Chulalongkorn University Theses and Dissertations (Chula ETD)
Cholangiocarcinoma (CCA) is a highly heterogeneous and aggressive malignancy of the bile ducts, characterized by a poor prognosis and high mortality rate. Despite advancements in treatment, effective targeted therapies and reliable prognostic biomarkers remain limited. Ferroptosis, a regulated form of cell death, has emerged as a potential therapeutic target in various cancers. However, its role in CCA remains poorly understood. In this study, we conducted a comprehensive analysis of ferroptosis-related genes (FRGs) to predict the prognosis of CCA and identify potential therapeutic targets. Using the least absolute shrinkage and selection operator (LASSO) Cox regression analysis on a training dataset, followed …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Dry Dipterocarp Forests (DDFs) in Thailand face severe degradation, primarily from human-induced fires, which significantly diminishes ectomycorrhizal (ECM) fungal diversity and the productivity of valuable wild mushrooms, such as Astraeus odoratus. However, concrete guidelines for restoring ECM fungal diversity in these degraded areas are currently lacking. This research aimed to develop a sustainable approach to enhance ECM fungal diversity and facilitate the restoration of degraded DDFs by utilizing A. odoratus mycelia. Furthermore, we developed alginate-encapsulated A. odoratus mycelial beads to extend inoculum viability and applicability. The study was conducted at two degraded DDF sites: Ban Kor Thung and Ban Mae …

Machine Learning To Predict Outcome Of Inulin Supplementation In Children And Adolescent Obesity From Gut Microbiome Profile And Clinical Factors, Vitavat Aksornkitti
Chulalongkorn University Theses and Dissertations (Chula ETD)
Obesity is a non-communicable disease that arises from multiple factors, including lifestyle behaviors, dietary habits, and genetics. Despite the growing prevalence of obesity, traditional interventions such as increased physical activity and changes in diet and lifestyle often fail to achieve sustainable, long-term weight loss. Therefore, more effective strategies are needed to address this condition. Recent studies have highlighted the role of gut microbiota in obesity, illustrating that the obesogenic effects are transferable via fecal microbiota transplantation. However, research in this area has been limited, particularly among children in developing countries such as Thailand. Early studies have shown that variations in …

Large Language Models (LLMs) have revolutionized various sectors, including healthcare where they are employed in diverse applications. Their utility is particularly significant in the context of rare diseases, where data scarcity, complexity, and specificity pose considerable challenges. In the clinical domain, Named Entity Recognition (NER) stands out as an essential task and it plays a crucial role in extracting relevant information from clinical texts. Despite the promise of LLMs, current research mostly concentrates on document-level NER, identifying entities in a more general context across entire documents, without extracting their precise location. Additionally, efforts have been directed towards adapting ChatGPTfor token-level …

Liver transplantation is a life-saving procedure for patients with end-stage liver disease. There are two main challenges in liver transplant: finding the best matching patient for a donor and ensuring transplant equity among different subpopulations. The current MELD scoring system evaluates a patient's mortality risk if not receiving an organ within 90 days. However, the donor-patient matching should also consider post-transplant risk factors, such as cardiovascular disease, chronic rejection, etc., which are all common complications after transplant. Accurate prediction of these risk scores remains a significant challenge. In this study, we used predictive models to solve the above challenges. Specifically, …

Introduction: Frailty is the most common medical condition affecting the aging population, and its prevalence increases in the population aged 65 or more. Frailty is commonly diagnosed using the frailty index (FI) or frailty phenotype (FP) assessments. Observational studies have indicated the association of frailty with Alzheimer's disease (AD). However, the shared genetic and biological mechanism of these comorbidity has not been studied.
Methods: To assess the genetic relationship between AD and frailty, we examined it at single nucleotide polymorphism (SNP), gene, and pathway levels.
Results: Overall, 16 genome-wide significant loci (15 unique loci) (p meta-analysis < 5 × 10-8) and 22 genes (21 unique genes) were identified between AD and frailty using cross-trait meta-analysis. The 8 shared loci implicated 11 genes: CLRN1-AS1, CRHR1 …

The Mpro is a conserved viral enzyme essential for antiviral drug development. This study employed an integrated computational strategy combining ligand-binding pathway exploration and machine learning-based QSAR modeling to design potent SARS-CoV-2 Mpro inhibitors based on organoselenium (ebselen) and thioester scaffolds. Parallel Cascade Selection Molecular Dynamics-based Ligand Binding-Path Sampling (LB-PaCS-MD) simulations were utilized to elucidate plausible ligand binding pathways and conformations within the Mpro catalytic site. Ebselen …
Akt Targeting Of Resveratrol Derivatives In Non-Small Cell Lung Cancer, Bhurichaya Innets
Akt Targeting Of Resveratrol Derivatives In Non-Small Cell Lung Cancer, Bhurichaya Innets
Chulalongkorn University Theses and Dissertations (Chula ETD)
The Akt signaling pathway plays a crucial role in the survival and proliferation of cancer cells and has emerged as an attractive target for anticancer drug development. This study aimed to modify and enhance the efficacy of resveratrol in non-small cell lung cancer (NSCLC). Five resveratrol derivatives (RD) were evaluated for their anticancer activity and their ability to target the Akt/mTOR pathway in NSCLC cell lines, specifically A549 and H23. Structure-activity relationship (SAR) analysis revealed that two derivatives, RD2 and RD3, had very similar chemical structures but exhibited different levels of cytotoxicity. RD2 demonstrated significant anti-proliferative effects and induced apoptosis …
Investigation Of Soil Fungal Communities In Agricultural Areas From Northeast Thailand, Chayaporn Lakmuang
Investigation Of Soil Fungal Communities In Agricultural Areas From Northeast Thailand, Chayaporn Lakmuang
Chulalongkorn University Theses and Dissertations (Chula ETD)
Soil fungal communities are important for ecosystem processes and plant health, but their composition in agricultural areas is poorly understood, especially in Thailand nowadays. Furthermore, according to the WHO fungus Pathogen Priority List (WHO FPPL), several fungus species pose significant harm to human health, with some demonstrating high death rates. The aims of this study are to assess the diversity of soil fungi in northeast Thailand and identify potential pathogen reservoirs in response to environmental factors. Samples were collected from rice, cassava, rubber tree, and sugarcane fields across 18 provinces with 8 zones of geological features. We used next-generation sequencing …
Determination Of Nutrients Digestibility In Commercial Dry Dog Food Containing Various Protein Sources, Phuong Anh Ta
Determination Of Nutrients Digestibility In Commercial Dry Dog Food Containing Various Protein Sources, Phuong Anh Ta
Chulalongkorn University Theses and Dissertations (Chula ETD)
This study evaluated the nutritional adequacy and digestibility of commercial dry dog foods formulated with different protein sources (animal versus plant) and market tiers (premium versus economy). Eight commercial dog foods were analyzed for nutrient composition and amino acid profiles using proximate analysis and high-performance liquid chromatography (HPLC). Of these, four representative products were selected to assess digestibility through in vivo total fecal collection in Beagle dogs and in vitro digestion assays. All products exceeded 18% crude protein (the minimum crude protein requirement by AAFCO). However, deficiencies were examined in essential amino acids, particularly leucine, isoleucine, methionine, lysine, tryptophan, valine …

Executive Summary: Climate change is driving the intensification of marine heatwaves (MHWs), which pose a significant threat to seagrass ecosystems globally. Understanding the response of seagrass species to these extreme thermal events is crucial for predicting their resilience and informing conservation strategies. In this study, we evaluated the impacts of prolonged MHW conditions (30 °C) on the photophysiology, metabolism, and growth of three populations of Posidonia sinuosa from Western Australia (Shoalwater, Garden Island, and Jurien Bay). We aimed to determine whether populations with higher thermal optima exhibit higher levels of resilience to MHWs compared to populations with lower thermal optima.
