Two-Dimensional Gel Analysis Of The Heat-Shock Response In Marine Snails (Genus Tegula): Interspecific Variation In Protein Expression And Acclimation Ability, Lars Tomanek Aug 2005

Two-Dimensional Gel Analysis Of The Heat-Shock Response In Marine Snails (Genus Tegula): Interspecific Variation In Protein Expression And Acclimation Ability, Lars Tomanek

Biological Sciences

The degree to which temperature acclimation modifies the acute synthesis of the entire heat-shock protein (Hsp) complement is still unknown, but it may constitute an important mechanism for understanding the differences in acclimation ability among closely related ectothermic species that occupy widely varying thermal environments. In general, eurythermal (heat-tolerant) species modify physiological function in response to an increase in acclimation temperature to a greater extent than stenothermal (heat-sensitive) species. In the present work I used 35S-labelled amino acids and two-dimensional gel electrophoresis to test this assumption for how acclimation affects acute Hsp expression (referred to as phenotypic plasticity) in …

K4, K9 And K18 In Human Histone H3 Are Targets For Biotinylation By Biotinidase, Kenya A. Kobza, Gabriela Camporeale, Brian Rueckert, Alice Kueh, Jacob B. Griffin, Gautam Sarath, Janos Zempleni Aug 2005

K4, K9 And K18 In Human Histone H3 Are Targets For Biotinylation By Biotinidase, Kenya A. Kobza, Gabriela Camporeale, Brian Rueckert, Alice Kueh, Jacob B. Griffin, Gautam Sarath, Janos Zempleni

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Histones are modified post-translationally, e.g. by methylation of lysine and arginine residues, and by phosphorylation of serine residues. These modifications regulate processes such as gene expression, DNA repair, and mitosis and meiosis. Recently, evidence has been provided that histones are also modified by covalent binding of the vitamin biotin. The aims of this study were to identify biotinylation sites in histone H3, and to investigate the crosstalk among histone biotinylation, methylation and phosphorylation. Synthetic peptides based on the sequence of human histone H3 were used as substrates for enzymatic biotinylation by biotinidase; biotin in peptides was probed using streptavidin peroxidase. …

Perennials That Beat The Heat, Bob Henrickson Aug 2005

Perennials That Beat The Heat, Bob Henrickson

Institute of Agriculture and Natural Resources: News Releases

I have always admired plants that seem to enjoy blooming in hot weather. Perennial favorites like the lavender-blue spikes of Russian sage, the dependable black-eyed susan, purple coneflower and "Autumn Joy" sedum, have become mainstays in the border. We can also mention the "Moonbeam" coreopsis, dazzling daylilies, garden phlox, yarrow, hosta and the striking hardy hibiscus. All of the previous plants are excellent choices for the hot summer perennial garden, providing beauty without much care. Thankfully, there are many more summer bloomers to choose from that are starting to gain the attention they deserve. The next time you think about …

Estimation Of Genetic Parameters For Growth, Feed Consumption, And Conformation Traits For Double-Muscled Belgian Blue Bulls Performance-Tested In Belgium, N. Gengler, C. Seutin, F. Boonen, L. Dale Van Vleck

Department of Animal Science: Faculty Publications

For 1,442 Belgian Blue bulls performance- tested at the Centre de Selection de la Race Blanc-Bleue Belge, nine traits were observed: height at withers at 7 mo, height at withers at 13 mo, weight at 7 mo, weight at 13 mo, average feed consumption of concentrates, average daily gain, average feed consumption of concentrates per average daily gain, average feed consumption of concentrates per mean metabolic weight, and price per kilogram of live weight. This price is based on muscle conformation and is therefore used as muscle conformation score. Restricted maximum likelihood with a derivative-free algorithm was used to estimate …

Runx-Dependent Expression Of Pkc Is Critical For Cell Survival In The Sea Urchin Embryo, Carrie Dickey-Sims, Anthony J. Robertson, Dawn E. Rupp, John J. Mccarthy, James A. Coffman Aug 2005

Runx-Dependent Expression Of Pkc Is Critical For Cell Survival In The Sea Urchin Embryo, Carrie Dickey-Sims, Anthony J. Robertson, Dawn E. Rupp, John J. Mccarthy, James A. Coffman

Physiology Faculty Publications

BACKGROUND: Runx transcription factors play critical roles in the developmental control of cell fate and contribute variously as oncoproteins and tumor suppressors to leukemia and other cancers. To discover fundamental Runx functions in the cell biology of animal development, we have employed morpholino antisense-mediated knockdown of the sea urchin Runx protein SpRunt-1. Previously we showed that embryos depleted of SpRunt-1 arrest development at early gastrula stage and underexpress the conventional protein kinase C SpPKC1.

RESULTS: We report here that SpRunt-1 deficiency leads to ectopic cell proliferation and extensive apoptosis. Suppression of the apoptosis by pharmacological inhibition of caspase-3 prevents the …

Analysis Of Affymetrix Genechip Data Using Amplified Rna, Leslie Cope, Scott M. Hartman, Hinrich W.H. Gohlmann, Jay P. Tiesman, Rafael A. Irizarry Aug 2005

Analysis Of Affymetrix Genechip Data Using Amplified Rna, Leslie Cope, Scott M. Hartman, Hinrich W.H. Gohlmann, Jay P. Tiesman, Rafael A. Irizarry

Johns Hopkins University, Dept. of Biostatistics Working Papers

The standard method of target synthesis for hybridization to Affymetrix GeneChip® expression microarrays requires a relatively large amount of input total RNA (1-15 micrograms). When small biological samples are collected by microdissection or other methods, amplification techniques are required to provide sufficient target for hybridization to expression arrays. One amplification technique used is to perform two successive rounds of T7-based in vitro transcription. However, the use of random primers required to re-generate cDNA from the first round transcription reaction results in shortened copies of the cDNA, and ultimately the cRNA, transcripts from which the 5' end is missing. In this …

Effects Of Stretching On Jumping Performance, Carisa Chavez Aug 2005

Effects Of Stretching On Jumping Performance, Carisa Chavez

Masters Theses & Specialist Projects

Stretching has been a part of warm-up for a very long time. Some recent research has shown that stretching could possibly hinder performance and others have shown it enhances performance. The purpose of this study was to examine whether stretching has an effect on vertical jump, and if so, how long the effects last. Twenty Western Kentucky students (10 M and 10 F) performed three trials each. There was a non-stretching group, a pre-stretching group and a between-stretching group; each of the subjects performed all three. The stretching protocol included a static stretching routine of the gluteus maximus, hamstrings, quadriceps, …

Influence Of Neck-Rail Placement On Free-Stall Preference, Use, And Cleanliness, Cassandra B. Tucker, Daniel M. Weary, David Fraser Aug 2005

Influence Of Neck-Rail Placement On Free-Stall Preference, Use, And Cleanliness, Cassandra B. Tucker, Daniel M. Weary, David Fraser

Housing and Confinement of Farm Animals Collection

Three experiments examined how the presence of a neck rail at different heights and locations influenced dairy cattle behavior and stall cleanliness. Experiment 1 compared 4 levels of neck-rail height (102, 114, and 127 cm and no neck rail; presented at 160 or 180 cm from the curb) in a preference test. Cows (n = 10) showed no consistent preference based on neck-rail height, regardless of the horizontal position of the neck rail. When cows were restricted to each treatment in turn, however, time spent standing fully (with all 4 hooves) in the stall was least in the stall with …

The Clearance Of Protein-Bound Solutes By Hemofiltration And Hemodiafiltration, Timothy W. Meyer, Jason L. Walther, Maria Enrica Pagtalunan, Andres W. Martinez, Ali Torkamani, Patrick D. Fong, Natalie S. Recht, Channing R. Robertson, Thomas H. Hostetter

Chemistry and Biochemistry

Background. Hemofiltration in the form of continuous venovenous hemofiltration (CVVH) is increasingly used to treat acute renal failure. Compared to hemodialysis, hemofiltration provides high clearances for large solutes but its effect on protein-bound solutes has been largely ignored.

Methods. Standard clinical systems were used to remove test solutes from a reservoir containing artificial plasma. Clearances of the protein-bound solutes phenol red (CPR) and indican (CIN) were compared to clearances of urea (CUREA) during hemofiltration and hemodiafiltration. A mathematical model was developed to predict clearances from values for plasma flow Qp, …

Genetic Analysis Of Two Bile Salt Hydrolase Activities In Lactobacillus Acidophilus Ncfm, Olivia Mcauliffe, Raul J. Cano, Todd R. Klaenhammer

Biological Sciences

Two genes, bshA and bshB, encoding bile salt hydrolase enzymes (EC 3.5.1.24) were identified in the genome sequence of Lactobacillus acidophilus NCFM. Targeted inactivation of these genes via chromosomal insertion of an integration vector demonstrated different substrate specificities for these two enzymes.

Elevated Levels Of Trace Elements In Cores Of Otoliths And Their Potential For Use As Natural Tags, Benjamin I. Ruttenberg, Scott L. Hamilton, Michael J.H. Hickford, Georges L. Paradis, Michael S. Sheehy, Julie D. Standish, Ofer Ben-Tzvi, Robert R. Warner Aug 2005

Elevated Levels Of Trace Elements In Cores Of Otoliths And Their Potential For Use As Natural Tags, Benjamin I. Ruttenberg, Scott L. Hamilton, Michael J.H. Hickford, Georges L. Paradis, Michael S. Sheehy, Julie D. Standish, Ofer Ben-Tzvi, Robert R. Warner

Biological Sciences

Variation in the chemical composition of fish otoliths has been used in recent years to address a range of ecological questions, including levels of stock mixing, variation in habitat use, and rates of larval exchange. While some of these questions have been answered with varying success, the degree to which discrete populations are connected via larval exchange remains unknown. To identify larval sources using natural variation in otolith chemistry, we must distinguish and measure the chemical composition of the otolith core, the portion of the otolith formed at the spawning site. Using laser ablation inductively coupled plasma mass spectrometry (LA-ICPMS), …

The Development Of A Rigorous Sampling Methodology For A Long-Term Annual Index Of Recruitment For Finfish Species From South-Western Australia, Daniel J. Gaughan, Suzanne G. Ayvazian, G Nowara, M Craine Aug 2005

The Development Of A Rigorous Sampling Methodology For A Long-Term Annual Index Of Recruitment For Finfish Species From South-Western Australia, Daniel J. Gaughan, Suzanne G. Ayvazian, G Nowara, M Craine

Fisheries Research Reports

This study has determined the optimal strategies to achieve statistically robust indices of recruitment of five key finfish species in south-western Australia. For another two species such indices could not be established. The sampling strategies developed will initially focus on the most appropriate location, month, and lunar phase, after which fine-tuning of the sampling activity will be undertaken with respect to time of day, tide height, weather conditions etc. A significant outcome from the project is that five finfish species have a sampling strategy for recruits that can be utilized as a monitoring program for the recreational and commercial fisheries …

Management Of The Proposed South Coast Trawl Fishery. A Discussion Paper., Department Of Fisheries. Aug 2005

Management Of The Proposed South Coast Trawl Fishery. A Discussion Paper., Department Of Fisheries.

Fisheries Management Papers

In late 1987, the Minister approved a development plan for the South Coast Inshore Trawl Fishery (Fisheries Management Paper No. 13) and made a number of notices regulating trawling activities in the Recherche Archipelago. Fishing Boat Licences were subsequently ‘endorsed’ by reference to those notices. The development plan was declared to be for a period ending 31 December 1989, subject to extension for a further year if necessary. Despite the considerable interest in the fishery during 1986 to 1989, fishing results demonstrated that the economic returns were marginal and a number of operators withdrew from participation in the fishery (at …

Human Activity Mediates A Trophic Cascade Caused By Wolves, Mark Hebblewhite, Clifford A. White, Clifford G. Nietvelt, John A. Mckenzie, Tomas E. Hurd, John M. Fryxell, Suzanne E. Bayley, Paul C. Paquet Aug 2005

Human Activity Mediates A Trophic Cascade Caused By Wolves, Mark Hebblewhite, Clifford A. White, Clifford G. Nietvelt, John A. Mckenzie, Tomas E. Hurd, John M. Fryxell, Suzanne E. Bayley, Paul C. Paquet

Biological Sciences Faculty Publications

Experimental evidence of trophic cascades initiated by large vertebrate predators is rare in terrestrial ecosystems. A serendipitous natural experiment provided an opportunity to test the trophic cascade hypothesis for wolves (Canis lupus) in Banff National Park, Canada. The first wolf pack recolonized the Bow Valley of Banff National Park in 1986. High human activity partially excluded wolves from one area of the Bow Valley (low-wolf area), whereas wolves made full use of an adjacent area (high-wolf area). We investigated the effects of differential wolf predation between these two areas on elk (Cervus elaphus) population density, adult …

Using Airborne Multispectral Imagery To Evaluate Geomorphic Work Across Floodplains Of Gravel-Bed Rivers, M. S. Lorang, D. C. Whited, Richard F. Hauer, J. S. Kimball, Jack Arthur Stanford Aug 2005

Using Airborne Multispectral Imagery To Evaluate Geomorphic Work Across Floodplains Of Gravel-Bed Rivers, M. S. Lorang, D. C. Whited, Richard F. Hauer, J. S. Kimball, Jack Arthur Stanford

Biological Sciences Faculty Publications

Fluvial processes of cut and fill alluviation and channel abandonment or avulsion are essential for maintaining the ecological health of floodplain ecosystems characteristic of gravel-bed rivers. These dynamic processes shape the floodplain landscape, resulting in a shifting mosaic of habitats, both above and below ground. We present a new and innovative methodology to quantitatively assess the geomorphic work potential necessary to maintain a shifting habitat mosaic for gravel-bed river floodplains. This approach can be used to delineate critical habitats for preservation through land acquisition and conservation easements, often critical elements of river restoration plans worldwide. Spatially explicit modeling of water …

Analyses Of High Resolution Hyperspectral Imagery For Characterization Of Ponderosa Pine Woodlands, Cullen R. Robbins

School of Natural Resources: Dissertations, Theses, and Student Research

The Bessey Unit of the Nebraska National Forest is a planted ponderosa pine forest located in the Nebraska Sand Hills. Planted in the early 20th Century, it provides a unique opportunity to study the effects of ponderosa pine establishment on the surrounding grassland ecosystem and the effects of increasing pine density on the forest ecosystem. It has been hypothesized that there are key levels of canopy cover at which shifts in ecosystem function occur. The goal of this research was to use remotely sensed data to develop a reliable method for estimating canopy cover. More specifically, canopy cover was estimated …

Ontology Learning As A Use-Case For Neural-Symbolic Integration, Pascal Hitzler, Sebastian Bader, Artur Garcez

Computer Science and Engineering Faculty Publications

We argue that the field of neural-symbolic integration is in need of identifying application scenarios for guiding further research. We furthermore argue that ontology learning - as occurring in the context of semantic technologies - provides such an application scenario with potential for success and high impact on neural-symbolic integration.
