Prion diseases are fatal neurodegenerative disorders caused by the misfolding of the normal prion protein (PrPC) into its infectious form (PrPSc). While the zoonotic potential of chronic wasting disease (CWD) remains uncertain, the presence of prions in food products raises public health concerns. Additionally, therapeutic strategies for prion diseases remain limited. This thesis presents a methodological exploration of the Protein Misfolding Cyclic Amplification (PMCA) technique to address these challenges in two key areas: (1) detecting CWD prions in processed meats subjected to common cooking procedures, and (2) identifying potential anti-prion compounds through a high-throughput PMCA adaptation. …

Inflammation of the mammary gland, also known as mastitis, is a prevalent and financially impactful disease in sheep production. The objectives of this survey were to 1) evaluate current testing methods: somatic cell count (SCC), California Mastitis Test (CMT), and bacterial culture, as well as novel biomarkers: lamb performance, milk composition, and udder conformation, for their impact on ewes categorized as clinically healthy or unhealthy under intensive management systems at 15 days in milk (DIM) and 30 DIM, and 2) identify biological markers that are correlated with clinical and subclinical mastitis. The objectives of these studies were to evaluate the impact of retail display case type and muscle type on meat quality attributes, including color, lipid oxidation, texture, and volatile compound profiles, as well as to assess economic implications based on retail color degradation. Case type effects were further evaluated by considering environmental factors such as temperature, air velocity, and relative humidity and their influence on the shelf life of beef patties. Lighting influences broiler production, health, and behavior. Due to their advanced visual system, avian species perceive a wide spectrum of light. The objective of this study was to investigate the effects of white (W, 350 to 780 nm) blue (B, 450 nm), and green (G, 560 nm) light on broiler production, processing performance, activity, fear, stress, blood cell profiles, tibiotarsi (tibia) morphology, and depth perception. The rumen microbiome plays a critical role in ruminant health and performance. As a consequence, many studies investigate microbial community composition using 16S rDNA based sequencing. As such, data quality and data filtering are critical to accurately identify microbial community composition. In this study we use mock communities to empirically select data filtering parameters to reduce artifact populations and compared the effect of filtering using different bioinformatic pipelines using rumen bacterial community data. The filtering parameters identified provides consistent estimates of rumen microbial diversity, regardless of bioinformatic pipeline utilized and provides a more accurate view of microbiome structure and composition. …

This study focuses on Balistes capriscus (Gray Triggerfish), a reef-associated species dependent on hard-bottom habitats in the Gulf of Mexico (GOM). Using mitochondrial DNA barcoding and phylogenetic analyses, genetic structure across four sites was assessed: Rio Grande Valley (RGV), Veracruz, Chinchorro Bank, and Xahuayxol. Samples from RGV and Veracruz exhibited limited haplotype diversity, minimal nucleotide divergence, and weak genetic differentiation-patterns consistent with recent population expansion and ongoing connectivity within the GOM. While the study centered on B. capriscus, all individuals from Chinchorro and Xahuayxol unexpectedly grouped with Canthidermis maculata (Spotted Oceanic Triggerfish), suggesting misidentification, cryptic diversity, or taxonomic uncertainty in …

Project 1 focused on the central role that mitochondria play in ATP production, highlighting that variation within the mtDNA may influence feed efficiency. Variation in the mitochondrial genome was uncovered, and associations with feed efficiency in beef cattle were identified. Growth and feed intake phenotypes, as well as low-pass sequencing, were collected on beef steers across two populations. Protein coding variants were identified and used to classify animals into mitochondrial haplotypes. A linear mixed model was employed, which detected a significant association between mitochondrial haplotype and DMI. It is well understood that lactating dairy cows require metabolizable protein (MP) for milk yield and milk protein synthesis. Diets with conventional feed sources like corn and soybean meal (SBM) may not sufficiently meet all MP and amino acid (AA) requirements so supplementation with more rumen undegradable protein (RUP) rich feeds or rumen-protected AA can be fed to support animal performance. Asian carp species pose significant ecological threats to North American freshwater systems due to their invasive nature and prolific reproduction. Their semi buoyant eggs develop while drifting downstream with the river current. Understanding early life-stage transport is therefore critical for predicting recruitment potential and guiding management efforts. Existing fluvial egg drift models, including those relying on 1-D hydraulics and fully 3-D CFD-based models, are either inadequate for complex braided rivers or too computationally demanding for large-scale application. Heat stress (HS) is one of the most critical environmental challenges negatively impacting global poultry production, significantly compromising broiler performance, welfare, and economic viability. Specifically, heat stress profoundly disrupts physiological and metabolic processes, including hepatic lipid metabolism. Two broiler lines were divergently selected for high (HWE) or low (LWE) water efficiency. Heat-stressed HWE birds showed reduced fat content compared to their LWE counterparts, and HS reduced abdominal fat content in both lines. Pasture-raised broiler chickens face temperature extremes that can threaten welfare. In the study, we evaluated whether a 0.26 % tryptophan-supplemented diet (T) improved heat stress (HS) resilience in Ross-708 broilers raised on pasture. At D14, birds began the T or remained on the control diet (C) and were moved outdoors. On D20 and 41, half of each group underwent six-hour HS; the remainder stayed under thermoneutral (TN) conditions. Weekly body weight, feed intake on D20 and D41 were recorded, and fear was measured using the Tonic Immobility (TI) test. Neither diet nor HS affected body weight. Poly- and perfluoroalkyl substances (PFAS) are an anthropogenic group of chemicals known for their persistence in the environment and their potential to bioaccumulate. PFAS can bioaccumulate in largemouth bass (Micropterus nigricans), yet the effects of exposure on bass larvae are unknown. While largemouth bass are commonly raised for aquaculture, techniques for rearing bass which enable controlled experiments at known densities, a necessity for toxicity studies, are poorly understood. Background/Objectives: Chronic inflammation and Western-style diets elevate colorectal cancer (CRC) risk, particularly in individuals with colitis, a feature of inflammatory bowel disease (IBD). Diets rich in polyphenol-containing functional foods, such as cocoa, may reduce gut inflammation and modulate the gut microbiome. This study investigated the impact of cocoa polyphenol (CP) supplementation on inflammation and microbiome composition in mice with colitis, fed either a healthy or Western diet, before, during, and after the onset of disease. Blanding’s Turtle (Emydoidea blandingii) is a species of concern in North America as many populations have declined across its distribution. Environmental factors associated with alteration of natural ecosystems have played a role in declines leading to many isolated populations. Most populations exist in the Great Lakes region with a number of outlying populations, such as Nebraska. Understanding the current distribution of the species will facilitate regional management strategies to protect this species into the future. Anoplocephalid cestodes collected from the southern mountain viscacha, Lagidium viscacia, in Boliva in 1986 are here examined, and a new species of Viscachataenia is described. The new species has numerous body measurements that are larger than the existing species, V. quadrata, but the eggs and corresponding structures of the new species are all smaller than those of V. quadrata.

Aquatic oligochaetes include the union of the two multispecies families: Naididae and Tubificidae. Roads can negatively impact the wildlife communities they transect. Road mortalities from wildlife-vehicle collisions are one direct effect, but roads may also indirectly affect wildlife behavior, movement, or local distribution. Quantifying the spatial extent of these road-effects is important for understanding how roads indirectly affect species of concern, identifying potential sources of disturbance, and for estimating the effectiveness of road mitigation structures. While consequences of equine obesity are well defined, the extent to which persistent hyperleptinemia and accompanying leptin (Lep) resistance affect the fetal-maternal environment warrants further exploration. This study compared endocrine, fetal, placental, and uterine parameters during early pregnancy (through day 70) in mares with elevated leptin versus normal leptin (NL) concentrations. Fourteen mares were classified as either NL (n = 6; Lep 0.9 ± 0.3 ng/mL), moderate leptin (ML; n = 4; Lep 3.0 ± 0.3 ng/mL), or high leptin (HL; n = 4; Lep 5.8 ± 0.7 ng/mL) based on annual plasma leptin. Weaning a beef calf is likely the most stressful event in the animal’s life. Future growth and reproductive performance can be compromised by health challenges encountered through the weaning process. Consequently, proper management aimed at reducing stress and preparing the beef animal for the next stage of production is critical to reach its genetic potential. Proper management involves applying good animal husbandry techniques, using sound science in developing health and feeding protocols, and implementing low-stress animal handling practices.

The hippopotamus, commonly known as the hippo, is classified into two extant species: the common hippopotamus (Hippopotamus amphibius) and the pygmy hippopotamus (Choeropsis liberiensis). Pygmy hippos are fascinating creatures native to the forests and swamps of West Africa. Their survival is increasingly threatened by habitat loss resulting from agricultural expansion and urban development. This study presents the first report of sexual cannibalism in the medically significant scorpion Leiurus abdullahbayrami Yağmur, Koç & Kunt, 2009, that was recorded during a herpetological field trip to the semi-desert area in northern Lebanon. It broadens the understanding of sexual cannibalism in scorpions and adds a new prey item to the dietary spectrum of the scorpion.

Stream fishes in the southeastern United States are adapted to hydrological disturbances such as droughts. However, this region is projected to experience increasingly frequent and severe disturbances that will elevate water demands given increasing agricultural and urban development. Further, road-stream crossings (RSCs) are abundant features in these streams that can exacerbate drought-mediated stream fragmentation and impede movements by many aquatic organisms, including fishes, when poorly constructed. These compounding effects may adversely affect the diversity and abundances of resident stream fishes, but their interactions remain poorly understood, warranting rigorous testing of how droughts and RSCs influence stream fish dynamics.
