Purifying Circular Rna Therapeutics, Karen Guillen-Cuevas Dec 2025

Purifying Circular Rna Therapeutics, Karen Guillen-Cuevas

All Dissertations

Circular RNA (circRNA) has emerged as a promising therapeutic modality due to its enhanced stability and resistance to exonuclease degradation compared with linear mRNA. However, its stability and functionality depend strongly on achieving high purity, and current methods such as size-exclusion chromatography suffer from peak overlap, low yields, and poor scalability. This dissertation addresses these limitations by developing ultrafiltration-based strategies to purify circRNA directly from in vitro transcription (IVT) and self-splicing (circularization) reactions. First, the feasibility of ultrafiltration was demonstrated using polyethersulfone membranes, where sieving coefficients and critical flux values of circRNA, precursor RNA, and nicked RNA were measured to …

Design And Characterization Of Lignin-Based, Thermoresponsive Soft Composites With Improved Mechanical And Transport Properties, Missoury D. Wolff May 2025

Design And Characterization Of Lignin-Based, Thermoresponsive Soft Composites With Improved Mechanical And Transport Properties, Missoury D. Wolff

All Dissertations

In an attempt to reduce reliance on petroleum-based polymers, lignin – an abundant biopolymer – has gained popularity as a sustainable alternative. However, research on lignin-containing soft composites – i.e., hydrogel (bio)composites – has largely been heuristic, providing limited insight into the fundamental structure-processing-property relationships that govern performance improvements in this emerging class of ‘green’ soft composites. As such, this dissertation presents a systematic investigation of how lignin molecular weight (MW), composite composition, and the concentrations of crosslinker and accelerator impact the mechanical and transport characteristics of soft composites composed of lignin, poly(N-isopropylacrylamide) (PNIPAm), and poly(vinyl alcohol) (PVA).

Frameworks For The Techno-Economic Assessment Of Membrane-Based Bioprocessing Platforms, Juan Jose Romero Conde

All Dissertations

This dissertation describes developing and implementing computational frameworks for simulating and optimizing purification processes in the biopharmaceutical industry. The framework performs techno-economic analyses to establish value propositions for new process alternatives, especially membrane technologies. Initially, the focus is developing a framework capable of simulating monoclonal antibody (mAb) capture using membrane and resin media in multi-column chromatography (MCC) platforms for continuous manufacturing. Subsequently, the impact of capture MCC is compared against other intensification strategies in established mAb manufacturing facilities. Finally, the framework application expands to simulate the purification of adeno-associated virus (AAV) vectors for gene therapy.

Chapter 2 details the framework …

Elucidating The Impact Of Casting Solvent On Vanadium Ion Transport In Solution-Cast Ionomer Biocomposites Containing Lignin, Zhongyu Wang May 2024

Elucidating The Impact Of Casting Solvent On Vanadium Ion Transport In Solution-Cast Ionomer Biocomposites Containing Lignin, Zhongyu Wang

All Theses

This paper attempts to investigate the influence of solvent choice to the Nafion-lignin proton exchange membranes that used in the vanadium redox flow batteries. The influence of two different solvents: DMF and DMSO were investigated in this paper. Results show that when using DMSO as solvent, Nafion-lignin membranes' vanadium ion cross over can be greatly reduced in comparing with membranes prepared from DMF. Also, membranes prepared from DMSO also show higher water uptake ability and lower ion exchange capacity than the membranes prepared from DMF. This paper has also investigated the influence of lignin type to the membranes. Results show …

Sulfonated Lignin-Based Ionomer Biocomposite Membranes For Redox Flow Batteries, Aiswarya Mechoor May 2024

Sulfonated Lignin-Based Ionomer Biocomposite Membranes For Redox Flow Batteries, Aiswarya Mechoor

All Theses

The current benchmark membrane for vanadium redox flow batteries (VRFBs), Nafion™, accounts for up to 40% of the cost of the battery and suffers from poor ion selectivity, leading to diminished battery performance. Consequently, alternative ionomer membranes, such as sulfonated poly(ether ether ketone) (SPEEK), have recently garnered attention due to their lower cost and similar performance properties to Nafion. However, like Nafion, SPEEK also suffers from poor ion selectivity. In this study, we attempt to address the issue of poor ion selectivity by developing SPEEK–lignin composite membranes containing fractionated and functionalized lignin. Specifically, two lignin fractions – a low molecular …

Sulfonated Ionomer Composite Membranes For Use In Vanadium Redox Flow Batteries, Xueting Wang May 2024

Sulfonated Ionomer Composite Membranes For Use In Vanadium Redox Flow Batteries, Xueting Wang

All Dissertations

While vanadium redox flow batteries (VRFBs) have garnered significant attention as a promising scalable energy storage technology, the current membrane material used in this technology, Nafion™, suffers from high cross mixing of vanadium ions in the liquid electrolytes, directly impacting battery performance properties such as the voltage and coulombic efficiencies, as well as resulting in a reduction in the overall battery lifetime. The use of ionomer nancomposites has proven to be one promising route of directly addressing the issue of high ion crossover, though the mechanism by which the nanoparticles act to reduce crossover remains in open question. Herein, the …

Amine-Functionalized Lignin-Based Ionomer Nanocomposites For Redox Flow Batteries, Joshua Wilder Dec 2023

Amine-Functionalized Lignin-Based Ionomer Nanocomposites For Redox Flow Batteries, Joshua Wilder

All Theses

As the development of energy storage technologies for renewable energy sources continues to garner attention, vanadium redox flow batteries (VRFBs) have emerged as promising source for grid-scale energy storage. A major component of the VRFB is the ion exchange membrane (IEM), as it serves to physically separate the two liquid electrolytes and also directly impacts the battery’s capacity, efficiency, and cost. The current industry standard IEM, Nafion™, not only accounts for a large portion of the cost of the VRFB but also suffers from poor proton selectivity. Herein, we have synthesized sulfonated poly(ether ether ketone) (SPEEK) composite membranes containing amine …

Development Of A New Affinity Membrane For Rapidly Purifying Non-Antibody Proteins, Friendship Edioma

All Theses

This thesis project describes the modification and evaluation of a new affinity membrane for rapid chromatographic purification of non-antibody proteins. The affinity membrane utilizes Im7/CL7 coupling technology developed by Dr. Vassylyev's lab at the University of Alabama Birmingham (UAB), licensed by TriAltus Bioscience. The behavior of the membrane was evaluated using purified CL7-tagged Cas9 as my model protein for static and dynamic binding capacity analysis.

Chapter one provides an overview on biopharmaceutical drug production process development. I discuss how protein drugs are produced, isolated, and purified from the cell supernatant after upstream phases are completed. Despite increasing demands for biologics, …

Reactive Thin Film Polymers And Thin Film Composite Membranes For The Rapid Screening Of Uranium Isotopes, Abenazer W. Darge

All Dissertations

Traditional radiochemistry approaches for the detection of trace-level alpha-emitting radioisotopes in water require lengthy offsite sample preparations and do not lend themselves to rapid quantification. Therefore, a novel platform is needed that combines onsite purification, concentration, and isotopic screening with a fieldable detection system. My dissertation research objective was to develop novel reactive thin polymer films and thin film composite membranes for the selective separation of uranium from environmental water followed by direct isotopic analysis by alpha spectroscopy. Chapter 1 reviews progress made on uranium separation from aqueous matrices and discusses methods used for the determination of isotopic composition.

Extractive Membranes For The Detection And Screening Of Waterborne Plutonium, James Foster May 2022

Extractive Membranes For The Detection And Screening Of Waterborne Plutonium, James Foster

All Dissertations

The development of rapid screening tools for special nuclear materials remains a crucial focus for nonproliferation efforts. Traditional approaches for the analysis of trace-level Pu isotopes in water requires tedious and time-consuming sample preparation steps that do not lend well to expeditious screening. Therefore, a novel analytical method that combines both Pu concentration and source preparation into a single detection system would make for an invaluable tool for nuclear security applications. Extractive membranes absorbers can help to fulfill this role as they are capable of concentrating Pu to detectable limits while subsequently serving as alpha spectrometry sample sources. In Chapter …
