Circular RNA (circRNA) has emerged as a promising therapeutic modality due to its enhanced stability and resistance to exonuclease degradation compared with linear mRNA. However, its stability and functionality depend strongly on achieving high purity, and current methods such as size-exclusion chromatography suffer from peak overlap, low yields, and poor scalability. This dissertation addresses these limitations by developing ultrafiltration-based strategies to purify circRNA directly from in vitro transcription (IVT) and self-splicing (circularization) reactions. Adsorbent components in face masks are crucial for protecting individuals exposed to dangerous situations. In this study, one porous TiO2 layer was deposited on zeolite 13X particles by coating hybrid organic/inorganic titanium alkoxide film via molecular layer deposition (MLD), followed by heat treatment to remove organic components in the MLD film. Various concentrations of Ag2O nanoparticles were impregnated on the TiO2-coated 13X particles, offering a wide range of antibacterial, antifungal, and antiviral characteristics. This study investigates the hydrodynamics of particles and drops driven by forces generated by an asymmetrical physical and chemical surrounding environment. In the first part, a novel colloidal motor design driven by surface tension forces is proposed, utilizing an active Janus particle encapsulated in an immiscible liquid drop to form a compound drop/particle. Marangoni forces induced by asymmetric solute adsorption at the liquid-liquid interface of the drop propels the compound system. Clinical studies have shown that patients undergoing renal replacement therapy are more susceptible to developing hemolysis, or the rupturing of red blood cells. Rapid hemolysis can cause symptoms such as anorexia, vomiting, and even death in severe cases. The aim of this study is to identify how shear stress within a hollow fiber membrane impacts the level of hemolysis that occurs. This allows for the optimization of the ultrafiltration membranes that are typically used for hemofiltration treatments. The use of lignin in the fabrication of soft composites has become an emerging area of research in polymer science and polymer chemistry. These lignin-based materials present numerous benefits, notably, a reduction in the use of petroleum-based precursor, improved structural benefits to otherwise soft host polymers, as well as the inherent antimicrobial and antioxidant properties of lignin, making it suitable for biomaterials. Wastewater sludge is an overlooked source of fat, oil, and grease (FOG) that could be converted into biodiesel. The United States produces about 8 million tons of sludge per year. The disposal cost for this amount of sludge is about 2 billion US dollars. The widespread availability and low cost of sludge compared to other biodiesel raw materials make it an economical choice for a renewable fuel. With the looming dominance of poorly water-soluble chemical entities within the pharmaceutical pipeline, the pharmaceutical industry has leaned on the use of supersaturating drug delivery systems (SDDSs) to achieve efficacious concentrations within the gastrointestinal fluids. SDDSs aim to achieve concentrations in solutions greater than the solubility of the lowest energy crystalline form. However, the generation of supersaturated solutions of active pharmaceutical ingredients (APIs) creates a strong crystallization potential, which is undesirable.

The generation of biofuels from lignocellulosic biomass involves innovative process technology that is being investigated worldwide. Enzymatic hydrolysis is a major step in the contemporary process of the generation of biofuels. Guided by pore size distribution measured using NMR cryoporometry, we developed pore-enzyme diffusion and adsorption models at the particle level coupled with a kinetic model for cellulose, cellobiose, and glucose production at flask level. The structural, transport, and thermodynamic properties related to cellulose dissolution by tetrabutylphosphonium chloride (TBPCl) and tetrabutylphosphonium hydroxide (TBPH)-water mixtures have been calculated via molecular dynamics simulations. For both ionic liquid (IL)-water solutions, water veins begin to form between the TBPs interlocking arms at 80 mol % water, opening a pathway for the diffusion of the anions, cations, and water. The water veins allow for a diffusion regime shift in the concentration region from 80 to 92.5 mol % water, providing a higher probability of solvent interaction with the dissolving cellulose strand. Wall shear stress (WSS) is commonly accepted as the primary influence affecting characteristics of anchored endothelial cells when subjected to fluid flow. Orbital shakers are commonly used to study cellular responses due to their ease of use, ability to run several experiments simultaneously, and since they exert physiologically relevant oscillatory shear. The purpose of this project was to model the size of calcium alginate microbeads produced by extruding a solution of 1.5 wt.% alginate from a syringe and needle with air shear generated from a pneumatic line flowing in an annular tube around the extrusion needle. The air shear would create microdroplets that would fall into a solution of 2 wt.% calcium chloride in water, crosslinking the droplets into beads. These microbeads can be used in drug dispersion applications or in encapsulation of cells throughout the body due to their biocompatibility. The goal was to create uniform beads that ranged from …
