Molecular Characterization Of Wetland Soil Bacterial Communities In Constructed Mesocosms, Elisabeth M. Leon

16S rRNA gene analysis was performed on soil samples from a subsurface flow mesocosm experiment, mimicking a constructed wetland. Three typical wetland species of plant were used in nine of the columns and three columns were unplanted controls. Analyses showed that plant presence, plant species, and depth did not have a significant effect on microbial community composition. However, richness analyses illustrated that plant presence had a positive effect on richness, plant species analyses indicated that there was evidence that plant species affected richness in a significant manner, and lastly richness was negatively affected in the bottom depth for the control …

Bifidobacteria have been recommended as potential indicators of human fecal pollution in surface waters even though very little is known about their presence in nonhuman fecal sources. The objective of this research was to shed light on the occurrence and molecular diversity of this fecal indicator group in different animals and environmental waters. Genus- and species-specific 16S rRNA gene PCR assays were used to study the presence of bifidobacteria among 269 fecal DNA extracts from 32 different animals. Twelve samples from three wastewater treatment plants and 34 water samples from two fecally impacted watersheds were also tested. The species-specific assays …

Global change stressors, including climate change and variability and changes in land use, are major drivers of ecosystem alterations. Invasive species, which are non-native species that cause environmental or economic damages or human-health impacts, also contribute to ecosystem changes. The interactions between stressors and invasive species, although not well understood, may exacerbate the impacts of climate change on ecosystems, and likewise, climate change may enable further invasions. This report reviews available literature on climate-change effects on aquatic invasive species (AIS) and examines state-level AIS management activities. The objective of this paper is to examine the changes in training needs arising from the increase of off-site construction in the Republic of Ireland. The methodology employed was a case study using a participant observation research technique. For a period of three weeks, the researcher observed the construction of residential housing, constructed using off-site techniques. From the practices observed and from interviews with the operatives on-site, an account of the differences in skills required between those employed in traditional construction and off-site construction was compiled. Binders manufactured using a blend of gypsum, ground granulated blast furnace slag and Portland cements are technically viable and possess considerable environmental and economic advantages when compared to binders manufactured using Portland cement alone. As such, the evaluation of binders made from these materials offers a promising research focus in the quest to produce technically sound, environmental and economical binders for specialist uses as an alternative to traditional concrete binders of higher carbon footprint. Highway maintenance activities usually require lane closures and disrupt traffic operations. Because of budget constraints, project deadlines, and the resulting traffic impact, the objective of this dissertation is to improve the efficiencies of traffic operation and maintenance work, and minimize the total project cost (i.e., agency cost and road user cost) by optimizing work zone schedules.

This dissertation focuses on the maintenance projects on multiple-lane highways. The objective total cost function is formulated while considering a discrete maintenance time-cost function and time-dependent traffic diversions. Hobble Creek is one of several inflowing streams and rivers into Utah Lake, Utah, USA. Historically, June sucker (Chasmistes liorus), a federally listed endemic fish, spawned up all the major inflowing streams and rivers but is now limited to just the Provo River. The State of Utah has recently proposed restoring the lower reaches of Hobble Creek for additional spawning and rearing needs. This restoration effort will likely involve removal of migration barriers, re-aligning the stream, and removing existing levees that prevent floodplain access. Steel slag is an industrial byproduct obtained from the steel manufacturing industry. It is produced in large quantities during the steel-making operations which utilize Electric Arc Furnaces (EAF). Steel slag can also be produced by smelting iron ore in the Basic Oxygen Furnace (BOF). Steel slag can be used in the construction industry as aggregates in concrete by replacing natural aggregates. Natural aggregates are becoming increasingly scarce and their production and shipment is becoming more difficult. Current methodologies of origin investigation have yet to include the structural deformations seen in steel buildings as a viable indicator of the area of origin of a given fire. As many steel structures are of relatively large size, it is often difficult to determine the area of origin using the typical dig and sift methods advocated in NFPA 921, especially if the extent of the fire was large and there were no witnesses as to the origin of the fire. The asphalt pavement analyzer (APA) has been widely used to evaluate hot-mix asphalt (HMA) rutting potential in mix design and quality control – quality assurance (QC-QA) applications, because the APA testing and its data analyses are relatively simple, rapid, and easy.
