Chronic kidney disease occurs when the kidneys lose the ability to effectively filter the blood, which causes waste build up. End-stage kidney disease (ESKD) is the most severe case of kidney disease and requires either dialysis or a kidney transplant. Hemodialysis, the most common form of dialysis, is an extremely time-intensive and physically burdensome process that is currently limited by membrane performance. Human mesenchymal stem cells (hMSCs) have gained popularity in clinical trials due to their multipotent differentiation characteristics, ability to secrete bioactive molecules, migrate into diseased or damaged tissues, and their immunosuppressive properties. HMSC cultures are heterogeneous, containing stem cells, partially differentiated progenitor cells, and fully differentiated cells. One of the major challenges with hMSCs therapeutic potential is the inability to select specific cell subpopulations due to an insufficient number of biomarkers. Neuronal membrane disruption and mechanoporation are nanoscale damage mechanisms that critically affect brain cell viability during traumatic brain injury (TBI). These nanoscale cellular impairments are elusive in experiments and necessitate in silico approaches such as molecular dynamics (MD) simulations. Implementing MD, this research aims to investigate the effects of different key factors related to membrane deformation and damage, including force field resolutions, lipid compositions, and loading conditions.
To examine the impact of force field resolution, MD deformation simulations were conducted on 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC) lipid bilayer membranes, using all-atom (AA), united-atom (UA), and coarse-grained Martini (CG-M) force fields. The mechanical responses …

Produced water (PW) generated from hydraulic fracturing operations was treated using an integrated electrocoagulation, ultrafiltration, membrane distillation, and crystallization process (EC UF MDC). The aim was to determine the viability of this integrated process for maximizing water recovery. The results obtained here indicate that optimizing the various unit operations could lead to increased recovery of PW. Membrane fouling limits all membrane separation processes. A pretreatment step to suppress fouling is essential. Here, removal of total suspended solids (TSS) and total organic carbon (TOC) was achieved by electrocoagulation (EC) followed by ultrafiltration (UF). Tangential flow filtration has many advantages for bioreactor harvesting as the permeate could be introduced directly to the subsequent capture step, the process is easy to scale up, and fouling of the filter is limited by the cross flow. However, membrane fouling has limited its widespread use. This is particularly problematic given the high cell densities encountered today. This thesis covers a two part project: the production methods to create a double collagen binding domain molecule with a growth factor for wound healing applications and the development of a new in-house production method for isolating C1q from bovine blood. Virus filtration is an integral part of the downstream purification of mammalian cell culture-derived biotherapeutics to assure the viral safety of the products. Virus filtration membranes remove viruses based on a size-exclusion mechanism. Commercial parvovirus filers possess unique membrane structure and are designed to remove smaller non-enveloped parvoviruses with size 18-26 nm. However, some filters face issues, such as pre-mature fouling, the decline of filtrate flux, and reduction in virus retention. This report investigates the most reasonable, cost efficient, and reliable ways to integrity test a filter made from a permeable silicone membrane for future biomedical and pharmaceutical applications. The goal of the project was to create a fixture that a pharmaceutical company can use to quickly and reliably test the integrity of the filter. Motivated to produce electricity with photon activated ion pumps, the main purpose of this work was to characterize the photosynthetic membrane proteins bacteriorhodopsin (proton pump) and halorhodopsin (chloride pump). The proteins were re-suspended in lipid bilayers. For this work, an experimental set-up was built which included: chambers for lipid bilayer formation and characterization, lasers for ion pump activation, and an AxoPatch electrophysiology system for small photocurrent measurement. Lipid bilayer membranes were formed using mostly folding method: folding two monolayers together. The membranes were characterized by their resistance, capacitance, and generated photocurrent. Selectively permeable biological membranes composed of lipophilic barriers inspire the design of biomimetic carrier-mediated membranes for aqueous solute separation. This work imparts selective permeability to lipid-filled pores of silica thin film composite membranes using carrier molecules that reside in the lipophilic self-assemblies. The lipids confined inside the pores of silica are proven to be a more effective barrier than bilayers formed on the porous surface through vesicle fusion, which is critical for quantifying the function of an immobilized carrier. Currently there are external artificial lungs circulating blood outside the body, taking out the carbon dioxide, and inserting oxygenated blood back into the body. An example of this type of machine is the ExtraCorporeal Membrane Oxygenation (ECMO) machine currently used in hospitals. Amyloid Beta (Aβ) was the major focus of this study. It is a peptide that is present in the brain with a high tendency to self-aggregate. When this protein aggregates, it forms oligomers and protofibrils which in turn are deposited as senile plaques in the brain. The reason for the concern with these plaques is their association with the neurological disorder Alzheimer’s disease. Hollow fiber membranes are widely used in blood oxygenators to remove carbon dioxide and add oxygen during cardiopulmonary bypass operations. These devices are now widely used off-label by physicians to perform extracorporeal blood oxygenation for patients with lung failure. Unfortunately, the hollow fiber membranes used in these devices fail prematurely due to blood plasma leakage and gas emboli formation.
This project formed ultrathin (~100nm) polymer coatings on polymer hollow fiber membranes. The coatings were intended to “block” existing pores on the exterior surfaces while permitting high gas fluxes. This coating is synthesized using surface imitated control radical polymerization.

Polylactic acid (PLA) is becoming an increasingly important biopolymer for packaging applications. PLA brittleness limits its applicability. This study evaluated PLA properties with increasing amounts of added polyethylene glycol (PEG) plasticizer. Oxygen transmission rate (OTR) of cast films was determined using the newly available Dynamic Accumulation (DA) method. Arrhenius temperature sensitivity of OTR and polymer Permeability was also determined.
