Bone mineral density (BMD), among other factors, largely effect the initial stability of the cementless tibial tray component in a total knee replacement (TKR), where increased motion at the tray-bone interface hinders bony ingrowth. With a lack of bony ingrowth, the cementless implant will not experience long-term success. Understanding which factors influence initial stability yields insight into surgical technique considerations and help inform a surgeon’s implant choice. Engineering of bulk tissues has been limited by the lack of nutrient and waste exchange in these tissues without an adjacent capillary network. To produce microvasculature, a scaffold must be produced that provides temporary mechanical support and stimulate endothelial cell adhesion, growth, and morphogenesis into a vessel. Hypoplastic left heart syndrome (HLHS) is a type of heart defect where the left ventricle is underdeveloped or not developed, resulting in only a single functioning right ventricle. Approximately 7.5% of patients with congenital heart disease are born with a single ventricle (SV) which is accompanied by a spectrum of other malformations such as atrophied ascending aorta, atrial septal defects, and ventricular septal defects (VSD). Endothelial cells (ECs) form the innermost layer of all vasculature and constantly receive both biochemical and biomechanical signals, yielding a plethora of biomechanical responses. In response to various biochemical or biomechanical cues, ECs have been documented to generate biomechanical responses such as tractions and intercellular stresses between the cell and substrate and between adjacent cells in a confluent monolayer, respectively. Thus far, the ability of endothelial tight junctions and adherens junctions to transmit intercellular stresses has been actively investigated, but the role of gap junctions is currently unknown. Motivation: Long term tissue survivability ex-vivo can greatly facilitate research on the influence of external stimulus (loading, radiation, microgravity) on the tissue, including mechanisms of disease transmission and subsequent drug discoveries. Total Shoulder Arthroplasty (TSA) is a surgery which replaces the shoulder joint, or the interface between the humerus and the scapula glenoid. To test TSA success, most prior research compares patients with TSA to healthy controls. However, the shoulder anthropometry, motion, and musculature of individuals varies widely across the population making it important to assess TSA performance in individuals. This thesis is an examination of two material systems derived from polylactic acid (PLA) and polyethylene glycol (PEG). PLA is a polymer commonly sourced from renewable sources such as starches and sugars. It is a relatively strong, biodegradable polymer, making it ideal for use in the body. Even though it has a relative high strength, PLA is also brittle leading to the use of plasticizers to increase flexibility. The high heat transfer capabilities of impinging jets have led to their widespread use in industrial applications, such as gas turbine cooling. These impinging jets are usually manufactured on the walls of super-alloy metals and are influenced by being positioned with a confined setting. Studies have been shown to enhance the heat transfer of impinging jets by fluctuating the flow which will be analyzed in this project with two designs. Three-dimensional scaffolding is an emerging research area in biomedical and tissue engineering. Scaffolds provide the possibility of growing tissues in a controlled environment, with desired characteristics and properties towards a specific application. A new method to 3D print biodegradable and biocompatible material called Polyglycerol Sebacate Acrylate (PGSA) is demonstrated. PGSA is essentially an acrylated form of PGS using photoinitiator to become a photocurable resin suitable for liquid crystal display (LCD) 3D printing. Congenital Heart Disease (CHD) occurs in about 1\% (40,000) of newborn babies each year in the United States alone. About 10.9\% (960) of whom suffer from Hypoplastic Left Heart Syndrome (HLHS) - a subset of CHD where children are born with a single-ventricle (SV). A series of three surgeries are carried out to correct HLHS culminating in the Fontan procedure where venous flow returns passively to the lungs. The current configuration for the Fontan results in elevated Central Venous Pressure (CVP), inadequate ventricular preload, and elevated Pulmonary Vascular Resistance (PVR) leading to a barrage of disease. Current forensic approaches are limited in assessing biomechanical compatibility of short-distance falls involving children due to a lack of reliable witnessed falls with known injury outcomes. Powder injection molding (PIM) process simulations can be performed to minimize the number of injection molding experiments by estimating material properties necessary for PIM simulations. In current work, lead zirconate titanate (PZT) powder-polymer binder feedstock was compounded for 45 vol. % and 52 vol. % solids loading. PIM experiments on designed micro-pillar array geometry were performed using 52 vol. % PZT. Using PIM experiments results as basis, PIM simulations were performed on designed micro-pillar array geometries to understand the effectiveness of PIM simulations with the use of estimated feedstock properties in predicting molding behavior that have micro-features. Based on the principles of cutting edge bone remodeling research, a unique therapeutic exercise device was designed specifically to improve bone quality at the most critical location of the proximal femur prone to fracture: the superior-lateral femoral neck where the fracture first initiates during a fall. The exercise/device is intended to work by inducing enough strain in the bone to stimulate the body’s natural bone remodeling mechanisms to increase bone density in the proximal femur and consequently prevent a fracture from arising if a fall to the side does occur.

Human lumbar spine mechanics are influenced by soft tissue structures. Understanding and properly modeling these structures can help determine pathology, treatment, and implant design and performance. Finite element models of the L4-L5 level of the lumbar spine are often used, which include a representation of the intervertebral disc and spinal ligaments. Validation of these models are typically based on torque rotation data from a single subject or the models use average properties reported in literature. The natural knee is a hinge joint with significant functional requirements during activities of daily living; as a result, acute and chronic injuries can occur. Pathologies are influenced by joint anatomy and may include patellar maltracking, cartilage degeneration (e.g. osteoarthritis), or acute injuries such as meniscal or ligamentous tears. Human lung undergoes respiration-induced deformation due to sequential inhalation and exhalation. Accurate determination of lung deformation is crucial for tumor localization and targeted radiotherapy in patients with lung cancer. Numerical modeling of human lung dynamics based on underlying physics and physiology enables simulation and virtual visualization of lung deformation. Dynamical modeling is numerically complicated by the lack of information on lung elastic behavior, structural heterogeneity as well as boundary constrains. An effective treatment method for chronic pain is postural correction through therapist. Therapeutic proteins are used to successfully treat hemophilia, Crohn's Disease, diabetes, and cancer. Recent product recalls have occurred because of sub-visible particle formation resulting from the inherent instability of proteins. It has been suggested that particle formation is associated with late stage processing steps of filling, shipping, and delivery.
