Src-Dependent Tyrosine Phosphorylation At The Tips Of Growth Cone Filopodia Promotes Extension, Estuardo Robles, Stephanie Woo, Timothy M. Gomez
Neuroscience: Faculty Publications
Extracellular cues guide axon outgrowth by activating intracellular signaling cascades that control the growth cone cytoskeleton. However, the spatial and temporal coordination of signaling intermediates remains essentially unknown. Live imaging of tyrosine phosphorylation in growth cones revealed dynamic phospho-tyrosine (PY) signals in filopodia that directly correlate with filopodial behavior. Local PY signals are generated at distal tips of filopodia during extension and are lost during retraction. Active Src family kinases localize to the tips of filopodia, and Src activity regulates both filopodial dynamics and local PY signaling. Positive guidance cues stimulate filopodial motility by locally increasing tyrosine phosphorylation in a …
Fatigue Resistance Of Rat Extraocular Muscles Does Not Depend On Creatine Kinase Activity, Colleen A. Mcmullen, Katrin Hayeß, Francisco H. Andrade
Fatigue Resistance Of Rat Extraocular Muscles Does Not Depend On Creatine Kinase Activity, Colleen A. Mcmullen, Katrin Hayeß, Francisco H. Andrade
Physiology Faculty Publications
BACKGROUND: Creatine kinase (CK) links phosphocreatine, an energy storage system, to cellular ATPases. CK activity serves as a temporal and spatial buffer for ATP content, particularly in fast-twitch skeletal muscles. The extraocular muscles are notoriously fast and active, suggesting the need for efficient ATP buffering. This study tested the hypotheses that (1) CK isoform expression and activity in rat extraocular muscles would be higher, and (2) the resistance of these muscles to fatigue would depend on CK activity.
RESULTS: We found that mRNA and protein levels for cytosolic and mitochondrial CK isoforms were lower in the extraocular muscles than in …

Composition And Methods Of Using The Mirabilis Mosaic Caulimovirus Sub-Genomic Transcript (Sgt) Promoter For Plant Genetic Engineering, Indu B. Maiti, Nrisingha Dey
Kentucky Tobacco Research and Development Center Faculty Patents
The isolation of and methods of using a sub-genomic transcript (Sgt) promoter from Mirabilis mosaic virus (MMV) are described. A 333 bp MMV Sgt promoter fragment (sequence−306 to +27 from the transcription start site, TSS) was found to be sufficient for strongest promoter activity. This MMV Sgt promoter fragment shows comparable promoter activity to the MMV FLt promoter both in transgenic plants and in protoplasts. The MMV Sgt promoter also demonstrates much greater activity compared to Cauliflower mosaic virus (CaMV) 19S promoter and 35S promoter. The MMV Sgt promoter fragment and any chimeric gene to which it may be linked …
Isolation, Purification, And Chemical Characterization Of The Dihydroxamate-Type Siderophore, "Schizokinen," Produced By Rhizobium Leguminosarum Iari 917., Erin P. Storey
Electronic Theses and Dissertations
Iron is essential to the majority of microorganisms; it is an important cofactor in many cellular processes and enzymes. However in an aerobic environment and at biological pH, iron is primarily found as insoluble oxyhydroxides and is unavailable to microorganisms. Many bacteria have the ability to produce siderophores, low molecular weight compounds that have a high affinity for Fe3+. Siderophores are part of a multi-component system that actively transports the iron-siderophore complex into the cytoplasm. Rhizobia are characterized by their ability to form symbiotic relationships with leguminous plants, where they can fix nitrogen for the host plant and …

Effects Of The Implementation Of A Pilot Nutrition Education Program In A Rural Appalachian County., Leigh A. Davenport
Electronic Theses and Dissertations
The purpose of the study was to determine if nutrition knowledge would increase, indicated by improvements in pretest and posttest scores, following implementation of a nutrition lesson plan. The subjects included 532 fourth through eighth grade students from Johnson County, a rural county in northeast Tennessee. The lesson plans and pretests and posttests were developed to be appropriate for each grade level, with increased complexity in higher grades. The principal investigator graded the pretests and posttests and analyzed the data using SPSS. The results showed a significant improvement from pretests to posttests for all grades given a p-value of <0.05.

A Study Of The Relationship Of Self-Reported Resistance Training To Lipid Profiles., Melissa Davis
Electronic Theses and Dissertations
The primary purpose of this study was to determine the relationship of self-reported resistance training to lipid profiles. The study consisted of 10 subjects between the ages of 19 and 35. Participants were assigned to an exercise group or control group based on self-reported resistance training and according to ACSM standards. After means were determined for the data gathered, the exercise group had lower averages of weight, percent body fat, total cholesterol, low-density lipoprotein and triglycerides, as well as, a higher average of high-density lipoproteins. Two sample t-tests were performed to determine significance. It was determined that no significant difference …

Several primary murine and human B lymphomas and cell lines were found to constitutively express high levels of the activated form of c-jun N-terminal kinase (JNK), a member of the mitogen-activated protein (MAP) kinase family. Proliferation of murine B lymphomas CH31, CH12.Lx, BKS-2, and WEHI-231 and the human B lymphomas BJAB, RAMOS, RAJI, OCI-Ly7, and OCI-Ly10 was strongly inhibited by SP600125, an anthrapyrazolone inhibitor of JNK, in a dose-dependent manner. The lymphoma cells underwent apoptosis and arrested at the G2/M phase of cell cycle. Furthermore, JNK-specific small interfering RNA (siRNA) inhibited the growth of both murine and human B lymphomas. …

Comparative Genetic Studies Of Native And Introduced Coccinellidae In North America, Elliot S. Krafsur, John J. Obrycki, James D. Harwood
Entomology Faculty Publications
During the past four decades, several species of aphidophagous Coccinellidae became established in North America, including Coccinella septempunctata, Harmonia axyridis, Hippodamia variegata, and Propylea quatuordecimpunctata. After their establishment, unknown circumstances favoured a rapid increase in population densities and distribution of H. axyridis and C. septempunctata at localities hundreds and thousands of kilometers from their release sites. Propylea quatuordecimpunctata and Hippodamia variegata have spread more slowly after becoming established in northeastern North America. Comparative studies based upon allozyme variation in these four introduced species and in six native North American species of ladybird beetles revealed …
Quantifying Aphid Predation Rates Of Generalist Predators In The Field, James D. Harwood, John J. Obrycki
Quantifying Aphid Predation Rates Of Generalist Predators In The Field, James D. Harwood, John J. Obrycki
Entomology Faculty Publications
The community of predators within agroecosystems has the potential to restrict aphid populations, especially early in the season before exponential increases in density and prior to the arrival of specialist natural enemies. Although direct observations of predation, laboratory feeding trials and manipulative field studies have been used to estimate levels of biological control exerted by different species (or potentially negative interactions between them), it is often difficult to extrapolate results to naturally occurring interactions in the field.
Over 100 investigations have utilized gut-content analysis to estimate aphid predation rates by predators. Throughout the last century, gut dissection, which enables the …

Role Of Spontaneous Current Oscillations During High-Efficiency Electrotransformation Of Thermophilic Anaerobes, Michael V. Tyurin, Charles R. Sullivan, Lee R. Lynd
Dartmouth Scholarship
Current oscillations at about 24 MHz were observed during electrotransformation (ET) of the thermophilic anaerobes Clostridium thermocellum ATCC 27405, C. thermocellum DSM 1313, and Thermoanaerobacterium saccharolyticum YS 485, using a pulse gated by a square signal generated by a custom generator. In experiments in which only the field strength was varied, all three of these strains resulted in a one-to-one correspondence between the appearance of current oscillations and successful ET. Oscillations accompanied ET of both C. thermocellum strains only at field strengths of >12 kV/cm, and ET was only observed above the same threshold. Similarly, for T. saccharolyticum, oscillations were …

Interpretive Signs Designed To Trigger Naturalist Intelligence At Two American Zoos, Martha Bryant
Graduate Student Dissertations, Theses, Capstones, and Portfolios
An investigation of interpretive graphics was conducted in 2005 at two mid-sized AZA-accredited zoos, Lowry Park Zoo, Tampa, Florida and Knoxville Zoo, Knoxville, Tennessee. The Lowry Park Zoo study investigated signs at a red-tailed hawk and sandhill crane exhibit. Combination signs and wordless signs were more effective helping visitors see animals, increasing holding time, and number of engagements than treatments of no signs, or signs with words only. A second study, at Knoxde Zoo, tested combination and wordless signs in a children's zoo, investigating 31 signs at a 3.5-acre exhibit. Comparisons of visitors seeing the animals/using interactive exhibit elements, holding …

G protein betagamma subunits associate with many binding partners in cellular signaling cascades. In previous work, we used random-peptide phage display screening to identify a diverse family of peptides that bound to a common surface on Gbetagamma subunits and blocked a subset of Gbetagamma effectors. Later studies showed that one of the peptides caused G protein activation through a novel Gbetagamma-dependent, nucleotide exchange-independent mechanism. Here we report the X-ray crystal structure of Gbeta(1)gamma(2) bound to this peptide, SIGK (SIGKAFKILGYPDYD), at 2.7 A resolution. SIGK forms a helical structure that binds the same face of Gbeta(1) as the switch II region …

Application Of A Multiple Testing Procedure Controlling The Proportion Of False Positives To Protein And Bacterial Data, Merrill D. Birkner, Alan E. Hubbard, Mark J. Van Der Laan
U.C. Berkeley Division of Biostatistics Working Paper Series
Simultaneously testing multiple hypotheses is important in high-dimensional biological studies. In these situations, one is often interested in controlling the Type-I error rate, such as the proportion of false positives to total rejections (TPPFP) at a specific level, alpha. This article will present an application of the E-Bayes/Bootstrap TPPFP procedure, presented in van der Laan et al. (2005), which controls the tail probability of the proportion of false positives (TPPFP), on two biological datasets. The two data applications include firstly, the application to a mass-spectrometry dataset of two leukemia subtypes, AML and ALL. The protein data measurements include intensity and …
