A mixed Eimeria spp. challenge model was designed to assess the effects of challenge on broiler chicken performance, intestinal integrity, and the gut microbiome for future use to evaluate alternative strategies for controlling coccidiosis in broiler chickens. The experimental design involved broiler chickens divided into two groups: a control group (uninfected) and a positive control group, infected with Eimeria acervulina (EA), Eimeria maxima (EM), and Eimeria tenella (ET). At day-of-hatch, 240 off-sex male broiler chicks were randomized and allocated to one of two treatment groups. Staphylococcus agnetis is an emerging pathogen in chickens but has been most commonly isolated from sub-clinical mastitis in bovines. Previous whole-genome analyses for known virulence genes failed to identify determinants for the switch from mild ductal infections in cattle to severe infections in poultry. We now report identification of a family of 15 kbp, 17–19 gene mobile genetic elements (MGEs) specific to chicken osteomyelitis and dermatitis isolates of S. agnetis. These MGEs can be present in multiple copies per genome. The MGE has been vectored on a Staphylococcus phage that separately lysogenized two S. agnetis osteomyelitis strains. …

Introduction: Coccidiosis caused by the Eimeria spp., an Apicomplexan protozoon, is a major intestinal disease that affects the poultry industry. Although most cases of coccidiosis are subclinical, Eimeria infections impair bird health and decrease overall performance, which can result in compromised welfare and major economic losses. Viable sporulated Eimeria oocysts are required for challenge studies and live coccidiosis vaccines. Potassium dichromate (PDC) is typically used as a preservative for these stocks during storage. The undomesticated wild populations of our domestic species deserve heightened consideration when it comes to their conservation. Not only do they carry significant socio-cultural and economic roles, but they offer an exceptionally unique case study for population genetics. The red junglefowl (Gallus gallus) of southern and south-eastern Asia is considered to be the living ancestor and progenitor of the domestic chicken. Heat stress (HS) is one of the most challenging stressors to poultry production sustainability. The adverse effects of HS range from feed intake and growth depression to alteration of meat quality and safety. As phytase supplementation is known to improve nutrient utilization and consequently growth, we undertook the present study to evaluate the effects of dietary phytase on growth and meat quality in heat-stressed broilers. Historical distribution and range expansion -- We reviewed existing information to evaluate conclusions by previous authors (e.g., Sharpe et al. 2001, Silcock and Jorgensen 2023) that woodcock have expanded their range west since settlement of Nebraska by Euro-Americans. We also provide additional context about the species’ distribution over time using regional resources.

Temporal occurrence and abundance -- We used data from woodcock outfitted with satellite transmitters 2021-2022 and compared it with occurrence data from two sources, eBird (eBird.org 2023) from all years through 2022 and the SFRs in the Nebraska Bird Review 1994–2019. The present study was designed to evaluate the effects of DL-methionine (DL-Met) 2-hydroxy-4-(methylthio) butanoic acid (HMTBa), or S-(5′-Adenosyl)-L-methionine chloride (SAM), using feeding trial and central administration, on live performance, plasma metabolites, and the expression of feeding-related hypothalamic neuropeptides in broilers raised to a market age (35 d). Final average body weight (BW) and feed conversion ratio (FCR) from the feeding trial exceeded the performance measurements published by the primary breeder. At d35, the MTBHa group had better BW and lower feed intake, which resulted in a better FCR than the DL-Met group at 87 TSAA to lysine. At the molecular …

Introduction: Drug-sensitive live coccidiosis vaccines have been used to control coccidiosis and renew drug sensitivity in commercial chicken operations. However, only limited species coverage vaccines have been available for commercial turkey producers. This study aimed to assess the effect of an E. meleagrimitis vaccine candidate, with and without amprolium intervention, on performance and oocyst shedding. Additionally, the effect of vaccination, amprolium treatment, and E. meleagrimitis challenge on intestinal integrity and microbiome composition was evaluated.

Mycoplasma gallisepticum (MG) can cause chronic respiratory disease in chickens and infectious sinusitis in turkeys. Impacts of MG infections can include increased morbidity and mortality, decreased egg production, hatchability and feed efficiency. Biosecurity and bio-surveillance remain the primary means of deterrence as the current means of control are not wholly effective. The objective of this study was to investigate the role of fine (F) and coarse (C) limestone ratios and exogenous phytase at standard and superdosing activity on performance, egg quality, and bone quality of post-peak and late-lay Hy-Line W-36 hens. For post-peak, treatments with 40F:60C at 0 and 400 FTU/kg (standard phytase activity) increased hen-day egg production (HDEP). Treatments with 15F:85C decreased feed intake (FI) but also increased unsaleable eggs (UE) and eggshell-breaking strength (EBS). The Obese strain (OS) of chickens spontaneously develops autoimmune thyroiditis and is a well-established biomedical model for Hashimoto’s thyroiditis in humans. Both conditions are characterized by the infiltration of the thyroid glands with mononuclear cells resulting in destruction of the thyroid tissue and impairment of the thyroid’s endocrinological functions. Past studies have described mononuclear cell infiltration in thyroids of the OS chickens, but the time-course, cell composition, and relative amounts of the various mononuclear cells infiltrating the thyroids has not been well defined. Lameness is a health condition threatening the welfare of broiler chickens in the poultry industry. It can be caused by a bacterial infection called Bacterial Chondronecrosis with Osteomyelitis (BCO). Zinpro has created Avalia-ZMC, a feed additive that has been proven to reduce lameness by 20% in wire-flooring pens and 25% in litter-flooring pens (Alrubaye et al., 2020). Breast myopathies have caused significant economic losses to the poultry industry over the last decade. The induction of woody breast (WB), white striping (WS), and spaghetti meat (SM) have caused textural issues, functional issues, and a decline in consumer acceptance. Of these three myopathies, WB causes the most pronounced effects on product quality and reduces functional use. Woody breast is also commonly associated with expansive breast meat growth/high performing broilers. The poultry industry has significant coccidiosis and necrotic enteritis (NE) challenges, leading to high mortality and unacceptable growth without antibiotic treatment. This research explored supplementing Nigella sativa (black cumin) seed oil in poultry feed to mitigate coccidiosis and prevent or lessen NE in broilers. In vivo studies consisted of 384 and 320 Cobb 500 male broiler chicks distributed in a randomized complete block experimental design for trials 1 and 2, respectively. A rising problem in agriculture is the increase of antibiotic-resistant Salmonella cases associated with chicken eggs, which transmit infection to humans. To counter this, new approaches to combat Salmonella in chickens and desiccated on eggshells are vital in the prevention of human foodborne illness. Disrupting signaling pathways with naturally occurring compounds provides a potential novel avenue for prevention of Salmonella infections, as this would disrupt sensing of these environments and inhibit subsequent optimal gene expression. Streptococcus gallolyticus (SG) is a Gram-positive found as commensal gut flora in animals and humans. SG has been associated with acute mortality in poults 2 to 3 weeks of age, with lesions mainly consisting of splenomegaly, hepatomegaly, and pericarditis. The objective of this thesis was to reproduce disease in poults caused by SG recovered from clinical field isolates and evaluate different routes of administration. The objective of this thesis was to investigate the effects of Enterococcus cecorum (EC) administration into the amnion at day 18 of embryogenesis on performance and septicemia development in broiler chickens. Chapter I consists of a brief overview of the thesis. Chapter II provides a literature review on EC-related disease in broiler chickens and methods to isolate, detect, and differentiate between commensal and pathogenic strains of EC. Commercial hatch cabinet environments promote replication of microorganisms. These pathogenic or apathogenic microorganisms may serve as pioneer colonizers of the gastrointestinal tract (GIT) of poultry. Some of these pioneer colonizers, such as Escherichia coli and Enterococcus spp., are opportunistic pathogens that lead to reduced performance in commercial poultry. Effective hatchery sanitation is imperative to limit contamination of naïve neonatal chicks and poults. Formaldehyde fumigation has been traditionally used to reduce the pathogen load in commercial hatch cabinets. Femur head necrosis (FHN), also known as bacterial chondronecrosis with osteomyelitis (BCO), has remained an animal welfare and production concern for modern broilers regardless of efforts to select against it in primary breeder flocks. Characterized by the bacterial infection of weak bone, FHN has been found in birds without clinical lameness and remains only detectable via necropsy. This presents an opportunity to utilize untargeted metabolomics to elucidate potential non-invasive biomarkers and key causative pathways involved in FHN pathology. Essential oils (EO) affect performance, intestinal integrity, bone mineralization, and meat quality in broiler chickens subjected to cyclic heat stress (HS). Day-of-hatch Cobb 500 male broiler chicks (n = 475) were randomly divided into four groups. Mitigating the negative effects of heat stress (HS) is a critical challenge for the global poultry industry. This study evaluated the thermoregulatory potential of 3 in ovo delivered bioactive substances using selected gut health parameters. Eggs were incubated and allotted to 5 groups, and respective bioactive substances delivered. In recent times, meat quality has become a key aspect of poultry production. In the current study, 2,400 sex separate broilers from 4 commercial strains were placed in 6 replications to evaluate various meat quality characteristics when grown to 2 market weights. Broilers were fed 1 of 2 diets with varying degrees of amino acid inclusion to evaluate meat quality of broiler raised on varying planes of nutrition.
