Bacteria are constantly threatened with infection by mobile genetic elements (MGE) such as bacteriophage and plasmids. Bacteriophage and plasmids require the bacteria's cellular infrastructure to replicate their genomes. Rampant replication can lead to cell death which is one reason why bacteria have developed a diverse array of immune systems to prevent or limit infection. This thesis studies three types of bacterial immune systems, type IV-A CRISPR-Cas (Clustered Regularly Interspaced Short Palindromic Repeat –CRISPR associated), type V-A2 CRISPR-Cas systems, and Wadjet systems.

Introduction: Determining the biological significance of pteridines in cancer development and progression remains an important step in understanding the altered levels of urinary pteridines seen in certain cancers. Our companion study revealed that several folate-derived pteridines and lumazines correlated with tumorigenicity in an isogenic, progressive breast cancer cell model, providing direct evidence for the tumorigenic origin of pteridines. Objectives: This study sought to elucidate the pteridine biosynthetic pathway in a progressive breast cancer model via direct pteridine dosing to determine how pteridine metabolism changes with tumorigenicity. Electrochemical reduction of carbon dioxide into eco-friendly and clean products is a promising approach to eradicate pollution. Although carbon dioxide emission is inhibited by the advent of renewable sources of energy, it is present in the atmosphere and needs to be cleaned. The goal of this project is to explore a new method to efficiently remove Pb(II) ions from water by processing Undaria pinnatifida into immobilized beads using sodium alginate and calcium chloride. The resulting biosorbent was characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy (SEM-EDS). Using immobilized U. pinnatifida, we investigated the effect of various factors on Pb(II) ion removal efficiency such as temperature, pH, ionic strength, time, and underlying biosorption mechanisms. In June 2018 the team embarked on an ambitious project to address the slow development pace of Alaska's 20+ billion barrels heavy oil resource via the first ever polymer flood pilot. Following the successful commencement of the pilot in August 2018, the field demonstration, supporting laboratory experiments and numerical simulation have steadily progressed. A significant amount of valuable data and lessons learned have been collected and are reported in this paper. Due to increasing concentrations in the atmosphere, carbon dioxide has, in recent times, been targeted for utilisation (Carbon Capture Utilisation and Storage, CCUS). In particular, the production of CO from CO2 has been an area of intense interest, particularly since the CO can be utilized in Fischer–Tropsch synthesis. Lamellar lyotropic liquid crystal-silver nanoparticles (LLLC-AgNPs) are synthesized using the in-situ method. The polarizer microscope showed a mosaic-like pattern with fan shape textures. The high resolution transmission electron microscope (HRTEM) confirmed that the AgNPs are spherical in shape with 4 – 12 nm size. Differential scanning calorimetry (DSC) determined the LLLC-AgNPs transition anisotropic phase to the highly ordered isotropic phase at 300 °C, and crystallinity was completed at 350 °C. Thermogravimetric analysis (TGA) verified that the LLLC-AgNPs were completely decomposed at 700 °C. The structural, transport, and thermodynamic properties related to cellulose dissolution by tetrabutylphosphonium chloride (TBPCl) and tetrabutylphosphonium hydroxide (TBPH)-water mixtures have been calculated via molecular dynamics simulations. For both ionic liquid (IL)-water solutions, water veins begin to form between the TBPs interlocking arms at 80 mol % water, opening a pathway for the diffusion of the anions, cations, and water. The water veins allow for a diffusion regime shift in the concentration region from 80 to 92.5 mol % water, providing a higher probability of solvent interaction with the dissolving cellulose strand. Acute myocardial infarction (AMI), also termed as heart attack, is often clinically screened by blood test through the measurement of cardiac biomarkers (i.e., troponin (T or I), creatine kinase (CK−MB) and myoglobin) and electrocardiography (ECG) changes which are presented to the hospital. Device calibration for in vivo studies is difficult for current non-steady state analytes of interest correlated with both inflammatory response and microbial signaling molecules (QS); which exist in low ng/mL to pg/mL with molecular weights over a wide range of 170 Da to 70 kDa. This article introduces an alignment-free clustering method in order to cluster all the 66 DORs sequentially diverse protein sequences. Two different methods are discussed: one is utilizing twenty standard amino acids (without grouping) and another one is using chemical grouping of amino acids (with grouping). Two grayscale images (representing two protein sequences by order pair frequency matrices) are compared to find the similarity index using morphology technique. In E. coli, fatty acid synthesis is catalyzed by the enzyme acetyl-CoA carboxylase (ACC), which converts acetyl-CoA into malonyl-CoA. Malonyl-CoA is a major building block for numerous of bioproducts. Multiple parameters regulate the homeostatic cellular concentration of malonyl-CoA, keeping it at a very low level. Understanding how these parameters affect the bacterial production of malonyl-CoA is fundamental to maximizing it and its bioproducts. Modification of the carbohydrate cyclodextrin increases the ability to encapsulate hydrophobic pharmaceutical molecules by forming a carrier with a hydrophobic core and hydrophilic exterior. This paper aims to shed light on the dissonance caused by the superimposition of Dominant Human Systems on Natural Systems. I highlight the synthetic nature of Dominant Human Systems as egoic and linguistic phenomenon manufactured by a mere portion of the human population, which renders them inherently oppressive unto peoples and landscapes whose wisdom were barred from the design process. Over the last decade, Unconventional Liquids Rich Reservoirs (ULR) have become the main target for oil and gas investors as conventional formations started to deplete and diminish in numbers. These unconventional plays have a huge oil reserve; however, the primary oil recovery factor is predicted to be less than 10%. Unconventional Improved Oil Recovery (UIOR) techniques are still a new concept in the oil industry since there is no commercial project reported for any IOR technique yet. One of the EOR technologies is low-salinity water flooding (LSWF) is an EOR method that operates at a lower cost than other EOR methods, which makes it a preferred area of interest for oil industry economists, who continue to call for EOR costs to come down. The aim is to minimize the possible cost to recover as much oil as possible.
