The resistive pulse technique characterizes single analytes by detecting fluctuations in the ion current as they pass through a narrow orifice connecting two electrolyte-filled chambers, each containing a single electrode. The amplitude, shape, frequency, and duration of these fluctuations primarily provide information about the analyte’s size, shape, concentration, and net surface charge, respectively. One configuration for resistive pulse measurements is of an electrolyte-filled nanopipette immersed in a bath, where the nanopipette tip acts as the nanoscale orifice. Increasing industrial interest in reducing fossil fuels in virgin polymer production and incorporating bio-based polymeric materials has led researchers to explore the potential of utilizing plant-based polymers such as lignin. Chapter 1 of this thesis presents a comprehensive overview of copolymerization and blending techniques for lignin and synthetic polymers, emphasizing different types of modifications and copolymerization techniques as well as key parameters such as temperature, reaction time, pH level, and solution compositions. Porous polymer membrane filters are widely used in separation and filtration process. Micro- and ultra-filtration membranes are commonly used in biopharmaceutical applications such as filtering viruses and separating proteins from a carrier solution. The formation of these membrane filters via phase inversion is a complex and interconnected process where varying casting conditions can have a wide variety of effects on the final membrane morphol- ogy. The development of materials for tissue regeneration, cell manufacturing, and drug delivery is possible by the manipulation of polymer properties and the use of three different techniques including layer-by-layer, electrospinning, and molding technique.
By applying the layer-by-layer technique over biomaterials, it is possible to develop polymeric multilayers that promote a more favorable environment for cellular functionality, increasing the possibility of improving their acceptance in the area of implantation. Peptoids are peptidomimetic oligomers that predominantly harness similarities to peptides for biomimetic functionality. The incorporation of chiral, aromatic side chains in the peptoid sequence allows for the formation of distinct secondary structures and self-assembly into supramolecular assemblies, including microspheres. Peptoid microspheres can be coated onto substrates for potential use in biosensor technologies, tissue engineering platforms, and drug-delivery systems. They have the potential for use in biomedical applications due to their resistance to proteolytic degradation and low immunogenicity. Electrospun membranes are an attractive alternative to flat sheet membranes as absorbent with numerous advantages like high porosity, large specific surface area and ease of functionalization. This doctoral dissertation focuses on fabricating novel polymeric membrane adsorbents for protein separations and ammonium ion removal. Three distinctly different preparation methods including UV-initiated polymerization, atom transfer radical polymerization, and mixed-matrix formation, have been employed to fabricate the electrospun membranes. Overall, this study aimed to develop electrospun membranes with excellent separation efficiency for application in protein purification and ammonium ion removal.

Carbonaceous materials such as graphene oxide, and activated carbon have a wide range of applications in water and wastewater treatment due to the high surface area, unique structure, and chemical and thermal stability. Continued population growth and the decrease of existing energy platforms demands long-term solutions for development and implementation of scalable plasmonic metamaterials for energy and agricultural applications. Self-assembled nanoantenna into random and ordered arrangements are advanced herein for optical and thermal enhancements in scalable thin film. An analytical approach to estimating the thermal dynamics of random arrangements of nanoantenna resulted in estimates within 30% across a range of geometric parameters, nanoantenna-containing media, and thermal parameters. Ultrafiltration (UF) membranes developed out of a need for protein separation processes. Currently, they are used in a variety of industries ranging from food manufacturing to pharmaceuticals for two main purposes: concentration, separation, and buffer exchange. UF membrane processes in product streams undergo frequent use and like all membrane processes experience a gradual decline in performance due to fouling phenomena both irreversible and reversible. Ultimately, performance declines to a point where the UF membrane needs to be replaced.
