Department of Animal Science: Faculty Publications
We characterized 24 polymorphic tetranucleotide microsatellite loci for delta smelt (Hypomesus transpacificus) endemic to the San Francisco Bay Estuary, California, USA. Screening of samples (n = 30) yielded two to 26 alleles per locus with observed levels of heterozygosity ranging from 0.17 to 1.0. Only one locus deviated from Hardy–Weinberg equilibrium, suggesting these individuals originate from a single panmictic population. Linkage disequilibrium was found in two pairs of loci after excluding the locus out of Hardy–Weinberg equilibrium. Twenty-two primer pairs cross-amplified in wakasagi smelt (Hypomesus nipponensis), and 15 primer pairs cross-amplified in longfin smelt ( …

Assessment Of River Herring And Striped Bass In The Connecticut River: Abundance, Population Structure, And Predator/Prey Interactions, Justin P. Davis, Eric T. Schultz, Jason Vokoun
EEB Articles
Populations of anadromous alewife Alosa pseudoharengus and blueback herring A. aestivalis, collectively referred to as river herring, have declined in the Connecticut River. An explanatory hypothesis for these declines is that predation pressures have increased as a result of recent increases in abundance of sympatric striped bass Morone saxatilis. We sampled river herring and striped bass from the stretch of the Connecticut River between Wethersfield, CT and Holyoke, MA during the vernal migration seasons of 2005-2008. The objectives of the sampling program were to assess abundance, temporal/spatial distribution, and population structure of both river herring and striped bass, …

Estimating Predation On Declining River Herring: Tag-Recapture Study Of Striped Bass In The Connecticut River, Eric T. Schultz, Justin P. Davis, Jason Vokoun
EEB Articles
Populations of anadromous alewife Alosa pseudoharengus and blueback herring Alosa aestivalis, collectively referred to as river herring, have declined in the Connecticut River. A hypothesis for why river herring have declined is that predation pressures have increased associated with recent increases in abundance of striped bass Morone saxatilis. Information on striped bass abundance, size structure, and consumption rates are required to test this hypothesis. This study was designed to provide estimates of striped bass population size in the Connecticut River during the spring migration season, via an intensive mark-recapture exercise and either an open or robust mark-recapture model. …

Cutler Reservoir is located in Cache county, Utah and was created for the purposes of irrigation, water storage and flood control. High nutrient loading to Cutler has raised concerns about the health of this system and has resulted in it being listed on the state's 303(d) list of impaired waters. The TMDL plan being drafted for Cutler lists dissolved oxygen and phosphorous as the key issues of concern. The underlying problem created by nutrient loading is eutrophication. If Cutler is to remain as a valuable source of recreation, wildlife habitat, and water for the Cache Valley we must understand the …

Thirty-Seven Additional Microsatellite Loci In The Pacific Lion-Paw Scallop (Nodipecten Subnodosus) And Cross-Amplification In Other Pectinids, Jessica Lynn Petersen, Ana M. Ibarra, Bernie May
Department of Animal Science: Faculty Publications
We characterized 37 new microsatellite markers in the Pacific lion-paw scallop (Nodipecten subnodosus) and tested for cross-amplification in four other species. Genetic diversity was estimated using 24 individuals from the Lagoon Ojo de Liebre, B.C.S., Mexico. Allelic richness varied from 5 to 27 alleles per locus and the average expected heterozygosity was 0.76. Ten loci exhibited significant departure from Hardy–Weinberg equilibrium likely due to the presence of null alleles. Sixteen of these markers cross-amplified in closely related N. nodosus, while little or no amplification was observed in three Argopecten species.
Evaluating The Effectiveness Of Grassbed Treatments As Habitat For Juvenile Black Bass In A Drawdown Reservoir, D. R. Ratcliff, Wayne A. Wurtsbaugh, J. Zustak
Evaluating The Effectiveness Of Grassbed Treatments As Habitat For Juvenile Black Bass In A Drawdown Reservoir, D. R. Ratcliff, Wayne A. Wurtsbaugh, J. Zustak
Watershed Sciences Faculty Publications
Many reservoirs in arid regions experience highly variable water levels caused by seasonal inflow fluctuations and designated outflow requirements. At Shasta Lake, California, managers plant cereal-grain grassbeds on exposed drawdown shorelines to increase juvenile fish habitat, localize productivity, and increase invertebrate fish prey. To determine the efficacy of these plantings, the abundance of juvenile black basses Micropterus spp. (20–55 mm standard length) and the amount of periphyton and macroinvertebrate prey were compared among three treatment types: (1) planted grassbeds of cereal barley Hordeum vulgare; (2) artificial rope grassbeds, which eliminated physical deterioration and nutrient release; and (3) nonplanted control sites …

Application Of Multiple Mass Spectrometric Techniques In The Isolation, Determination And Characterisation Of The Marine Toxins; Azaspiracid, Keith O'Callaghan
Theses
The Azaspiracids are a class of polyether marine biotoxins that present a significant health risk to consumers of shellfish. Since their discovery in the late 1990s several human intoxications have been reported, and traced to Irish shellfish farms resulting in closures or harvest rejections. As such incidents involving azaspiracid shellfish poisoning have a major socio-economic impact on communities that rely on shellfish farming revenues.
Recent evidence suggests that azaspiracid poisoning events may become more commonplace. With this increased risk the understanding of azaspiracids mode of action and biological targets becomes crucial. To support these endeavours effective analytical methods need to …
Peptide Hydrolysis And The Uptake Of Dipeptides By Phytoplankton, Margaret R. Mulholland, Cindy Lee
Peptide Hydrolysis And The Uptake Of Dipeptides By Phytoplankton, Margaret R. Mulholland, Cindy Lee
OES Faculty Publications
Rates of peptide hydrolysis (using the fluorescent substrate, lucifer yellow anhydride-labeled tetra-alanine) and dipeptide uptake (using dually labeled, 15N and 13C, dialanine) were measured in phytoplankton cultures and in natural populations during algal blooms dominated by one or two taxa. During most sampling events, both peptide hydrolysis and dipeptide uptake were greatest in the size fraction containing the dominant phytoplankter, suggesting that phytoplankton contribute substantially to or may even dominate observed extracellular peptide hydrolysis and dipeptide uptake in the environment. These are the first data suggesting that dipeptides may be taken up directly by phytoplankton and this may …

Spatial Variation In Otolith Chemistry Of Atlantic Croaker Larvae In The Mid-Atlantic Bight, Jason J. Schaffler, Christian S. Reiss, Cynthia M. Jones
OES Faculty Publications
Larval Atlantic croaker Micropogonias undulatus (1 to 7 mm in standard length) were collected on the east coast of the United States from North Carolina to Delaware during 2000. We defined 3 water-mass boundaries for potential groups of spawning Atlantic croaker using temperature and salinity measured at each sampling station. We tested the hypothesis that distinct otolith chemistries existed among 3 groups of larval Atlantic croaker collected from these water masses using solution-based inductively coupled plasma-mass spectrometry. Multivariate analysis of variance indicated that otolith chemistry differed significantly among water masses. Using a quadratic discriminant function, we were able to correctly …

Patterns Of Larval Atlantic Croaker Ingress Into Chesapeake Bay, Usa, Jason J. Schaffler, Christian S. Reiss, Cynthia M. Jones
OES Faculty Publications
We compared ingress patterns of Atlantic croaker Micropogonias undulatus larvae into Chesapeake Bay, USA, with published ingress patterns through barrier island inlets, the accepted model for larval fish ingress. This model asserts that larvae ingress on night flood tides at the flooddominated side of the inlet and at all depths. At the Chesapeake Bay mouth and in the adjacent coastal waters, we compared the distribution of abundance, size, age, and growth rates of croaker prior to ingress, In contrast to the barrier island inlet model, croaker larvae were more abundant at depth than closer to the surface regardless of location. …

Purple rice bran contains high amounts of natural antioxidants that consist of water- and lipid-soluble compounds. Hexane-insoluble and hexane-soluble fractions were separated from 100% methanolic extract from purple rice bran (RBE-HI and RBE-HS, respectively). Total anthocyanin, tannin, flavonoid, and phenolics contents were determined in those fractions, and their antioxidant capacities were evaluated by measuring 1,1-diphenyl-2- picrylhydrazyl (DPPH) radical scavenging capability, oxygen radical absorbance capacity (ORAC), and iron chelating capability (ICC). RBE-HI and RBE-HSwere also added to restructured patties made fromminced channel catfish (Ictalurus punctatus) belly flap meat. Lipid oxidation, color, and/or textural properties were determined for raw and …

Is Seagrass An Important Nursery Habitat For The Caribbean Spiny Lobster, Panulirus Argus, In Florida?, Donald C. Behringer, Mark J. Butler Iv, William Herrnkind, John H. Hunt, Charles A. Acosta, William C. Sharp
Biological Sciences Faculty Publications
Caribbean spiny lobster (Panulirus argus) settle preferentially in macroalgal-covered hard-bottom habitat, but seagrass is more prevalent in Florida (United States) and the Caribbean, so even low settlement of lobsters within seagrass could contribute substantially to recruitment if post-settlement survival and growth were high. We tested the role of seagrass and hard-bottom habitats for P. argus recruitment in three ways. We first explored possible density-dependent regulation of early benthic juvenile lobster survival within cages deployed in seagrass and hard-bottom habitats. Second, we compared settlement and survival of P. argus in both habitats, by comparing the recovery of microwire-tagged early …
The Occurrence Of Batrachochytrium Dendrobatidis In Salamander Populations Of West Virginia, Casey John Bartkus
The Occurrence Of Batrachochytrium Dendrobatidis In Salamander Populations Of West Virginia, Casey John Bartkus
Theses, Dissertations and Capstones
Batrachochytrium dendrobatidis (Bd), otherwise known as the amphibian chytrid fungus, is a pathogen that causes the amphibian disease, chytridiomycosis. While Bd has been found in amphibian populations in other states across the country, little is known of its occurrence in West Virginia. Batrachochytrium dendrobatidis is highly associated with amphibians in montane habitats. Because West Virginia has the highest mean elevation of any state east of the Mississippi River, amphibians in the state may be at high risk of an outbreak. The goal of this study was to determine the status of Bd in salamander species of concern in West Virginia. …

Regional Characterisation Of Hard-Bottom Nursery Habitat For Juvenile Caribbean Spiny Lobster (Panulirus Argus) Using Rapid Assessment Techniques, Rodney D. Bertelsen, Mark J. Butler Iv, William F. Herrnkind, John H. Hunt
Biological Sciences Faculty Publications
Shallow, hard-bottom habitat constitutes approximately 30% of the coastal waters of south Florida, United States, yet it is a chronically understudied feature of the marine seascape in this region. In this study, we characterised the general biogeographic and structural features of shallow benthic hard-bottom communities in the Florida Keys, and related those to the abundance of juvenile Caribbean spiny lobster (Panulirus argus), the target of one of Florida's most economically valuable fisheries. We used rapid assessment techniques to survey more than 100 hard-bottom sites in the Florida Keys to estimate the percentage bottom coverage of vegetation (seagrass and …

Inheritance Of Shoulder Spotting In The Tetra, Hyphessobrycon Bentosi Characidae, Jack Frankel
Inheritance Of Shoulder Spotting In The Tetra, Hyphessobrycon Bentosi Characidae, Jack Frankel
Department of Biology Faculty Publications
The tetra (Hyphessobrycon bentosi) exhibits two phenotypes associated with shoulder spotting. Fish possess ei-ther a prominent black vertical spot located directly behind the operculum (H. b. bentosi, bentosi white tip tetra) or lack this spot (H. b. rosaceus, rosy tetra). Segregation patterns observed from the progenies of ten different crosses suggest that the inheritance of these phenotypes is controlled by two autosomal loci acting in a complementary fashion, with domi-nance at both loci required for the expression of the spotted phenotype.

Population Structure Of The Invasive Round Goby In Lake Michigan, Elizabeth A. Larue, Carl Ruetz Iii, Ryan Thum
Student Summer Scholars Manuscripts
The recent establishment of the round goby (Neogobius melanostomus), an invasive fish in Lake Michigan, provides a model system to view fine scale evolutionary and ecological processes that can create genetic structure within a population. We used seven nuclear polymorphic microsatellite markers (N = 11-17 per site) and measurements of fish total length, weight, and sex (N = 20-74 per site) on round gobies captured by minnow traps and angling among 12 sites around the entire shore of Lake Michigan to determine if evolutionary processes are present in Lake Michigan by characterizing population structure of the round goby. Specific objectives …

A feeding trial was performed to investigate levels of corn-based Distillers Dried Grains with solubles (DDGS), a co-product of fuel ethanol manufacturing, used as a nutrient source for juvenile Nile tilapia (Oreochromis niloticus). Three isocaloric (15.7±0.7 MJ kg-1 dry matter), isonitrogenous (29.6±1.0% crude protein) experimental diets were formulated to contain 20, 30 and 40% DDGS; a commercial diet containing fishmeal was used as a reference diet. Glass aquaria (110 L) were stocked with Nile tilapia (mean weight = 6.7 g) with four replicates per diet. Weight gains, conversion ratios and hepatosomatic indices did not significantly differ between …

Effect Of Using Threadfin Shad As Forage For Channel Catfish Fed Daily Or Every Third Day, Bartholomew W. Green, Peter Perschbacher, Gerald Ludwig
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
We evaluated whether stocking threadfin shad Dorosoma petenense as a forage fish in multiple-batch production ponds for channel catfish Ictalurus punctatus could substitute for formulated feed when channel catfish were fed daily or every 3 d. A completely randomized design in a 232 factorial arrangement was used for the experiment, which was conducted in twelve 0.1-ha earthen ponds. Prespawn adult threadfin shad were stocked at 404 kg/ha in six of the ponds, whereas the remaining ponds received no threadfin shad. Channel catfish stockers (0.35 kg/fish) were stocked at 5,040 kg/ha, and fingerlings (28.2 g/fish) were stocked at 14,820 fish/ha. Channel …

As part of the Coral Reef Targeted Research (CRTR) Program, a partnership between the Global Environment Facility and the World Bank, our research team examined the recruitment patterns of Caribbean spiny lobster (Panulirus argus) postlarvae among regions in the Caribbean, with a particular focus on Mesoamerica. Our goal was to collect comparable information on postlarval supply among regions and to provide data to test predictions of connectivity generated from a coupled biophysical oceanographic model of lobster larval dispersal. Here we present the results of the postlarval recruitment monitoring program. We monitored the catch of postlarvae on Witham-style collectors …

Mechanism(S) Of 3-Trifluoromethyl-4-Nitrophenol (Tfm) Toxicity In Sea Lamprey (Petromyzon Marinus) & Rainbow Trout (Oncorhynchus Mykiss), Oana Birceanu
Theses and Dissertations (Comprehensive)
The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), has been extensively used over the last 50 years to control invasive sea lamprey (P. marinus) populations in the Great Lakes, but its mechanism of toxicity is unresolved. Due to its structural similarity to phenolic compounds known to inhibit mitochondrial ATP production, one hypothesis on the mode of action of TFM is that it impairs mitochondrial oxidative phosphorylation. A second hypothesis is that TFM targets the gill, interfering with gill ion uptake. Exposure of larval sea lamprey and rainbow trout (O. mykiss), to each animal’s respective 12-h TFM LC50, resulted in marked …

The Amphipod Orchomenella Pinguis — A Potential Bioindicator For Contamination In The Arctic, Lis Bach, Valery E. Forbes, Ingela Dahllöf
Valery Forbes Publications
Indigenous organisms can be used as bioindicators for effects of contaminants, but no such bioindicator has been established for Arctic areas. Orchomenella pinguis is a benthic amphipod, ubiquitous in the Arctic and can be found in high numbers. We collected O. pinguis at sites with different contamination levels. Population characteristics (body length distribution, average dry weight and amphipod organic content) were related to sediment contaminant concentrations, in order to identify suitable endpoints for using this species as a bioindicator. We show that O. pinguis was prevalent in both clean and contaminated areas, easy to sample and that its population characteristics …
Gene Expression Profiles As Molecular Indicators Of Dissolved Oxygen Stress In Grass Shrimp Palaemonetes Pugio Holthuis 1949, Tiandao Li
Dissertations
Occurrence and severity of hypoxia is increasing in coastal and estuarine environments, and recovery of impacted habitats and living resources is slow. Detection of early biological effects of hypoxia is needed for timely remedial action to be taken. The overall objectives of this research was to develop molecular indicators of dissolved oxygen stress to assess the biological impact of hypoxia in coastal estuaries and validate their use through a combination of laboratory and field studies. To achieve these goals, grass shrimp, Palaemonetes pugio, oxygen-sensitive and hypoxia-tolerant species abundant in estuarine systems, were exposed to hypoxia under controlled laboratory conditions, and …

Yellow Head Virus: Transmission And Genome Analyses, Hongwei Ma
Dissertations
Yellow head virus (YHV) is an important pathogen to shrimp aquaculture. Among 13 species of naturally YHV-negative crustaceans in the Mississippi coastal area, the daggerblade grass shrimp, Palaemonetes pugio, and the blue crab, Callinectes sapidus, were tested for potential reservoir and carrier hosts of YHV using PCR and real time PCR. The YHV replicated in the daggerblade grass shrimp, and was still detectable on 36 d post-inoculation, causing 8% mortality after injection. However, YHV did not replicate in the blue crab. These data suggest that the daggerblade grass shrimp could act as a reservoir host for YHV. Storage conditions of …

Unintended Facilitation Between Marine Consumers Generates Enhanced Mortality For Their Shared Prey, F. Joel Fodrie, Matthew D. Kenworthy, Sean P. Powers
University Faculty and Staff Publications
We manipulated predator densities and prey vulnerability to explore how interactions between two predators affect overall mortality of their shared prey. Our threemember study system included eastern oysters (Crassostrea virginica) and two of its major consumers: southern oyster drills (Stramonita haemastoma) and stone crabs (Menippe adina). Field experiments demonstrated that drills and crabs foraging together generated higher than expected oyster mortality based on each species operating independently, even though crabs also killed some drills. In subsequent laboratory trials, we experimentally mimicked the handling of oysters by foraging crabs and confirmed that crabs facilitated drills by breeching oyster valves, thereby granting …

Fisheries Research Report No. 182 - A Likelihood Analysis Of Non-Indigenous Marine Species Introduction To Fifteen Ports In Western Australia, Justin I. Mcdonald
Fisheries Research Reports
As an island continent, Australia is heavily dependent upon maritime transport with over 95% of its imports and exports transported by ship (Australian State of the Environment Committee, 2001). With about one third of Australia’s coastline, Western Australia ranks fourth of the six states and territory in the number of known non-indigenous marine species.
In this study fifteen ports in Western Australia were assessed on the potential for non-indigenous marine species to become introduced through ballast water and biofouling. The overall vesselmediated incursion risk to Western Australian ports was calculated by summing the relative incursion threat posed by visits to …

Movement Patterns And Multi-Scale Factors That Influence Exotic Brook Trout And Endemic Bonneville Cutthroat Trout Distribution And Abundance In The Mill Creek Drainage, Utah, Benjamin Keith Nadolski
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
Introduced brook trout (Salvelinus fontinalis) are implicated as a primary factor leading to the decline in distribution and abundance of native cutthroat trout (Oncorhynchus clarkii). However, not all introductions are successful, suggesting local conditions influence the success of invasions. Therefore, I sought to determine the multi-scale factor(s) that influence brook trouts’ invasion success of native Bonneville cutthroat trout (Oncorhynchus clarkii utah) habitats in Mill Creek, Utah. I conducted patch occupancy surveys to determine watershed-scale brook trout and cutthroat trout distribution. I also determined the relative abundance of brook trout and cutthroat trout at the …
