Circular RNA (circRNA) has emerged as a promising therapeutic modality due to its enhanced stability and resistance to exonuclease degradation compared with linear mRNA. However, its stability and functionality depend strongly on achieving high purity, and current methods such as size-exclusion chromatography suffer from peak overlap, low yields, and poor scalability. This dissertation addresses these limitations by developing ultrafiltration-based strategies to purify circRNA directly from in vitro transcription (IVT) and self-splicing (circularization) reactions. This dissertation explores the combination of two sophisticated techniques for addressing computational fluid dynamics: the discrete velocity Boltzmann equation (DVBE) and the localized collocation meshless model with upwinding (U-LCMM). The DVBE is a high-level model that describes the foundations of transport phenomena by addressing the microscale motions of particles themselves and the effect of their aggregate behaviors on continuum principles. This equation integrates multiple scales of phenomena; while it can be used for fluid flow at Navier-Stokes scales, it can also resolve fine features that can only be described at the molecular level. Electrocatalytic carbon dioxide (CO2) reduction to C2+ products holds promise for low carbon intensity fuels, chemicals, and food; however, durability challenges, particularly bicarbonate (HCO3-)and carbonate (CO32-) salt precipitation, hinder their commercialization. This report considers water (H2O) and ion transport properties of conventional anion exchange membranes (AEMs) in zero-gap membrane electrode assembly (MEA) configurations using Cu electrocatalysts. In an attempt to reduce reliance on petroleum-based polymers, lignin – an abundant biopolymer – has gained popularity as a sustainable alternative. However, research on lignin-containing soft composites – i.e., hydrogel (bio)composites – has largely been heuristic, providing limited insight into the fundamental structure-processing-property relationships that govern performance improvements in this emerging class of ‘green’ soft composites. As such, this dissertation presents a systematic investigation of how lignin molecular weight (MW), composite composition, and the concentrations of crosslinker and accelerator impact the mechanical and transport characteristics of soft composites composed of lignin, poly(N-isopropylacrylamide) (PNIPAm), and poly(vinyl alcohol) (PVA).

Gypsum, a naturally occurring mineral salt, is frequently encountered in industrial processes such as seawater desalination and power plant cooling. Over the years, significant effort has been devoted to controlling the crystallization of gypsum through various chemical approaches, including acidification, chelating agents, and antiscalant polymers. Nevertheless, the cost associated with chemical treatments may be reduced by incorporating physical operational strategies to complement chemical methods in regulating gypsum crystallization.

In this work, palladium nanoparticles (PdNPs)/p1,5-DAN/ carbon paste electrode (CPE) and p1,5-DAN/CPE sensors have been developed for determination of hydrogen peroxide. Both sensors showed a highly sensitive and selective electrochemical behaviour, which were derived from a large specific area of poly 1,5 DAN and super excellent electroconductibility of PdNPs. PdNPs/p1,5-DAN/CPE exhibited excellent performance over p1,5-DAN/CPE. Thus, it was used for detecting hydrogen peroxide (H2O2) with linear ranges of 0.1 to 250 μM and 0.2 to 300 μM as well as detection limits (S/N= 3) of 1.0 and 5.0 nM for square wave voltammetry (SWV) and cyclic voltammetry (C.V) techniques, respectively. …

The current benchmark membrane for vanadium redox flow batteries (VRFBs), Nafion™, accounts for up to 40% of the cost of the battery and suffers from poor ion selectivity, leading to diminished battery performance. Consequently, alternative ionomer membranes, such as sulfonated poly(ether ether ketone) (SPEEK), have recently garnered attention due to their lower cost and similar performance properties to Nafion. However, like Nafion, SPEEK also suffers from poor ion selectivity. Anaerobic digestate (AD) of various centralized wastewater facilities contains substantial amounts of ammonium (NH4+) (300-800 mg/L), potassium (K+) (13-120 mg/L), and phosphorus (P) (30-100 mg/L)- based nutrients. A downside of plastic is that the most common type is a product of oil and oil is not only a limited resource but also a climate-damaging resource. Polylactic acid (PLA) is a bio-based, biodegradable plastic. However, the process of converting biomass to polylactic acid polymer has the largest environmental impact of the PLA production process, so alternative methods of conversion are needed (Moretti et al., 2021). Polymer-enhanced ultrafiltration has gained increasing attention as a selective separation process to remove small solutes, such as metal ions, from water or wastewater. Hexavalent chromium Cr(VI) is one of the metal ions in wastewater that has received considerable attention because of its toxicity. Chitosan was used in this study as the polymer to bind Cr(VI) to enhance its removal by ultrafiltration. The effects of chitosan-to-Cr(VI) ratio on Cr(VI) rejection and permeate flux were investigated. In addition, the fouling tendency was evaluated. The objective of this research was to test the feasibility of using forward osmosis (FO) with polyelectrolyte draw solutions to recover water from bioreactor mixed liquors. When combined with an Anaerobic Osmotic Membrane Bioreactor (AnOMBR), such a system could process fecal and food waste from astronauts aboard the International Space Station (ISS) and reclaim important nutrients and water. This project focused on measuring the obtainable water recovery rates from bioreactor effluent, and the identifying challenges associated with the operation.

SDDSs aim to achieve concentrations in solutions greater than the solubility of the lowest energy crystalline form. Though membrane separation systems appeared superior to the conventional counterparts for water and wastewater treatment, wider applications of membrane systems have been limited by some factors, most notably is membrane fouling. Capillarity is often exploited in self-cleaning, drag reducing and fluid absorption/storage (sanitary products) purposes just to name a few. Formulating the underlying physics of capillarity helps future design and development of optimized structures. This work reports on developing computational models to quantify the capillary pressure and capillary forces on the fibrous surfaces. To this end, the current study utilizes a novel mass-spring-damper approach to incorporate the mechanical properties of the fibers in generating virtual fibrous structures that can best represent fibrous membranes. Future nanofiltration (NF) membranes used for selective separations of ions and small organic molecules must maintain performance in environments where high concentrations of total dissolved solvents or foulants are present. These challenges can be addressed through the development of composite membranes, as well as the engineering of enhanced surface properties and operating conditions for existing commercial membranes.
In this work, ion transport through commercial thin film composite (TFC) polyamide NF membranes were studied in both lab-prepared salt solutions and industrial wastewater. Among the next generation materials being investigated for membrane development, partially reduced Graphene Oxide (rGO) has received increasing attention from the membrane community. rGO-based nanofiltration membranes have shown promising results in applications such as partial desalination, organic contaminant removal, gas-phase separations, and separations from solvent media. rGO offers a unique platform compared to common polymeric membranes since it can be used for separation applications in both aqueous and organic solvent media. An rGO-based platform could also be utilized to synthesize reactive membranes, giving rGO membranes the additional capability of reactively removing organic contaminants.
