Chronic kidney disease occurs when the kidneys lose the ability to effectively filter the blood, which causes waste build up. End-stage kidney disease (ESKD) is the most severe case of kidney disease and requires either dialysis or a kidney transplant. Hemodialysis, the most common form of dialysis, is an extremely time-intensive and physically burdensome process that is currently limited by membrane performance. The polysulfone (PSf) membranes that are widely used for dialysis today have many limitations, including fouling, toxin rejection, and essential protein loss. Cell therapy is a technology that relies on replacing diseased or dysfunctional cells with healthy functioning ones. One of the cells used for such advanced therapies are stem cells, owing to their ability to differentiate into specific cells required for repairing damaged or defective tissues or cells. Tangential flow filtration has many advantages for bioreactor harvesting as the permeate could be introduced directly to the subsequent capture step, the process is easy to scale up, and fouling of the filter is limited by the cross flow. However, membrane fouling has limited its widespread use. This is particularly problematic given the high cell densities encountered today. Virus filtration is an integral part of the downstream purification of mammalian cell culture-derived biotherapeutics to assure the viral safety of the products. Virus filtration membranes remove viruses based on a size-exclusion mechanism. Commercial parvovirus filers possess unique membrane structure and are designed to remove smaller non-enveloped parvoviruses with size 18-26 nm. However, some filters face issues, such as pre-mature fouling, the decline of filtrate flux, and reduction in virus retention. End-stage renal disease (ESRD) is currently the ninth leading cause of death in the United States, and of the 661,00 Americans diagnosed with ESRD, approximately 468,800 were on hemodialysis in 2016. The ninth leading cause of death in the United States is kidney disease, and hemodialysis is the process most commonly prescribed for treatment. It utilizes a selectively permeable membrane filter to remove toxins such as urea from the blood and retain necessary protein levels. Motivated to produce electricity with photon activated ion pumps, the main purpose of this work was to characterize the photosynthetic membrane proteins bacteriorhodopsin (proton pump) and halorhodopsin (chloride pump). The proteins were re-suspended in lipid bilayers. For this work, an experimental set-up was built which included: chambers for lipid bilayer formation and characterization, lasers for ion pump activation, and an AxoPatch electrophysiology system for small photocurrent measurement. Lipid bilayer membranes were formed using mostly folding method: folding two monolayers together. The membranes were characterized by their resistance, capacitance, and generated photocurrent. Selectively permeable biological membranes composed of lipophilic barriers inspire the design of biomimetic carrier-mediated membranes for aqueous solute separation. This work imparts selective permeability to lipid-filled pores of silica thin film composite membranes using carrier molecules that reside in the lipophilic self-assemblies. The lipids confined inside the pores of silica are proven to be a more effective barrier than bilayers formed on the porous surface through vesicle fusion, which is critical for quantifying the function of an immobilized carrier. Artificial lungs are in use, but difficult issues remain in the field of membrane development related to fouling issues. Currently there are external artificial lungs circulating blood outside the body, taking out the carbon dioxide, and inserting oxygenated blood back into the body. An example of this type of machine is the ExtraCorporeal Membrane Oxygenation (ECMO) machine currently used in hospitals. The ECMO takes over the functions for both the lungs and the heart but is only available for short term use by patients with respiratory failure due to infections [1]. Hollow fiber membranes are widely used in blood oxygenators to remove carbon dioxide and add oxygen during cardiopulmonary bypass operations. These devices are now widely used off-label by physicians to perform extracorporeal blood oxygenation for patients with lung failure. Unfortunately, the hollow fiber membranes used in these devices fail prematurely due to blood plasma leakage and gas emboli formation.

This project formed ultrathin (~100nm) polymer coatings on polymer hollow fiber membranes. The coatings were intended to “block” existing pores on the exterior surfaces while permitting high gas fluxes. This coating is synthesized using surface imitated control radical polymerization.

Polylactic acid (PLA) is becoming an increasingly important biopolymer for packaging applications. PLA brittleness limits its applicability. This study evaluated PLA properties with increasing amounts of added polyethylene glycol (PEG) plasticizer. Oxygen transmission rate (OTR) of cast films was determined using the newly available Dynamic Accumulation (DA) method. Arrhenius temperature sensitivity of OTR and polymer Permeability was also determined.
