Bovine mastitis, or inflammation of the mammary gland, has substantial economic and animal welfare implications. A genetic basis for mastitis resistance traits is recognized and can be used to guide selective breeding programs. The discovery of regions of the genome associated with mastitis resistance, and knowledge of the underlying molecular mechanisms responsible, can facilitate development of efficient mastitis control and therapeutic strategies. The objectives of this dissertation research were to identify sites of genetic variation associated with mastitis resistance, and to define the contributions of the milk-secreting epithelial cells to mammary gland immune responses and mastitis resistance. Zika virus (ZIKV) infection of pregnant women is associated with pathologic complications of fetal development. Here, we infect pregnant rhesus macaques (Macaca mulatta) with a minimally passaged ZIKV isolate from Rio de Janeiro, where a high rate of fetal development complications was observed. The infection of pregnant macaques with this virus results in maternal viremia, virus crossing into the amniotic fluid (AF), and in utero fetal deaths. We also treated three additional ZIKV-infected pregnant macaques with a cocktail of ZIKV-neutralizing human monoclonal antibodies (nmAbs) at peak viremia. Background
In food animal agriculture, there is a need to identify the mechanisms that can improve the efficiency of muscle growth and protein accretion. Callipyge sheep provide excellent machinery since the up-regulation of DLK1 and RTL1 results in extreme postnatal muscle hypertrophy in distinct muscles. The aim of this study is to distinguish the genes that directly respond to DLK1 and RTL1 signaling from the genes that change as the result of muscle specific effects.
Results
The quantitative PCR results indicated that DLK1 expression was significantly increased in hypertrophied muscles but not in non-hypertrophied muscles. However, RTL1 was up-regulated in …

Syringomyelia is a form of myelodysplasia defined by the formation of one or more fluid-filled cavities within the spinal cord that do not communicate with the central canal. The defect may be congenital or acquired. Clinical signs correlate to the segment of spinal cord affected and include pain, paresis, proprioceptive deficits, alterations in sensation, scoliosis, and autonomic dysfunction. This report describes the clinical and pathologic changes in a case of acquired syringomyelia in a 10-year-old American Paint Horse mare. The horse had a six-week history of progressive proprioceptive deficits in all four limbs, bilateral pelvic limb ataxia, and muscle fasciculations …

Yogurt has been recognized as a healthy food. Nutritional value of yogurt can be increased with the addition of aloe vera juice (AVJ) and inulin. People are interested in consuming products without artificial ingredients and with high content of bioactive compounds. The aims of this study were to develop a functional plain yogurt enhanced with inulin and AVJ, to evaluate the physicochemical properties, the enumeration of Streptococcus thermophilus and Lactobacillus bulgaricus, inulin content, and consumer acceptability. Epithelial cells lining secretory units and ducts of bovine mammary glands perform an important role in regulating movement of various macromolecules and whole cells during normal lactation and mastitis. During mastitis, host and bacterial produced substances can affect the “barrier” function of epithelial monolayers. One potential component is adenosine triphosphate (ATP). ATP likely interacts with P2X7, a purinergic receptor, in mediating some effects associated with mastitis. Bovine mammary gland epithelial cell line, Mac-T cells, were examined for cytoskeletal changes as result of P2X7 interactions. Zona pellucida hardening is a natural process that occurs after oocyte fertilization to prevent polyspermic fertilization and to protect embryonic development. Pre-fertilization hardening of the zona pellucida however, decreases fertilization rates. Cryoprotectants have also been shown to negatively affect fertilization rates, one possible mechanism of which being through zona hardening. This experiment was conducted to determine the effect of different cryoprotectants on hardening of the zona pellucida using fresh mature bovine oocytes and vitrified mature bovine oocytes. Domestication fundamentally reshaped animal morphology, physiology and behaviour, offering the opportunity to investigate the molecular processes driving evolutionary change. Here we assess sheep domestication and artificial selection by comparing genome sequence from 43 modern breeds (Ovis aries) and their Asian mouflon ancestor (O. orientalis) to identify selection sweeps. Next, we provide a comparative functional annotation of the sheep genome, validated using experimental ChIP-Seq of sheep tissue. Using these annotations, we evaluate the impact of selection and domestication on regulatory sequences and find that sweeps are significantly enriched for protein coding genes, proximal regulatory elements of genes …

Zika virus (ZIKV) has received widespread attention because of its effect on the developing fetus. It is becoming apparent, however, that severe neurological sequelae, such as Guillian-Barrë syndrome (GBS), myelitis, encephalitis, and seizures can occur after infection of adults. This study demonstrates that a contemporary strain of ZIKV can widely infect astrocytes and neurons in the brain and spinal cord of adult, interferon α/β receptor knockout mice (AG129 strain) and cause progressive hindlimb paralysis, as well as severe seizure-like activity during the acute phase of disease. The objective of this study was to identify the major electricity and water-consuming components of a pasture-based automatic milking (AM) system and to establish the daily and seasonal consumption trends. Electricity and water meters were installed on 7 seasonal calving pasture-based AM farms across Ireland. Electricity-consuming processes and equipment that were metered for consumption included milk cooling components, air compressors, AM unit(s), auxiliary water heaters, water pumps, lights, sockets, automatic manure scrapers, and so on. The food-borne mycotoxin aflatoxin B1 (AFB1) poses a significant risk to poultry, which are highly susceptible to its hepatotoxic effects. Domesticated turkeys (Meleagris gallopavo) are especially sensitive, whereas wild turkeys (M.g. silvestris) are more resistant. AFB1 toxicity entails bioactivation by hepatic cytochrome P450s to the electrophilic exo-AFB1-8,9-epoxide (AFBO). Domesticated turkeys lack functional hepatic GST-mediated detoxification of AFBO, and this is largely responsible for the differences in resistance between turkey types. Enterovirus D68 (EV-D68) caused a large outbreak in the summer and fall of 2014 in the United States. It causes serious respiratory disease, but causation of associated paralysis is controversial, because the virus is not routinely identified in cerebrospinal fluid. To establish clinical correlates with human disease, we evaluated EV-D68 infection in non-lethal paralysis mouse models. Ten-day-old mice lacking interferon responses were injected intraperitoneally with the virus. Paralysis developed in hindlimbs. After six weeks of paralysis, the motor neurons were depleted due to viral infection. Hindlimb muscles were also infected and degenerating. Even at the earliest stage of paralysis, muscles …

Despite the increasing frequency of integrated automatic milking (AM) and pasture-based systems, there is limited knowledge available on the suitability of different dairy cow breeds to these systems. Thus, the objective of this experiment was to establish the performance of three breeds in a pasture-based AM system with respect to milk production, cow traffic and milking characteristics. The breeds examined were Holstein Friesian (HF), Jersey x HF (JEX) and Norwegian Red x HF (NRX), all of which have been previously identified as being compatible with conventional milking pasture-based systems. The pronuclear (PN) injection technique was first established in mice to introduce foreign genetic materials into the pronuclei of one-cell stage embryos. The introduced genetic material may integrate into the embryonic genome and generate transgenic animals with foreign genetic information following transfer of the injected embryos to foster mothers. Following the success in mice, PN injection has been applied successfully in many other animal species. Detection of coding/functional SNPs that change the biological function of a gene may lead to identification of putative causative alleles within QTL regions and discovery of genetic markers with large effects on phenotypes. This study has two-fold objectives, first to develop, and validate a 50K transcribed gene SNP-chip using RNA-Seq data. To achieve this objective, two bioinformatics pipelines, GATK and SAMtools, were used to identify ∼21K transcribed SNPs with allelic imbalances associated with important aquaculture production traits including body weight, muscle yield, muscle fat content, shear force, and whiteness in addition to resistance/susceptibility to bacterial cold-water disease (BCWD). With the abolishment of milking quotas across all European Union member states in April 2015, dairy farmers must adjust their farming practises to minimise milk production costs to adequately prepare for potential periods of reduced revenue. Milk production is an intense energy and water consuming process. Six experiments were conducted for this dissertation research to evaluate strategies to improve growth performance and health of calves. Growth performance, nutrient utilization, and health effects were evaluated when calves were offered water treated with reverse osmosis system (RW), fed starter pellets supplemented with microbially-enhanced soy protein (MSP) or supplemented condensed whey solubles (CWS). The first two experiments were conducted to determine drinking preference, growth performance, and health of dairy calves offered RW or by a municipal water treatment plant (MW), compared with local untreated well water (WW). Phytochemicals such as tannins included in dairy cow nutritional programs, as a natural feed additive, promise to improve rumen bypass protein and consequently reduce the feeding cost by decreasing the amount of CP needed in the ration. Additionally, if tannins are fed along with high quality protein sources, this could improve the AA profile reaching the small intestine and potentially improving health and performance of dairy cows. The black pepper, specifically its alkaloid piperine, is a nontoxic, natural dietary compound with a broad range of physiological activity. The main purpose of this body of research was to evaluate carinata meal as a feedstuff for dairy heifers and lay the foundations for its future evaluation in lactating dairy cow diets. This research focused on evaluating the taste preference of carinata meal compared with other oilseeds meals and distillers dried grains with solubles (DDGS). Despite containing glucosinolates, carinata meal was preferred similarly to canola meal, had greater preference compared to camelina meal, and less preference compared to the other meals. The objective of this study was to examine the effects of feeding exogenous amylolytic and proteolytic enzymes on lactation performance of dairy cows fed freshly ensiled corn silage-based diets. We hypothesized that the addition of these enzymes would improve nutrient utilization and consequently lactation performance. Thirty-six Holstein cows [15 multiparous and 15 primiparous; DIM = 132 ± 48 and 6 cannulated (3 multiparous and 3 primiparous; DIM = 164 ± 50] were blocked by milk yield, DIM, parity, and body weights and used in a 9 wk randomized complete block design study.. Precision dairy monitoring is used to supplement or replace human observation of dairy cattle. This study examined the value dairy producers placed on disease alerts generated from a precision dairy monitoring technology. A secondary objective was calculating the accuracy of technology-generated disease alerts compared against observed disease events. A final objective was determining the economic viability of investing in a precision dairy monitoring technology for detecting estrus and diseases.

Understanding the total cost per case of disease-specific lameness can help producers select better treatment, prevention, and control strategies for their herds. The first objective of our research was to calculate the costs associated with 3 lameness causing hoof diseases: digital dermatitis, sole ulcer, and white line disease. Mastitis and milk quality affect every dairy farmer across the globe. Sand bedded freestalls are the industry standard for cow comfort, welfare, and the control of environmental mastitis. Compost bedded packs may be a viable alternative to the sand bedded freestall.
Compost bedded packs are maintained at a consistent level of moisture, nutrients, and aeration to favor compost microorganisms. The emergence of antibiotic resistances has raised concerns in society about the widespread use of antibiotics, such as monensin, as growth enhancers in agriculture. Pressure to find alternatives has increased since the European Union banned the use of ionophorous antibiotics. Butyrate supplementation has been found to enhance growth in pre-weaned calves and it has been recently suggested to enhance growth in post-weaned heifers. In a recent study by Rice in 2017, there was a quadratic (P=0.03) response for coccidia counts as sodium butyrate was increased, with the lowest counts being at the intermediate doses.
