Strategies for mitigating the impacts of cyberattacks on control systems using a control-oriented perspective have become of greater interest in recent years. Our group has contributed to this trend by developing several methods for detecting cyberattacks on process sensors, actuators, or both sensors and actuators simultaneously using an advanced optimization-based control strategy known as Lyapunov-based economic model predictive control (LEMPC). Though dynamic operation of chemical processes has been extensively explored theoretically in contexts such as economic model predictive control or even considering the potential for cyberattacks on control systems creating non-standard operating policies, important practical questions remain regarding dynamic operation. In this work, we look at two of these with particular relevance to process safety: (1) evaluating dynamic operating policies with respect to process equipment fidelity and (2) evaluating procedures for determining the parameters of an advanced control law that can promote both dynamic operation as well as safety if appropriately designed. Typical control designs in the process systems engineering literature have assumed that the primary sensing methodologies are traditional instruments such as thermocouples. Dig- italization is changing the landscape for manufacturing, and data-based sensing modalities (e.g., image-based sensing) are becoming of greater interest for plant control. These considerations require novel test/evaluation solutions. For example, process systems engineering researchers may wish to test image-based sensors in simulation. In this work, we provide preliminary thoughts on how image-based technologies might be evaluated via simulation for process systems.

There were two parts of the assignment, each with slightly different end goals. The production of styrene from ethylbenzene in Unit 500 is designed to start up in 2024 and operate for 12 years after startup. The engineering team was tasked with designing the process, creating an economic model, and optimizing the net present value (NPV). The process was simulated in AVEVA PRO/II Simulation for the design process and to estimate the size of equipment. Parametric and topological optimization was performed subsequently on the unit operations in the process. The NPV was improved by $423M from a base case of -$919M to an optimized case of -$496M. One of the limitations of conventional monitoring tools in crystallization is the inability to deal with high solids concentration. Image-based monitoring, in which RGB images of the solution are captured and analyzed by an object detection software, has been a promising alternative. Virus filters are single-use devices that use a size-based separation process. In virus filters, contaminating virus particles are retained while the therapeutic molecules pass through the membrane pores. Virus filters are an essential component of the overall virus clearance strategy. Sections 1 and 2 of this dissertation provide an introduction and extensive review of monoclonal antibody (mAb) process development, where virus filtration is pivotal. In section 3, prefiltration studies were performed with an industrially relevant IgG1 type mAb using adsorptive and size-exclusion-based prefilters with different mechanisms of action. The controllers for a cyber-physical system may be impacted by sensor measurement cyberattacks, actuator signal cyberattacks, or both types of attacks. Prior work in our group has developed a theory for handling cyberattacks on process sensors. However, sensor and actuator cyberattacks have a different character from one another. Economic model predictive control (EMPC) is a flexible control design strategy that can be modified to achieve many operating goals while also ensuring safe operation (e.g., by adding Lyapunov-based stability constraints to form Lyapunov-based EMPC, or LEMPC). Microcystin-LR (MC-LR) is a toxin produced by cyanobacteria that can bloom in freshwater supplies. This study describes a new strategy for remediation of MC-LR that combines linearization of the toxin using microcystinase A, MlrA, enzyme with rejection of linearized byproducts using membrane filtration. The MlrA enzyme was expressed in Escherichia coli (E. coli) and purified via a His-tag with 95% purity. Additionally, composite membranes made of 95% polysulfone and 5% sulfonated polyether ether ketone (SPEEK) were fabricated and used to filter a solution containing cyclic and linearized MC-LR. The most common gas sensors are based on chemically induced changes in electrical resistivity and necessarily involve making imperfect electrical contacts to the sensing materials, which introduce errors into the measurements. We leverage thermal- and chemical-induced changes in microwave propagation characteristics (i.e., S-parameters) to compare ZnO and surface-anchored metal-organic-framework (HKUST-1 MOF) thin films as sensing materials for detecting ethanol vapor, a typical volatile organic compound (VOC), at low temperatures. We show that the microwave propagation technique can detect ethanol at relatively low temperatures (<100 >°C), and afford new mechanistic insights that are inaccessible with the traditional dc-resistance-based measurements.
