Apios (Apios americana; “apios”), a tuberous perennial legume in the Phaseoleae tribe, was widely used as a food by Native Americans. Work in the last 40 years has led to several improved breeding lines. Aspects of the pollination biology (complex floral structure and tripping mechanism) have made controlled crosses difficult, and the previous reports indicated that the plant is likely primarily an outcrosser. We used a pseudo-testcross strategy to construct a genetic map specific to the maternal parent. Bangladesh faces huge challenges in achieving food security due to its high population, diet changes, and limited room for expanding cropland and cropping intensity. The objective of this study is to assess the degree to which Bangladesh can be self-sufficient in terms of domestic maize, rice and wheat production by the years 2030 and 2050 by closing the existing gap (Yg) between yield potential (Yp) and actual farm yield (Ya), accounting for possible changes in cropland area. While the vast majority of genome size variation in plants is due to differences in repetitive sequence, we know little about how selection acts on repeat content in natural populations. Here we investigate parallel changes in intraspecific genome size and repeat content of domesticated maize (Zea mays) landraces and their wild relative teosinte across altitudinal gradients in Mesoamerica and South America. We combine genotyping, low coverage whole-genome sequence data, and flow cytometry to test for evidence of selection on genome size and individual repeat abundance. In many areas of the US, fresh locally grown berries are not available during the winter. With this in mind, a research study comprised of three experiments was conducted focused on cultivar selection for berry yield, number, sweetness and phytonutrient content. Using a capillary mat system with under bench heating within a double-layer polyethylene greenhouse, strawberries were grown in the Great Plains Region of the US during the winter. During experiment 1, 12 cultivars were grown; berries were weighed, counted and analyzed for sugars and phytonutrients. “Albion” plants produced a high number/mass of berries, had relatively high sugar content but …

Recently, imaged-based approaches have developed rapidly for high-throughput plant phenotyping (HTPP). Imaging reduces a 3D plant into 2D images, which makes the retrieval of plant morphological traits challenging. We developed a novel LiDAR-based phenotyping instrument to generate 3D point clouds of single plants. The instrument combined a LiDAR scanner with a precision rotation stage on which an individual plant was placed. A LabVIEW program was developed to control the scanning and rotation motion, synchronize the measurements from both devices, and capture a 360◦ view point cloud. Background: Switchgrass breeders need to improve the rates of genetic gain in many bioenergy-related traits in order to create improved cultivars that are higher yielding and have optimal biomass composition. One way to achieve this is through genomic selection. However, the heritability of traits needs to be determined as well as the accuracy of prediction in order to determine if efficient selection is possible.

As one of the key crop traits, plant height is traditionally evaluated manually, which can be slow, laborious and prone to error. Rapid development of remote and proximal sensing technologies in recent years allows plant height to be estimated in more objective and efficient fashions, while research regarding direct comparisons between different height measurement methods seems to be lagging. In this study, a ground-based multi-sensor phenotyping system equipped with ultrasonic sensors and light detection and ranging (LiDAR) was developed. Recycling of carbon (C) and nitrogen (N) from plants into soils is decisive for maintaining soil organic matter and soil fertility. Therefore, we quantified plant biomass and C and N in the shoots and roots from the topsoil layer for a wide range of annual crops grown under subtropical conditions. We grew 26 species, 13 main crops, and 13 cover crops, in the field in standard sowing arrangements. Root biomass was recovered from the 0.00-0.20 m soil layer at flowering, and shoot biomass was measured at flowering for all crops and at maturity only for the main crops. Root dry …

Uncertainty exists with corn (Zea mays L.) N management due to year-to-year variation in crop N need, soil N supply, and N loss from leaching, volatilization, and denitrification. Activeoptical reflectance sensing (AORS) has proven effective in some fields for generating N fertilizer recommendations that improve N use efficiency, but locally derived (e.g., within a US state) AORS algorithms have not been tested simultaneously across a broad region. The Wild Free-Roaming Horses and Burros Act (WFRHBA) of 1971 established all “unbranded or unclaimed” equids on U.S. public lands as “living symbols of the historic and pioneer spirit of the West.” Today, >72,000 feral horses (Equus ferus caballus) and burros (E . asinus ; WHB) live on western U.S. public rangelands. The number of WHBs exceeds the Bureau of Land Management’s maximum Appropriate Management Level (AML) of 26,715 by a factor of approximately 2.7 and has nearly doubled from 2007–2015. The AML was set to balance WHB numbers with rangeland health and support other uses such as wildlife habitat …

Our understanding of the spatial ecology of feral horses (Equus ferus caballus ) and burros (E . asinus ) in the United States is limited. Robust location data are needed to better understand the permeability of Bureau of Land Management Herd Management Area boundaries, relative to feral horse movement patterns and home ranges. Predicting developmental growth stages in soft red winter wheat (Triticum aestivum L.) (SRWW) could improve agronomic management in Kentucky. However, predicting SRWW development is complex due to vernalization requirement and photoperiod sensitivity differences of cultivars. The objectives of this study are to (1) determine ability of Kompetitive Allele Specific PCR (KASP) genotyping to predict phenotype; (2) determine the relative vernalization requirement (RVR) of 50 SRWW cultivars in a greenhouse (GH) assay; and (3) measure growing degree-days (GDD) required by cultivars to reach eight growth stages in a field assay. Soil and root microbial communities have been studied for decades, and the incorporation of high-throughput techniques and analysis has allowed the identification of endophytic/non-culturable organisms. This has helped characterize and establish the core microbiome of many model plant species which include underground and aboveground organs. Unfortunately, the information obtained from some of these model plants is not always transferable to other agronomic species. In this project, we decided to study the microbiome of the Nicotiana genus because of its importance in plant physiological and plant-microbe interactions studies. The act of topping tobacco (Nicotiana tabacum L.) involves the removal of the terminal bud or inflorescence of the tobacco plant. This practice ordinarily is accomplished by manually removing the top of each tobacco plant in an entire field which is labor intensive and costly. Chemical topping utilizes sucker control products to inhibit the terminal bud and axillary bud growth without manually removing the top of the tobacco plant. The relationship between biodiversity and ecosystem functioning is important in maintaining agroecosystem sustainability. Plant-microbe symbioses, such as exists between the grass tall fescue (Schedonorus arundinaceum) and the asexual fungal endophyte Epichloë coenophiala, can be utilized to enhance agroecosystem functions, such as herbivore resistance. “Novel” E. coenophiala strains that vary in the production of mammal- and insect-toxic compounds have been identified, inserted into tall fescue cultivars, and are planted in pastures globally. Novel fungal endophyte-tall fescue associations may have divergent ecosystem function effects. Zinc deficient soils, or soils with low Zn bioavailability, are widespread, which exacerbates Zn deficiency in human as crops grown on these soils have low Zn content. Often crop yields are also compromised. Fertilizers based on soluble Zn salts often have limited efficacy in such soils. In this research, we evaluate the performance of polymer coated and bare ZnO nanoparticles (NPs) in an attempt to overcome limitations of soluble Zn salts in alkaline soils. The area of vegetable production is growing rapidly world-wide, as are efforts to increase production on existing lands in these labor- and input-intensive systems. Yet information on nutrient losses, greenhouse gas emissions, and input efficiency is lacking. Sustainable intensification of these systems requires knowing how to optimize nutrient and water inputs to improve yields while minimizing negative environmental consequences. Mycobacterium avium subsp. paratuberculosis is responsible for paratuberculosis in animals. This disease, leading to an inflammation of the gastrointestinal tract, has a high impact on animal health and an important economic burden. The environmental life cycle of M. avium subsp. paratuberculosis is poorly understood and several studies suggest that free-living amoebae (FLA) might be a potential environmental host. FLA are protozoa found in water and soil that are described as reservoirs of pathogenic and non-pathogenic bacteria in the environment. Efforts to develop live attenuated vaccines against Mycobacterium avium subspecies paratuberculosis (Map), using indirect methods to screen Map deletion mutants for potential efficacy, have not been successful. A reduction in the capacity to survive in macrophages has not predicted the ability of mutants to survive in vivo. Previous studies for screening of three deletion mutants in cattle and goats revealed one mutant, with a deletion in relA (ΔMap/relA), could not establish a persistent infection. Further studies, using antigen presenting cells (APC), blood dendritic cells and monocyte derived DC, pulsed with ΔMap/relA or a 35 kDa Map membrane protein (MMP) revealed …

Phytochemicals such as tannins included in dairy cow nutritional programs, as a natural feed additive, promise to improve rumen bypass protein and consequently reduce the feeding cost by decreasing the amount of CP needed in the ration. Additionally, if tannins are fed along with high quality protein sources, this could improve the AA profile reaching the small intestine and potentially improving health and performance of dairy cows. The black pepper, specifically its alkaloid piperine, is a nontoxic, natural dietary compound with a broad range of physiological activity. In pregnant pigs, amino acid (AA) requirements represent the sum of those required for maintenance functions, protein retention and efficiency of utilizing AA intake for the aforementioned body processes. The NRC (2012) model assumed AA efficiency is constant across period of gestation; however this is not reflective of the changes in metabolic demand during gestation. Therefore, two experiments were conducted to evaluate the efficiency of utilizing SID Lys and Thr for whole body protein retention (kSIDLys and kSIDThr) in pregnant gilts during early, mid and late gestation.
