In an attempt to reduce reliance on petroleum-based polymers, lignin – an abundant biopolymer – has gained popularity as a sustainable alternative. However, research on lignin-containing soft composites – i.e., hydrogel (bio)composites – has largely been heuristic, providing limited insight into the fundamental structure-processing-property relationships that govern performance improvements in this emerging class of ‘green’ soft composites. As such, this dissertation presents a systematic investigation of how lignin molecular weight (MW), composite composition, and the concentrations of crosslinker and accelerator impact the mechanical and transport characteristics of soft composites composed of lignin, poly(N-isopropylacrylamide) (PNIPAm), and poly(vinyl alcohol) (PVA).

Smart biomaterials are becoming promising for therapeutic deliveries due to their ability to respond to changes in the environment. Redox-responsive hydrogels are a promising type of smart polymer that can swell and degrade according to cellular oxidative stress levels. Glutathione, abundant in cells, is crucial in redox-responsive biomaterials, serving as a key reactive molecule. This study explored the degradation kinetics of glutathione-responsive poly(β-amino ester) (PBAE) polymers created through a single-step Michael addition. We examined how disulfide content influences the degradation kinetics of these PBAE networks. Widespread distribution of poly- and perfluoroalkyl substances (PFAS) in the environment combined with concerns for their potentially negative health effects has motivated regulators to establish strict standards for their surveillance. The United States Environmental Protection Agency issued a cumulative domestic threshold of 70 ppt for water supplies, and this bar is even lower in some local districts and other countries. Monitoring PFAS consequently requires sensitive analytical equipment to meet regulatory specifications, and liquid chromatography with tandem mass spectroscopy (LC/MS/MS) is the most common technique used to satisfy these requirements. Hydrogels are known to be weak and brittle, due to the amounts of water that they hold. Gelatin-based hydrogels specifically are inexpensive and easily accessible, but often have poor physical properties. The goal of my project was to strengthen gelatin-based hydrogels and improve their mechanical properties by forming these hydrogels in different aqueous solutions containing kosmotropic Hofmeister's ions. Prior research has shown that these ions are able to take water away from gelatin-based gels, which forms a tougher hydrogel. The treatment of ocular diseases poses a significant challenge to drug delivery design, due to the eye’s complex anatomy, with a semi-permeable blood-ocular barrier and associated immune privilege. To address some of the delivery challenges, polycationic dendrimers are explored as vehicle for the treatment of posterior chamber disease. Dendrimers are a class of polymeric nanoparticles that are highly branched, multivalent and monodisperse, whose three-dimensional globular architecture has given them the nickname of “synthetic proteins”. Nano-structured material fabrication using functionalized membranes with polyelectrolytes is a promising research field for water pollution, catalytic and mining applications. These responsive polymers react to external stimuli like temperature, pH, radiation, ionic strength or chemical composition. Such nanomaterials provide novel hybrid properties and can also be self-supported in addition to the membranes.

Polyelectrolytes (as hydrogels) have pH responsiveness. The hydrogel moieties gain or lose protons based on the pH, displaying swelling properties. These responsive materials can be exploited to synthesize metal nanoparticles in situ using their functional groups, or to immobilize other polyelectrolytes and biomolecules.
