วยการวิเคราะห์ทรานสคริปโตมและโปรตีโอมแบบบูรณาการร่วมกัน ทำให้การศึกษานี้สามารถระบุโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มย่อยของออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษาอย่างรุนแรง ซึ่งโปรตีนเหล่านี้อาจทำหน้าที่เป็นตัวแทนในการช่วยระบุกลไกทางโมเลกุลของออทิซึมสเปกตรัมในอนาคต ตลอดจน อาจพัฒนาไปเป็น …

กระบวนการสร้างนิวไรท์ เป็นกระบวนการที่สำคัญในการเจริญพัฒนา การซ่อมแซมส่วนที่เสียหาย รวมถึงการสื่อสารระหว่างเซลล์ของเซลล์ประสาท ซึ่งหากกระบวนการการสร้างนิวไรท์เกิดความผิดปกติสามารถก่อให้เกิดโรคความผิดปกติของระบบประสาท เช่น โรคอัลไซเมอร์ โรคพาร์กินสัน และโรคฮันติงตัน เป็นต้น ในการศึกษาวิจัยนี้ศึกษาฤทธิ์การต้านสารอนุมูลอิสระ และการกระตุ้นการสร้างนิวไรท์จากสารสกัดเมล็ดของหมามุ่ยไทย และหมามุ่ยอินเดียในเซลล์ประสาทเพาะเลี้ยงชนิด Neuro2a การศึกษาคุณสมบัติในการต้านสารอนุมูลอิสระโดยวิธี DPPH และ ABTS assays นอกจากนั้นทำการศึกษาอัตราการรอดชีวิตของเซลล์ประสาทโดยวิธี MTT พร้อมกับศึกษาการแสดงออกของยีนที่เกี่ยวข้องกับการกระตุ้นการงอกของนิวไรท์ ประกอบด้วย Ten-4 และ GAP-43 โดยวิธี Real-time PCR ซึ่งจากการทดสอบพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียด้วยตัวทำละลายเอทานอลมีความสามารถในการต้านสารอนุมูลอิสระ และพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียไม่เป็นพิษต่อเซลล์ประสาท จากนั้นทำการศึกษาฤทธิ์ และกลไกของสารสกัดจากสมุนไพรต่อการกระตุ้นการงอกของนิวไรท์ โดยพบว่าสารสกัดจากเมล็ดหมามุ่ยไทยด้วยตัวทำละลายเอทานอล สามารถกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 และ GAP-43 ในขณะที่สารสกัดจากเมล็ดของหมามุ่ยอินเดียด้วยตัวทำละลายเฮกเซนกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 อีกทั้งศึกษาผลการยับยั้งการแสดงออกของยีน Ten-4 ต่อการกระตุ้นการสร้างนิวไรท์ของสารสกัดสมุนไพร ซึ่งพบว่ามีการสร้างนิวไรท์ และความยาวของนิวไรท์ลดลง จากการศึกษาสามารถบ่งชี้ได้ว่าสารสกัดจากเมล็ดหมามุ่ยไทย และหมามุ่ยอินเดียสามารถปกป้องเซลล์ประสาทจากสารอนุมูลอิสระได้ และสามารถกระตุ้นการงอกของนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน GAP-43 และ Ten-4 ซึ่งเป็นอีกทางเลือกหนึ่งในการพัฒนายาการกระตุ้นการเจริญพัฒนาของเซลล์ประสาทสำหรับป้องกัน หรือรักษาโรคความผิดปกติของระบบประสาท

โรคออทิซึมสเปกตรัม (Autism Spectrum Disorder, ASD) คือ โรคทางพัฒนาการระบบประสาทที่พบในเพศชายมากกว่าเพศหญิงโดยไม่ทราบสาเหตุ สาเหตุการเกิดโรคออทิซึมสเปกตรัมยังไม่เป็นที่ทราบแน่ชัดแต่การได้รับบิสฟีนอลเอขณะตั้งท้องมีความสัมพันธ์ต่อการเกิดโรคออทิซึมสเปกตรัมในลูกหนู อย่างไรก็ตามยังไม่มีการศึกษาแน่ชัดว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกของยีนที่เกี่ยวข้องกับโรคออทิซึมสเปกตรัมหรือไม่ โดยเฉพาะเมื่อวิเคราะห์แบบแยกเพศ ในการศึกษาครั้งนี้ ทางคณะผู้วิจัยจึงทำการศึกษาผลของการได้รับบิสฟีนอลเอในขณะตั้งท้องต่อการแสดงออกของยีนในสมองและความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมรวมไปถึงหน้าที่ของเซลล์ระบบประสาท ในการทำนายว่าการได้รับบิสฟีนอลเอสามารถเปลี่ยนแปลงการแสดงออกของยีนและมีความสัมพันธ์กับโรคออทิซึมสเปกตรัมหรือไม่ ทางผู้วิจัยได้ทำการวิเคราะห์แบบ meta-analysis โดยใช้ข้อมูลการแสดงออกของยีนที่ผิดปกติจากการได้รับบิสฟีนอลเอที่ได้ตีพิมพ์ไปแล้วและใช้การวิเคราะห์แบบ hypergeometric distribution หาความสัมพันธ์ระหว่างกลุ่มยีนดังกล่าวกับรายชื่อยีนจากฐานข้อมูลโรคออทิซึมสเปกตรัมต่าง ๆ สำหรับการศึกษาถึงผลของการได้รับบิสฟีนอลเอขณะตั้งท้องต่อการแสดงออกของยีนที่มีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมนั้น คณะผู้วิจัยได้นำสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลขณะตั้งท้องมาวิเคราะห์ทางทรานสคริปโตมิกส์ด้วยเทคนิค RNA-seq และใช้โปรแกรม Ingenuity Pathway Analysis วิเคราะห์อินเตอร์แอกโตมและบทบาทหน้าที่ทางชีวภาพของกลุ่มยีนที่มีการแสดงออกผิดปกติ ยีนที่มีความสัมพันธ์กับโรคออทิซึมสเปกตรัมถูกคัดเลือกเพื่อยืนยันด้วยเทคนิค quantitative RT-PCR นอกจากนี้เซลล์ระบบประสาทที่แยกได้จากลูกหนูถูกนำมาเพาะเลี้ยงเพื่อศึกษาความยาวของสายใยประสาท การศึกษาพบว่า จากการวิเคราะห์ทาง meta-analysis ยีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความสัมพันธ์อย่างมีนัยสำคัญกับหน้าที่และโรคออทิซึมสเปกตรัม การวิเคราะห์ RNA-seq ในสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลเอขณะตั้งท้องพบว่ายีนที่มีการแสดงออกผิดปกติมีความสัมพันธ์กับโรคออทิซึมสเปกตรัม เป็นที่น่าสนใจอย่างยิ่งว่ากลุ่มยีนที่เคยมีการรายงานว่ามีความผิดปกติในโรคออทิซึมสเปกตรัม ได้แก่ Auts2 Foxp2 และ Smarcc2 แสดงออกผิดปกติอย่างจำเพาะต่อเพศ การวิเคราะห์เชิงอินเตอร์แอกโตมพบว่ายีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความเกี่ยวข้องอย่างมีนัยสำคัญกับบทบาทหน้าที่ทางระบบประสาทที่เคยมีรายงานในผู้ป่วยออทิซึมสเปกตรัม การวิเคราะห์ความยาวของสายใยประสาทพบว่าการได้รับบิสฟีนอลเอส่งเสริมความยาวของสายใยประสาทและการแตกแขนงของสายใยประสาทแต่ลดขนาดของตัวเซลล์อย่างมีนัยสำคัญทางสถิติ จากการศึกษาครั้งนี้พบว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกที่ผิดปกติของยีนในสมองส่วนฮิปโปแคมปัสและกลุ่มยีนดังกล่าวมีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมและแสดงออกจำเพาะในแต่ละเพศประกอบกับการได้รับบิสฟีนอลเอขณะตั้งท้องส่งเสริมความยาวของสายใยประสาท การศึกษานี้แสดงให้เห็นว่าการได้รับบิสฟีนอลเอขณะตั้งครรภ์อาจเป็นสาเหตุหรือเพิ่มความเสี่ยงต่อโรคออทิซึมสเปกตรัมและอาจจะเป็นหลักฐานที่อธิบายถึงความชุกของโรคนี้ในเพศชายที่มากกว่าเพศหญิงซึ่งควรศึกษาเพิ่มเติมในอนาคต

Metaproteomics is the characterization of all proteins being expressed by a community of organisms in a complex biological sample at a single point in time. Applications of metaproteomics range from the comparative analysis of environmental samples (such as ocean water and soil) to microbiome data from multicellular organisms (such as the human gut). Classification method performance was evaluated using otolith chemistry of juvenile Atlantic menhaden Brevoortia tyrannus when assumptions of data normality were met and were violated. Four methods were tested [linear discriminant function analysis (LDFA), quadratic discriminant function analysis (QDFA), random forest (RF) and artificial neural networks (ANN)] using computer simulation to determine their performance when variable-group means ranged from small to large and their performance under conditions of typical skewness to double the amount of skewness typically observed. Using the kappa index, the parametric methods performed best after applying appropriate data transformation, gaining 2% better performance with LDFA performing slightly better …

Proteins that bind to both DNA and RNA embody the ability to perform multiple functions by a single gene product. These nucleic acid binding proteins in prokaryotes can play a vital role in many cellular processes, including replication, transcription, gene expression, recombination, and repair, to name a few. Nucleic acid binding proteins have unique functional characteristics that stem from their structural attributes that have evolved in a widely-conserved manner. SH3 and multiple ankyrin repeat domains 3 (SHANK3) is a multidomain scaffold protein that is highly augmented in the postsynaptic density (PSD) of excitatory glutamatergic synapses within the central and peripheral nervous systems. SHANK3 links neurotransmitter receptors, ion channels, and other critical membrane proteins to intracellular cytoskeleton and signal transduction pathways. Mutations in SHANK3 are linked with a number neuropsychiatric disorders including autism spectrum disorders (ASDs). Intellectual disability, impaired memory and learning, and epilepsy are some of the deficits commonly associated with ASDs that result from mutations in SHANK3. Niemann Pick Type C (NPC) disease is a fatal childhood neurological disease caused by mutations in the NPC-1 protein, resulting in cholesterol buildup in the late endosomes. StarD4 and StarD5 are cholesterol binding proteins that play a role in the intracellular cholesterol transport. In this study we overexpress StarD4 and StarD5 in in vitro and in vivo models, and find evidence of amelioration of the NPC phenotype. This study demonstrates that the overexpression of these proteins has the potential to be a therapeutic treatment for NPC disease.

The objective of this project was to identify important sources of bacterial loading in Plaster Creek (Kent County). The following goals were set to reach this objective: 1) the relative contributions of bacterial loading for 10 major tributaries will be assessed and the five most egregious contributors will be prioritized; 2) molecular markers will be used to try to identify the primary taxonomic sources, and 3) to pinpoint bacterial loading locations using molecular markers and scent-trained canines in sub-watersheds.

A key idea in de novo modeling of a medium-resolution density image obtained from cryo-electron microscopy is to compute the optimal mapping between the secondary structure traces observed in the density image and those predicted on the protein sequence. When secondary structures are not determined precisely, either from the image or from the amino acid sequence of the protein, the computational problem becomes more complex. We present an efficient method that addresses the secondary structure placement problem in presence of multiple secondary structure predictions and computes the optimal mapping. Three-dimensional density maps of biological specimens from cryo-electron microscopy (cryo-EM) can be interpreted in the form of atomic models that are modeled into the density, or they can be compared to known atomic structures. When the central axis of a helix is detectable in a cryo-EM density map, it is possible to quantify the agreement between this central axis and a central axis calculated from the atomic model or structure. We propose a novel arc-length association method to compare the two axes reliably. This method was applied to 79 helices in simulated density maps and six case studies using cryo-EM …
