Circular RNA (circRNA) has emerged as a promising therapeutic modality due to its enhanced stability and resistance to exonuclease degradation compared with linear mRNA. However, its stability and functionality depend strongly on achieving high purity, and current methods such as size-exclusion chromatography suffer from peak overlap, low yields, and poor scalability. This dissertation addresses these limitations by developing ultrafiltration-based strategies to purify circRNA directly from in vitro transcription (IVT) and self-splicing (circularization) reactions. Thermoplastic polyurethanes (TPUs) are suited for fused deposition modeling (FDM) of parts that require high levels of flexibility and strength. Predicting the deformation of TPU parts produced using FDM may be difficult, especially under large deformations, as their constitutive models depend on the printing process parameters. The lack of understanding led to the absence of constitutive models for TPU parts produced using FDM. This work aims to identify accurate hyperelastic constitutive models. This dissertation investigates the development and optimization of pathology-responsive polymersomes for simultaneous enhanced magnetic resonance imaging (MRI) and enzyme replacement therapy (ERT) in the treatment of GM1 gangliosidosis (GM1). While therapeutic strategies exist for similar, non-neuropathic lysosomal storage disorders, implementation towards GM1 faces significant challenges due to the blood-brain barrier.

Herein, we developed and characterized a range of low molecular weight hyaluronic acid-b-polylactic acid (HA-PLA) polymersomes (PSs) capable of encapsulating therapeutic enzymes in their hydrophilic core. Classical electrophoretic theory describes the linear response of particles suspended in Newtonian fluids under weak electric fields. When the electric field exceeds this weak limit, a nonlinear response arises due to non-uniform surface conduction over the curved particles. This nonlinear behavior introduces dependencies on fluid and particle properties not considered in the classical theory, offering potential advantages for microfluidic particle manipulation. The waste of polyethylene terephthalate (PET) plastic waste in Indonesia is a pressing concern due to its slow degradation and potential environmental damage. The Namib Desert beetle, Stenocara gracilipes, condenses fog that rolls in off the west coast of Namibia to provide itself with the water it needs for the day. By lifting its abdomen into the air to collect the water vapor, it condenses on hydrophilic bumps that are about 500μm in diameter and are spaced apart about 500-1500μm. Once the water reaches a critical size, the droplet rolls off the hydrophilic bump and onto the flat hydrophobic surface of the elytra. It has been hypothesized that poly(vinylmethylether) (PVME) could be used to replace poly(N-isopropylacrylamide) (pNIPAAM) in various thermoresponsive applications based on literature searches and previous lab experiments. PVME is cheaper and easier to process, and would make pNIPAAM applications more cost effective. In order to verify this, experimental data was gathered on the effects of four anions from the Hofmeister series (SO42-, Cl-, I-, and SCN-) on the lower critical solution temperature (LCST) of pNIPAAM and compared to previous data collected on PVME. Piezoelectric polymer materials such as polyvinylidene fluoride exhibit beneficial properties for use in personal protective equipment (PPE) against viral contaminants and other foreign microparticles. The electrospinning process of this material allows for surface charge to develop over a controlled surface area. Kynar 761 at 10 wt% demonstrates increased surface charge after withstanding a flow of aerated NaCl particles from a TSI 8130 instrument. Utilizing voltage measurements from a Faraday bucket, 18 wt% Kynar 761 and MG-15 exhibit surface charge when undergoing an electrospinning process; however, Kynar MG-15 displays lower charge values than that of Kynar 761. This project addressed burn wound healing through controlled release of the antibacterial drug mafenide encapsulated by a copolyster into 3D printed scaffolds. Scaffolds were printed at 25oC and 0.77-1 bar in a cross-hatch pattern with uniform thickness, distance between parallel layers, and diameter then photo-crosslinked. The number of scaffolds available for testing was limited by the amount of polymer that could be synthesized. The structural, transport, and thermodynamic properties related to cellulose dissolution by tetrabutylphosphonium chloride (TBPCl) and tetrabutylphosphonium hydroxide (TBPH)-water mixtures have been calculated via molecular dynamics simulations. For both ionic liquid (IL)-water solutions, water veins begin to form between the TBPs interlocking arms at 80 mol % water, opening a pathway for the diffusion of the anions, cations, and water. The water veins allow for a diffusion regime shift in the concentration region from 80 to 92.5 mol % water, providing a higher probability of solvent interaction with the dissolving cellulose strand. In the modern forest industry, the need for bio-based, renewable, and environmentally-benign wood preservatives is increasing. Preservatives are used to prevent or limit decay and there has been an increasing interest in developing wood preservatives from renewable materials. To support the need for bio-based, environmentally-friendly preservatives, this work employed kraft lignin as a raw material to produce a novel bio-based wood preservative.

Continued population growth and the decrease of existing energy platforms demands long-term solutions for development and implementation of scalable plasmonic metamaterials for energy and agricultural applications. Self-assembled nanoantenna into random and ordered arrangements are advanced herein for optical and thermal enhancements in scalable thin film. An analytical approach to estimating the thermal dynamics of random arrangements of nanoantenna resulted in estimates within 30% across a range of geometric parameters, nanoantenna-containing media, and thermal parameters. Multimodal thermal dynamics of polymer thin films containing gold nanoparticles (AuNPs) were observed through the natural log of the dimensionless temperature driving force plotted versus time …

The tested hypothesis was that features on polydimethylsiloxane (PDMS) surfaces coated with a poly(N-isopropylacrylamide)/aminopropyltriethoxysilane or pNIPAAM/APTES thin film would accelerate cell detachment than the film coated on a unfeatured surface. This project tested samples with features generated by molds, wrinkling, and sandpaper roughened substrates. Surface feature generation methods were limited to mechanical means, and characterized by microscopy and strain rates. 50/50 mixtures of 1.5 wt.% pNIPAAM/ APTES were used to coat thin films (30-40 nm) on PDMS membranes by spin-coating, and the coated membranes were thermally annealed to chemically graft pNIPAAm/APTES on the membrane and their thermo-responsive property was assessed …
