Circular RNA (circRNA) has emerged as a promising therapeutic modality due to its enhanced stability and resistance to exonuclease degradation compared with linear mRNA. However, its stability and functionality depend strongly on achieving high purity, and current methods such as size-exclusion chromatography suffer from peak overlap, low yields, and poor scalability. This dissertation addresses these limitations by developing ultrafiltration-based strategies to purify circRNA directly from in vitro transcription (IVT) and self-splicing (circularization) reactions. Wettability is a crucial surface property influencing various phenomena, including heat transfer, cell adhesion, and corrosion. Engineering devices can achieve superior performance by precisely manipulating surface wettability. As a result, extensive research has focused on developing surfaces that exhibit either extreme water attraction (superhydrophilic) or strong water repellency (superhydrophobic). Tailoring surface wettability paves the way for numerous applications, such as drag reduction in marine vessels, controlled drug delivery, efficient water collection, advanced liquid transport systems, oil-water separation, anti-corrosion coatings, friction reduction, and self-cleaning materials. In an attempt to reduce reliance on petroleum-based polymers, lignin – an abundant biopolymer – has gained popularity as a sustainable alternative. However, research on lignin-containing soft composites – i.e., hydrogel (bio)composites – has largely been heuristic, providing limited insight into the fundamental structure-processing-property relationships that govern performance improvements in this emerging class of ‘green’ soft composites. As such, this dissertation presents a systematic investigation of how lignin molecular weight (MW), composite composition, and the concentrations of crosslinker and accelerator impact the mechanical and transport characteristics of soft composites composed of lignin, poly(N-isopropylacrylamide) (PNIPAm), and poly(vinyl alcohol) (PVA).

The current benchmark membrane for vanadium redox flow batteries (VRFBs), Nafion™, accounts for up to 40% of the cost of the battery and suffers from poor ion selectivity, leading to diminished battery performance. Consequently, alternative ionomer membranes, such as sulfonated poly(ether ether ketone) (SPEEK), have recently garnered attention due to their lower cost and similar performance properties to Nafion. However, like Nafion, SPEEK also suffers from poor ion selectivity. As the development of energy storage technologies for renewable energy sources continues to garner attention, vanadium redox flow batteries (VRFBs) have emerged as promising source for grid-scale energy storage. A major component of the VRFB is the ion exchange membrane (IEM), as it serves to physically separate the two liquid electrolytes and also directly impacts the battery’s capacity, efficiency, and cost. The use of lignin in the fabrication of soft composites has become an emerging area of research in polymer science and polymer chemistry. These lignin-based materials present numerous benefits, notably, a reduction in the use of petroleum-based precursor, improved structural benefits to otherwise soft host polymers, as well as the inherent antimicrobial and antioxidant properties of lignin, making it suitable for biomaterials. Understanding the fate and transport of T. gondii through the soil and groundwater is vital in determining the risk it poses to water resources and human health. The objective of this research was to test the feasibility of using forward osmosis (FO) with polyelectrolyte draw solutions to recover water from bioreactor mixed liquors. When combined with an Anaerobic Osmotic Membrane Bioreactor (AnOMBR), such a system could process fecal and food waste from astronauts aboard the International Space Station (ISS) and reclaim important nutrients and water. This project focused on measuring the obtainable water recovery rates from bioreactor effluent, and the identifying challenges associated with the operation.

AnOMBRs feature several advantages over aerobic bioreactors, and non-osmotic anaerobic membrane bioreactors (AnMBR). Anaerobic bioreactors avoid the significant energy costs of …
