Nanomechanical Characterization Of Vascular Tissue Using Atomic Force Microscopy, Ana Isabel Delgado Peralta

Cardiovascular morphogenesis and pathophysiology have been associated with the mechanical properties of the vessel wall. In this work, nanomechanical properties of diseased arteries and control groups were quantified using the Atomic Force Microscopy (AFM). Both tip sizes of 20nm and 5µm were used for examining the local and global stiffness. First, we measured the stiffness as a potential index factor of the mice’s aortic aneurysm. Results have shown the stiffness of mice aortic aneurysm is 58.84 ± 30.26 kPa, which is significantly different (P < 0.05) from the stiffness of the control group of 4.70 ± 17.41 kPa. This was attributed to the degradation of elastin fibers and increment of collagen deposition in the vessel as observed in the histological analysis. The lesion heterogeneity was illustrated by the stiffness gradient from 40 kPa to 1.2 MPa within a 2µm2 region. We also examined the stiffness of the diabetic rat aorta as a potential biomarker for disease progression. Results have shown a significantly higher stiffness of 70.25 ± 14.10 kPa in diabetic rat aorta, compared with 54.19 ± 14.15 kPa in the control group. Lastly, we characterized the human aortic calcifications to fill the knowledge gap of scarce datasets for calcification stiffness. Results have demonstrated the calcification stiffness as 61.5 ± 35 kPa, which is significantly larger than the stiffness of the control group of 12.0 ± 4.4 kPa. The stiffness variation was attributed to the percentage of calcium deposits, observed from the histological analysis. These nanomechanical characterizations could be further used for a better understanding of arterial adaptation and to provide insights for effective vascular therapies.

This study describes the deployment, integration, and demonstration of the Smartphone Video Guidance Sensor (SVGS) as novel technology for autonomous 6 DOF proximity maneuvers and precision landing of a quadcopter drone. The proposed approach uses a vision-based photogrammetric position and attitude sensor (SVGS) to estimate the position of an illuminated landing target after video capture. A visual inertial odometry camera (VIO) is used to provide position estimates of the UAV in a ground coordinate system during flight on a GPS denied environment. While unidirectional microphones remain prohibitively expensive for ubiquitous usage, externally applying a custom structure around an inexpensive omnidirectional microphone can also grant it desired directionality. Additively manufactured tungsten was printed in pure nitrogen, nitrogen-2.5% hydrogen, and nitrogen-5% hydrogen atmospheres as part of a 2^3 full factorial designed experiment and subjected to room temperature and high-temperature three-point-bend testing, chemical analysis, hardness testing, and microstructural imaging techniques. The pure nitrogen specimens exhibited the highest strength and ductility at both high temperature and room temperature. Chemical analysis showed a 2-8x reduction in compositional oxygen relative to unprocessed powder. Hardness values for all samples was between 306.8 and 361.5 HV1. It is proposed that adding hydrogen into the build atmosphere reduced the available energy density for tungsten melting by …

Trips and falls are one of the most common causes for injuries among elderly. The existing trip-and-falls studies primarily focus on the proactive fall prevention approaches, while active prevention strategies remain largely unexplored. This thesis aims to provide first steps towards active trip-and-fall prevention by developing various algorithms capable of detecting trip in human walking faster than the human voluntary reactions (~200 ms). This work centers on the development and the down-selection of nano-manufactured devices to be used in conjunction with Raman spectroscopy for probing a branched chain amino acid. The nano-manufactured devices integrate plasmonic nanoantennas for the purpose of amplifying molecular fingerprints, which are otherwise difficult to detect, through Surface Enhanced Raman Spectroscopy (SERS). Plasmonic nanostructures can be utilized for a variety of biomedical and biochemical applications to detect the characteristic fingerprint provided by Raman Spectroscopy. The goal of this research project was to develop an infant air-jet dry powder inhaler (DPI) platform for high-efficiency nose-to-lung (N2L) aerosol administration to infants through optimization of individual system components followed by expansion of the system to higher doses and different formulations using a new approach. Machine learning techniques using artificial neural networks (ANNs) have proven to be effective tools to rapidly mimic first principles calculations. These tools are capable of sub meV/atom accuracy while operating with linear scaling with respect to the system size. Here novel interatomic potentials are constructed based on the rapid artificial neural network (RANN) formalism. This approach generates precise force fields for various metals that have historically been difficult to describe at the atomic scale. This work focuses on understanding the behavior of high strength concrete in high strain rate ballistic impact loading scenarios. A finite element analysis was created with the implementation of the Concrete Damage and Plasticity Model 2 (CDPM2) to represent the material behavior. This study produced cross-laminated timber panels at a range of four lamina thickness (5/8, 1, 1 1/8, and 1 1/4 inch) and three bonding pressures (50, 125, 200 psi), producing a total of 12 panels for mechanical testing. The goal of this study is to observe how the thickness and pressure trends affect the mechanical properties of CLT. Tests include flatwise bending, flatwise shear, internal-bond, and delamination. Results showed that bending MOE decreases as the panel thickness increases while bonding pressure had no significance. Tests and models are often based on monolithic armors, however, layered and spaced armors are common in real world applications. Remaining useful life (RUL) prediction is a problem that researchers in the prognostics and health management (PHM) community have been studying for decades. Both physics-based and data-driven methods have been investigated, and in recent years, deep learning has gained significant attention. When sufficiently large and diverse datasets are available, deep neural networks can achieve state-of-the-art performance in RUL prediction for a variety of systems. High hardness steels (HHS) are vulnerable to hydrogen embrittlement, which can lead to rapid degradation of mechanical properties. Improved resistance to hydrogen embrittlement would be beneficial to many industries including construction, automotive, and military. A comparison study was performed to assess the hydrogen susceptibility of select commercially available and in-house designed HHS alloys. Slow strain rate tensile tests, performed with specimens charged with various levels of hydrogen, provided a macroscopic view of the onset of hydrogen embrittlement. The Rotating Detonation Engine (RDE) has gained increasing attention in recent years for its potential advantages over typical deflagration combustion. A Micro-RDE design with an outer diameter of 28mm operating on Nitrous Oxide and Ethylene was recently developed, which stretched the limits of small-scale detonation engines. The testing on this rig has shown a stable one wave mode detonation with frequencies reaching 16.8 kHz. This research focuses on developing metal matrix composite reinforced by polymer-derived ceramic fibers. The force-spun AMT nanofibers were heat treated into tungsten carbide and then mixed with aluminum powder using cold pressing and sintering to produce the composites. The basic mechanical properties of this composite are investigated by density measurement, hardness and compression testing. SEM, EDS and XRD were used to test the morphology of tungsten carbide fibers. EDS identified a near-perfect W-C ratio in tungsten carbide fibers 1000 °C. Tungsten Carbide fibers were crushed using ball milling technique. Tungsten carbide nanofibers powder had a variety of particle size. Results …
