Virus filters are single-use devices that use a size-based separation process. In virus filters, contaminating virus particles are retained while the therapeutic molecules pass through the membrane pores. Virus filters are an essential component of the overall virus clearance strategy. Sections 1 and 2 of this dissertation provide an introduction and extensive review of monoclonal antibody (mAb) process development, where virus filtration is pivotal. In section 3, prefiltration studies were performed with an industrially relevant IgG1 type mAb using adsorptive and size-exclusion-based prefilters with different mechanisms of action. Microcystin-LR (MC-LR) is a toxin produced by cyanobacteria that can bloom in freshwater supplies. This study describes a new strategy for remediation of MC-LR that combines linearization of the toxin using microcystinase A, MlrA, enzyme with rejection of linearized byproducts using membrane filtration. The MlrA enzyme was expressed in Escherichia coli (E. coli) and purified via a His-tag with 95% purity. Additionally, composite membranes made of 95% polysulfone and 5% sulfonated polyether ether ketone (SPEEK) were fabricated and used to filter a solution containing cyclic and linearized MC-LR. Plastic waste generation is increasing at an unsustainable rate while recycling solutions remain stagnant. As a chemical means of recycling, mixed plastic waste pyrolysis can generate synthetic oil appropriate for use as fuel in power generation from plastic waste that otherwise accumulates in landfills. With the scaling of a commercial plastic pyrolysis process in Northwest Arkansas (NWA) modeled after an operational sawdust pyrolysis unit in Huntsville, Arkansas, economic analysis resulted in 26.3% internal rate of return. Therefore, construction of a commercial mixed plastic-to-fuel pyrolysis plant is economically justified and should be pursued. To effectively implement the proposed design, NWA must …

Enzyme-linked immunosorbent assay (ELISA) microarray performance is limited by low assay sensitivity and dynamic range. Increasing the surface area for reagent binding can help to improve performance, but standard techniques such as roughening the surface or adding a polymer coating lead to increased non-specific fluorescence and do not have reproducibly improved performance. Another approach to increase surface area is adding a microsphere coating on the surface. Poly-N-substituted glycine (peptoid) microspheres are ideal for this application due to low immunogenicity, protease-resistance, and biocompatibility. The purpose of this project was to model and design a plant to produce 75,000 tons per year of acrylic acid. Using the design in section B.9 of Richard Turton’s Analysis, Synthesis, and Design of Chemical Processes as a starting point, a plant was designed based around the partial oxidation of propylene to acrylic acid. The final plant design produces about 86,000 tons per year of acrylic acid. Using an interest rate of 10%, the plant has a discounted cash flow rate of return of 32% over 2 years of startup and 10 years of operation. Attached is an executive …

Along-wind dispersion, or stretching of the cloud in the direction of the wind, plays an important role in the concentration and modeling of contaminant releases in the atmosphere. Theoretical and empirical derivations were compared for appropriate parameterization of along-wind dispersion. Available field data were analyzed to evaluate and improve previous parameterizations of the along-wind dispersion coefficient, σ_x. An experimental test program was developed and executed in an ultra-low speed wind tunnel at the Chemical Hazards Research Center to determine the effects of the time distribution coefficient on true finite-duration releases from an original area source. Single celled eukaryotic organisms are advantageous in the production of proteins for use in biopharmaceuticals due to their ease of cultivation and manipulation. Because of high cell density during fermentation and an ability to allow post-translational protein modifications, yeast is particularly well-suited. To maximize the specific formation rate of the desired proteins, an optimum specific growth rate of the yeast should be found and maintained. It is the intent of this work to develop a process control apparatus and series of experiments that will help students visualize the PID (Proportional-Integral-Derivative) control of a process and enhance their understanding of the subject. The apparatus is a computer-controlled PID mixing system that responds quickly to set point changes and process disturbances which are directly observable. The system can easily be simulated with a transfer function model in Matlab's Simulink, so that the controller can be optimized for the desired system response. The scientific and industrial demand for controllable thin gold (Au) film and Au nanostructures is increasing in many fields including opto-electronics, photovoltaics, MEMS devices, diagnostics, bio-molecular sensors, spectro-/microscopic surfaces and probes. In this study, a novel continuous flow electroless (CF-EL) Au plating method is developed to fabricate uniform Au thin films in ambient condition. The enhanced local mass transfer rate and continuous deposition resulting from CF-EL plating improved physical uniformity of deposited Au films and thermally transformed nanoparticles (NPs). High value specialty chemicals are usually obtained from natural products by extracting with generally regarded as safe (GRAS) solvents. Because organic solvents are quite often used, high operating and disposal costs often occur. When compared to traditional solvents, water can be viewed as an interesting alternative because of its low operating and disposal costs. Milk thistle contains compounds (taxifolin, silychristin, silydianin, silybinin A and silybinin B) that display hepatoxic protection properties.
