This dissertation describes developing and implementing computational frameworks for simulating and optimizing purification processes in the biopharmaceutical industry. The framework performs techno-economic analyses to establish value propositions for new process alternatives, especially membrane technologies. Initially, the focus is developing a framework capable of simulating monoclonal antibody (mAb) capture using membrane and resin media in multi-column chromatography (MCC) platforms for continuous manufacturing. Subsequently, the impact of capture MCC is compared against other intensification strategies in established mAb manufacturing facilities. Finally, the framework application expands to simulate the purification of adeno-associated virus (AAV) vectors for gene therapy.

This work presents a theoretical investigation of the use of nanoporous carbon membranes for the separation of nitrogen from natural gas. A mathematical model to predict the performance of the membrane is developed. The model is a combination of the well known dusty gas model, which describes the transfer of multi-components mixture in porous media, together with a surface diffusion model. The model is first validated using the literature results for the separation of hydrogen from hydrocarbons mixture. The model is then applied to the nitrogen-hydrocarbons system. This paper presents an analysis of a filtration technique that uses ultrasound to aid the collection of small particles (tens of microns in diameter) from suspension. In this method, particles are retained within a porous mesh that is subjected to a resonant ultrasonic field, even though the pore size of the mesh is two orders of magnitude greater than the particle diameter. The role of acoustic forces in driving the retention phenomena has previously been studied on a micro-scale, which included modeling and experimental verification of particle motion and trapping near a single element of the mesh.
