Purpose: Cytoskeletal protein ensembles exhibit emergent mechanics where behavior in teams is not necessarily the sum of the components’ single molecule properties. In addition, filaments may act as force sensors that distribute feedback and influence motor protein behavior. To understand the design principles of such emergent mechanics, we developed an approach utilizing QCM-D to measure how actomyosin bundles respond mechanically to environmental variables that alter constituent myosin II motor behavior.
Methods: QCM-D is used for the first time to probe alterations in actin-myosin bundle viscoelasticity due to changes in skeletal myosin II concentration and motor nucleotide state. Actomyosin bundles were …

Alzheimer’s disease (AD) is a progressive neurodegenerative disease with the clinical symptoms of gradual cognitive decline and memory loss. Biologically, abnormal accumulation of Aβ (Aβ) plaque and tau tangles in the brain are the core pathological hallmarks in AD. Although inflammatory response is not specific to AD, emerging evidence suggests that the disrupted innate immune system within the central nervous system, particularly microglial activation, might act as the bridge linking Aβ aggregation to pathological phosphorylation and aggregation of tau. Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. Genetics plays a critical role in understanding the molecular mechanisms underlying neurodegenerative disorders and pathogen evolution in infectious diseases. For example, identifying genetic variants associated with a disease phenotype uncovers functional pathways that could lead to potential drug targets and therapeutic interventions. In addition, tracking the genetic evolution of pathogens enables early detection and warning of infectious disease outbreaks. In both applications, given the large amount of genetic data, advanced computational methods, including longitudinal and multivariate models, could significantly boost the statistical power and capture interrelationships among traits, environmental factors and genetic influences. Machine learning has emerged as a key technology for enabling advanced computer vision systems. These systems now permeate many industries, and have enhanced traditional processes through autonomous interpretation of visual data. In the field of orthopaedics, the increasing prevalence of imaging highlights a need for similar tools. In many cases, however, direct translation of available frameworks fails to resolve the complex problems currently facing this field. Abstract: Studies comparing student outcomes for online vs. in-person classes have reported mixed results, though with a majority finding that lower-performing students, on average, fare worse in online classes, attributed to the lack of built-in structure provided by in-person instruction. The online/in-person outcome disparity was normative for non-major geology classes at the University of Mississippi prior to COVID-19, but the difference disappeared in the years after 2020. Previously distinct trendlines of GPA-based predictions of earned-grade for online and in-person classes merged. Of particular concern, outcomes for in-person classes declined to match pre-COVID-19 online expectations, with lower-GPA students disproportionally impacted. Objective …

The Microphysiological Systems Laboratory aims to develop colorectal cancer tumor models under a hypoxic environment to assess model response to pharmaceutical compounds in vitro. To perform relevant studies, researchers have attempted to use different hypoxic inducing strategies such as a nitrogen pod and hypoxic incubator to recreate in vivo physiological responses to hypoxia. However, studies would be interrupted due to incubator functionality failure. The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. Electrostatic artificial muscles benefit from a microfluidic approach due to the non-linearity of electrostatic attraction creating disproportionate benefits when miniaturized. When designed …

The reconstruction of craniomaxillofacial bone defects remains clinically challenging. To date, autogenous grafts are considered the gold standard but present critical drawbacks. These shortcomings have driven recent research on craniomaxillofacial bone reconstruction to focus on synthetic grafts with distinct materials and fabrication techniques. Among the various fabrication methods, additive manufacturing (AM) has shown significant clinical potential. AM technologies build three-dimensional (3D) objects with personalized geometry customizable from a computer-aided design. These layer-by-layer 3D biomaterial structures can support bone formation by guiding cell migration/proliferation, osteogenesis, and angiogenesis. In this work, the synthesis and the scheme for obtaining an oligomer based on epiiodhydrin with diphenylguanidine were studied, the proportions of epiiodhydrin in the initial mixture, the yield of oligomers, and the reduced viscosity were studied. The results of spectroscopic and elemental analyzes are obtained, which show that changes in the ratio of monomers have almost no effect on the iodine content in the oligomer. This is apparently due to the fact that epiiodhydrin is much more active in reactions than amino compounds. Chitosan was synthesized from Apis Mellifera dead bees by traditional and cryogenic methods, and the degree of deacetylation, viscosity, and molecular mass of the synthesized sample was studied. Chitosan was synthesized by deacetylation of chitin under cryogenic conditions. NaOH concentration 40%, temperature 90 ± 3 °C, time 120 min. The degree of deacetylation, and molecular mass was determined using an Ubbelode viscometer. DDA was determined by conductometric titration under standard conditions. The combination of outsole, midsole and insole layers of new shoes provide proper cushioning to the foot during the daily activities of life. Subsequently, the wear-based degeneration or damage to one or more of these sole layers can reduce the cushioning effect to the plantar surface of the foot. This reduction of cushioning effect can impact the distribution of stress concentration regions along the foot plantar surfaces and lead to foot pathologies. Flow diverter devices are small stents used to divert blood flow away from aneurysms in the brain, stagnating flow and inducing intra-aneurysmal thrombosis which in time will prevent aneurysm rupture. Current devices are formed from thin (∼25 μm) wires which will remain in place long after the aneurysm has been mitigated. As their continued presence could lead to secondary complications, an absorbable flow diverter which dissolves into the body after aneurysm occlusion is desirable. This dissertation aimed to (1) quantify morphological variations in the pediatric hip joint and (2) evaluate the sensitivity of an infant musculoskeletal model (MSM) to these variations, considering hip joint center estimation errors. A shape statistical model (SSM) of decedent infant femurs from the Ortolani collection was created using ShapeWorks, capturing key morphological features, such as variations in the femoral neck-shaft and anteversion angles. Seven synthetic femurs were generated from the SSM to create SSM-informed MSMs, which were systematically evaluated through kinematics and kinetics analyses in OpenSim. Understanding and predicting the hydrodynamic interactions of micron-scale droplets is crucial in a wide range of industrial and real-life applications, including microfluidics, pharmaceutics, drug delivery, food science, and enhanced oil recovery. These multi-phase and multi-scale phenomena are further complicated by the presence of core droplets of an immiscible fluid within shell droplets, known as compound droplets. During the COVID-19 global pandemic, a shortage of personal protective equipment (PPE) occurred. Manufacturers of these critical products were unable to keep up with the growing demands as the situation evolved. Most face shields, a common type of PPE, include a headband component to provide the appropriate fit for the user and curvature for the clear face shield attachment. This dissertation explores the use of cold atmospheric pressure plasma (CAP) technology in eradicating bacterial biofilms from tissue surfaces, using Ps. fluorescens on chicken tissue to model human chronic wounds. It introduces a novel image-guided technique for precise biofilm removal, employing trypan blue staining coupled with advanced computer vision analysis, enhancing the CAP treatment's precision and efficacy. The study demonstrates the CAP device's ability to effectively disrupt biofilms through a non-thermal mechanism, based on its electrical characteristics and operational parameters. Total shoulder arthroplasty (TSA) is often considered a surgery of last resort for patients with debilitating pathologies of the glenohumeral joint such as osteoarthritis, humeral head fracture, and advancing rotator cuff tears. TSA replaces the articulation between the humeral head and the glenoid fossa, with the goals of relieving pain and restoring function. This paper presents the development of First Respire and their design of a powered air-purifying respirator (PAPR) specifically curated for wildland firefighter while simultaneously addressing the critical absence of respiratory protection for wildland firefighters operating in the challenging wildland-urban interface (WUI) environments. An infant's musculoskeletal and motor development is largely affected by their environment. Understanding how different mechanical environments affect an infant's movements and muscle use is necessary to inform the juvenile products industry and reduce incidents involving inclined nursery products each year.
