Altered Gene Response To Aflatoxin B1 In The Spleens Of Susceptible And Resistant Turkeys, Kent M. Reed, Kristelle M. Mendoza, Roger A. Coulombe Jr.

Animal, Dairy, and Veterinary Science Faculty Publications

Susceptibility and/or resistance to aflatoxin B1 (AFB1) is a threshold trait governed principally by glutathione S transferase (GST)-mediated detoxification. In poultry, domesticated turkeys are highly sensitive to AFB1, most likely due to dysfunction in hepatic GSTs. In contrast, wild turkeys are comparatively resistant to aflatoxicosis due to the presence of functional hepatic GSTAs and other possible physiological and immunological interactions. The underlying genetic basis for the disparate GST function in turkeys is unknown as are the broader molecular interactions that control the systemic response. This study quantifies the effects of dietary AFB1 on gene …

Currently, there are no licensed human vaccines or antivirals for treatment of or prevention from infection with encephalitic alphaviruses. Because epidemics are sporadic and unpredictable, and endemic disease is common but rarely diagnosed, it is difficult to identify all populations requiring vaccination; thus, an effective post-exposure treatment method is needed to interrupt ongoing outbreaks. To address this public health need, we have continued development of ML336 to deliver a molecule with prophylactic and therapeutic potential that could be relevant for use in natural epidemics or deliberate release scenario for Venezuelan equine encephalitis virus (VEEV). We report findings from in vitro …

Quantification Of Tylosin Antibiotics In Cattle Waste, Appala Keerthi Apr 2019

Quantification Of Tylosin Antibiotics In Cattle Waste, Appala Keerthi

Masters Theses & Specialist Projects

Antibiotics are used as prophylactic agents to promote growth and for treating infections in animals. However, the irrational use of antibiotics in livestock management is a significant cause of the development of antibioticresistant genes in the environment. Each year 2 million people suffer from the infections caused by bacteria which are resistant to antibiotics and 23,000 of these people are estimated to die because of antibiotic resistance. New drugs are continually coming into the market but are at the risk of developing resistance. Thus, there is a need for the development of analytical methods which can be used to monitor …

Islands In A Sea Of Aaruul: Globalization And Mongolian Cheesemakers, Pearse Anderson

Independent Study Project (ISP) Collection

Despite being a sparsely-populated country with less than 2% of its land arable, Mongolia has developed a rich and complex food culture, notably for its meat and dairy products, which could soon be at an historic high thanks to the tens of millions of animals on Mongolian pastureland. Many Mongolians and non-Mongolians view the countryside as a sea of milk that is currently being underutilized for economic exploitation. Various projects, whether funded and organized by international NGOs, the Mongolian government, or private companies, have tried to use Mongolian’s dairy resources to fill Mongolian demand, with more recent private ventures also …

Crystalline silica (cSiO2) is a widely recognized environmental trigger of autoimmune disease. In the lupus-prone female NZBWF1 mouse, airway exposure to cSiO2 triggers pulmonary ectopic lymphoid neogenesis, systemic autoantibody elevation, and glomerulonephritis. Here we tested the hypothesis that upregulation of adaptive immune function genes in the lung precedes cSiO2-triggering of autoimmune disease in this model. The study include three groups of mice, as follows: (1) necropsied 1 d after a single intranasal instillation of 1 mg cSiO2 or vehicle, (2) necropsied 1 d after four weekly single instillations of 1 mg cSiO2 or …

To address the unmet needs for human polyclonal antibodies both as therapeutics and diagnostic reagents, building upon our previously established transchromosomic (Tc) cattle platform, we report herein the development of a Tc goat system expressing human polyclonal antibodies in their sera. In the Tc goat system, a human artificial chromosome (HAC) comprising the entire human immunoglobulin (Ig) gene repertoire in the germline configuration was introduced into the genetic makeup of the domestic goat. We achieved this by transferring the HAC into goat fetal fibroblast cells followed by somatic cell nuclear transfer for Tc goat production. Gene and protein expression analyses …

Zika virus (ZIKV) infection can cause severe congenital diseases, such as microcephaly, ocular defects and arthrogryposis in fetuses, and Guillain–Barré syndrome in adults. Efficacious therapeutic treatments for infected patients, as well as prophylactic treatments to prevent new infections are needed for combating ZIKV infection. Here, we report that ZIKV-specific human polyclonal antibodies (SAB-155), elicited in transchromosomal bovine (TcB), provide significant protection from infection by ZIKV in STAT2 knockout (KO) golden Syrian hamsters both prophylactically and therapeutically. These antibodies also prevent testicular lesions in this hamster model. Our data indicate that antibody-mediated immunotherapy is effective in treating ZIKV infection. Because suitable …

Differential Transcriptome Responses To Aflatoxin B1 In The Cecal Tonsil Of Susceptible And Resistant Turkeys, Kent M. Reed, Kristelle M. Mendoza, Roger A. Coulombe Jr.

Animal, Dairy, and Veterinary Science Faculty Publications

The nearly-ubiquitous food and feed-borne mycotoxin aflatoxin B1 (AFB1) is carcinogenic and mutagenic, posing a food safety threat to humans and animals. One of the most susceptible animal species known and thus a good model for characterizing toxicological pathways, is the domesticated turkey (DT), a condition likely due, at least in part, to deficient hepatic AFB1-detoxifying alpha-class glutathione S-transferases (GSTAs). Conversely, wild turkeys (Eastern wild, EW) are relatively resistant to the hepatotoxic, hepatocarcinogenic and immunosuppressive effects of AFB1 owing to functional gene expression and presence of functional hepatic GSTAs. This study was designed to …

Perceptions Of On-Farm Emergency Slaughter For Dairy Cows In British Columbia, Katherine E. Koralesky, David Fraser

Slaughter and Slaughtering Practices Collection

Some jurisdictions permit on-farm emergency slaughter (OFES) as one end-of-life option for dairy cows and other animals that cannot be transported humanely but are deemed fit for human consumption. Anecdotal reports suggest that OFES is controversial among dairy industry professionals, but to date their perceptions of OFES have not been studied systematically. Twentyfive individual interviews and 3 focus groups with 40 dairy producers, veterinarians, and other professionals in British Columbia, Canada, revealed positive and negative perceptions of OFES influenced by (1) individual values, (2) the perceived operational legitimacy of OFES, and (3) concern over social responsibility and public perception of …

Using An Implantable Microchip For Measuring Body Temperature In Dairy Calves, Megan Woodrum, Melissa Cantor, Joao H. C. Costa

Posters-at-the-Capitol Presentations

Body temperature is frequently used as a method for determining if illness is present with fever detection. Taking rectal temperature or alternatively tympanic temperature can be time consuming and requires restraint of calves. Alternatives, such as implantable microchips that can be passively read using a radio frequency identification (RFID) scanner may allow for easier monitoring of body temperature.

This study aim was to validate an implantable microchip and to determine the best implant site.

Hydrodynamic Cavitation As In-Line Process To Control Common Dairy Sporeformers, Pratibha Chaudhary Jan 2019

Hydrodynamic Cavitation As In-Line Process To Control Common Dairy Sporeformers, Pratibha Chaudhary

Electronic Theses and Dissertations

Sporeforming bacteria and their endospores have the ability to survive thermal treatments and proliferate in the milk and dairy products to cause spoilages. We applied the effect of hydrodynamic cavitation in controlling these bacteria and their spores at pilot scale level. Our aim was to investigate a technology that would improve the microbial quality of milk and dairy products, and is capable of being scaled up to be adapted at industrial level. Cavitation damages the cell walls of the bacteria and their endospores (releasing dipicolinic acid, responsible for endospore resistance to extreme conditions), thereby making them susceptible to thermal treatments …

Evaluation Of Effects Of Methods Used For Forage Analysis And Dietary Inclusion Of Buffer On Fiber Digestibility And Rumen Fermentation In Diets High In Distillers Grains, Lydia Kehinde Olagunju

Electronic Theses and Dissertations

Forage quality is an important factor affecting intake and utilization of forage, thus making it imperative to evaluate the methods used to determine the nutritive values of forage in order to predict animal performance. Additionally, fibrous feeds for ruminants are less subject to competitive demand. The plant cell wall is the largest hindrance to complete digestion of feeds, particularly forages and by-products and to the utilization of the nutrients and energy they contain, necessitating effective strategies for increasing the rate and efficiency of utilization of forage fiber and the energy therein. It is critically important to increase fiber digestion for …

Assessing Listeria Risk During Different Stages Of Ice Cream Manufacturing And Storage, Neha Singh Jan 2019

Assessing Listeria Risk During Different Stages Of Ice Cream Manufacturing And Storage, Neha Singh

Electronic Theses and Dissertations

Listeriosis is a life-threatening infection caused by foods contaminated with Listeria monocytogenes. Some of the major ice cream recalls in recent years reaffirm the ability of this food-borne pathogen to survive in diverse dairy processing environments and cause cross contamination. Inspection reports revealed certain lapses in implementing adequate hygienic practices for Listeria persistence in the processing environment, leading to cross contamination of ice cream. The higher levels of cross contamination of raw ice cream mix might result in random heat-injured cells when exposed to minimum heat treatment (69°C for 30 min). Evidence about the presence of injured cells in ice …

Effects Of Yeast-Derived Microbial Protein On Transition Dairy Cow Health And Performance, Gustavo Mazon Correa Alves Jan 2019

Effects Of Yeast-Derived Microbial Protein On Transition Dairy Cow Health And Performance, Gustavo Mazon Correa Alves

Theses and Dissertations--Animal and Food Sciences

The transition period for dairy cows is defined as the three weeks pre and postpartum. During the transition period, dairy cows experience a myriad of metabolic, managerial, and nutritional requirement changes. These changes lead to stress and increased susceptibility to diseases which can negatively affect lactational performance in the short and long term. However, dietary amino acid availability can have a dramatic impact on the health and performance of dairy cows around parturition. Thus, the objective of the thesis was to evaluate the effects of supplementing yeast-derived microbial protein, as an alternative protein source for dairy cows during the transition …

Automated Body Condition Scoring: Progression Across Lactation And Its Association With Disease And Reproduction In Dairy Cattle, Carissa Marie Truman

Theses and Dissertations--Animal and Food Sciences

Body condition scoring is a technique used to noninvasively assess fat reserves. It provides an objective estimate to describe the current and past nutritional status of the dairy cow and has been associated with increased disease risk and breeding success. Traditionally body condition scores are taken manually by visual appraisal on a 1 to 5 scale, in one-quarter increments. However, recent studies have shown the potential of automating the body condition scoring of cows using images. The first objective was to estimate the likelihood of disease development and breeding success, using odds ratios, associated with body condition score scored automatically …
