Department of Agricultural and Biological Systems Engineering: Faculty Publications

Defining the final size and geometry of engineered tissues through precise control of the scalar and vector components of tissue growth is a necessary benchmark for regenerative medicine, but it has proved to be a significant challenge for tissue engineers. The growth plate cartilage that promotes elongation of the long bones is a good model system for studying morphogenetic mechanisms because cartilage is composed of a single cell type, the chondrocyte; chondrocytes are readily maintained in culture; and growth trajectory is predominately in a single vector. In this cartilage, growth is generated via a differentiation program that is spatially and …

The energy-water nexus is the inseparable connection linking water and energy. We are faced with a unique opportunity to co-manage these resources, as conservation of one is directly linked to the conservation of its counterpart. Therefore, immediately facing this critical challenge, will lead to tangible impacts on the water and energy crisis our food system is faced with. Determining the role of water and energy in the food industry has proved to be the starting point for reducing the distance between process productivity and resource efficiency. Therefore, this research focuses on determining opportunities for water-energy optimization and wastewater reduction and …

Sugar Versus Lipid For Sustainable Biofuels, Yaşar Demirel Jan 2017

Sugar Versus Lipid For Sustainable Biofuels, Yaşar Demirel

Yaşar Demirel Publications

Introduction

First‐generation biofuels, namely, ethanol and biodiesel, have led to far reaching impact on the peoples’ life world‐wide.[1] However, they inter-fere with the food supply chain and may not be sustainable although some of the biomass are converted to biofuels after those biomasses have met the human needs. Still, the first‐generation–based biofuels have proved that sugar and lipid platforms can be an answer to energy security and global warming concerns without the need for new infrastructure for feedstock delivery as well as for biomass‐to‐biofuel conversion tech-nologies. At the same time, we are discovering and assessing the long‐term environmental im-plications on …

Fractionation And Characterization Of Lignin Streams From Genetically Engineered Switchgrass, Enshi Liu Jan 2017

Fractionation And Characterization Of Lignin Streams From Genetically Engineered Switchgrass, Enshi Liu

Theses and Dissertations--Biosystems and Agricultural Engineering

Development of biomass feedstocks with desirable traits for cost-effective conversion is one of the main focus areas in biofuels research. As suggested by techno-economic analyses, the success of a lignocellulose-based biorefinery largely relies on the utilization of lignin to generate value-added products, i.e. fuels and chemicals. The fate of lignin and its structural/compositional changes during pretreatment have received increasing attention; however, the effect of genetic modification on the fractionation, depolymerization and catalytic upgrading of lignin from genetically engineered plants is not well understood. This study aims to fractionate and characterize the lignin streams from a wild-type and two genetically engineered …

Department of Agricultural and Biological Systems Engineering: Faculty Publications

Background: Type-C dose algorithms provide more accurate dosimetry for lung SBRT treatment planning. However, because current dosimetric protocols were developed based on conventional algorithms, its applicability for the new generation algorithms needs to be determined. Previous studies on this issue used small sample sizes and reached discordant conclusions. Our study assessed dose calculation of a Type-C algorithm with current dosimetric protocols in a large patient cohort, in order to demonstrate the dosimetric impacts and necessary treatment planning steps of switching from a Type-B to a Type-C dose algorithm for lung SBRT planning.

Methods: Fifty-two lung SBRT patients were included, each …

Using Elastin-Like Polypeptides For Better Retention Of Biofuels, Yu Hong Wang, Ethan T. Hilman, Kevin V. Solomon Aug 2016

Using Elastin-Like Polypeptides For Better Retention Of Biofuels, Yu Hong Wang, Ethan T. Hilman, Kevin V. Solomon

The Summer Undergraduate Research Fellowship (SURF) Symposium

Elastin-like polypeptides (ELPs) are synthetic molecules that exhibit an interesting property of inverse temperature phase transition; they exist as soluble monomers at low temperatures and form insoluble aggregates at higher temperatures. The transition temperature depends on the pH, salt concentration, and the amino acid sequence of the ELP. This unique and reversible behavior, along with their high biocompatibility has made them a strategic tool for various biomedical applications. However, their hydrophobic properties also make them a prime candidate for biofuel production. As high levels of many commercially important organic solvents are toxic to the cells that make them, ELPs can …

Using A Vnir Spectral Library To Model Soil Carbon And Total Nitrogen Content, Nuwan K. Wijewardane

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

n-situ soil sensor systems based on visible and near infrared spectroscopy is not yet been effectively used due to inadequate studies to utilize legacy spectral libraries under the field conditions. The performance of such systems is significantly affected by spectral discrepancies created by sample intactness and library differences. In this study, four objectives were devised to obtain directives to address these issues. The first objective was to calibrate and evaluate VNIR models statistically and computationally (i.e. computing resource requirement), using four modeling techniques namely: Partial least squares regression (PLS), Artificial neural networks (ANN), Random forests (RF) and Support vector regression …

Municipal Composting And Organic Waste Diversion: The Case Of Fayetteville, Arkansas, Michael E. Hoppe May 2016

Municipal Composting And Organic Waste Diversion: The Case Of Fayetteville, Arkansas, Michael E. Hoppe

Biological and Agricultural Engineering Undergraduate Honors Theses

It is estimated that 40% of food is wasted in the United States; representing $165 billion in wasted resources. A vast majority of that wasted food is ultimately placed in landfills where it decomposes and releases harmful greenhouse gases (GHGs). In fact, food waste alone is responsible for 23% of annual methane emissions for the US. This has a huge impact on global climate change due to the potency of methane as a greenhouse gas. Currently only 5% of the food waste produced is recovered across the nation. Source reduction would be the best solution to reducing this food waste, …

Protein-Targeted Corona Phase Molecular Recognition, Gili Bisker, Juyao Dong, Hoyoung D. Park, Nicole M. Iverson, Jiyoung Ahn, Justin T. Nelson, Markita P. Landry, Sebastian Kruss, Michael S. Strano

Department of Agricultural and Biological Systems Engineering: Faculty Publications

Corona phase molecular recognition (CoPhMoRe) uses a heteropolymer adsorbed onto and templated by a nanoparticle surface to recognize a specific target analyte. This method has not yet been extended to macromolecular analytes, including proteins. Herein we develop a variant of a CoPhMoRe screening procedure of single-walled carbon nanotubes (SWCNT) and use it against a panel of human blood proteins, revealing a specific corona phase that recognizes fibrinogen with high selectivity. In response to fibrinogen binding, SWCNT fluorescence decreases by >80% at saturation. Sequential binding of the three fibrinogen nodules is suggested by selective fluorescence quenching by isolated sub-domains and validated …

Use Of Lidar In The Design Of Grassed Waterways: Case Study In Agricultural Management In Oklahoma, Annette Sparks

Dissertations, Master's Theses and Master's Reports

Before the advent of Light Detection and Ranging (LiDAR) technology, farm-level infrastructure for sustainable agriculture was designed using topographic maps. LiDAR is a remote sensing method whereby reflected laser pulses are measured to generate high-resolution 3D images of the terrain. This report compares these two methods in the design of grassed waterways for flood drainage and evaluates how the quantitative difference between the two methods can affect the overall hydrologic design. The benefits of LiDAR are expected to be higher accuracy and precision in design, as well as greater reproducibility or consistency, regardless of the designer. To carry out this …

Automated Solid-Substrate Cultivation Of The Anaerobic Bacterium Clostridium Thermocellum, Mathew J. Ruwaya Jan 2016

Automated Solid-Substrate Cultivation Of The Anaerobic Bacterium Clostridium Thermocellum, Mathew J. Ruwaya

Theses and Dissertations--Biosystems and Agricultural Engineering

The organism Clostridium thermocellum grows on cellulosic substrates and produces ethanol, acetate, lactate, formic acid, and CO2. The organic acids produced alter the growth environment in which the bacteria grows and ultimately inhibit bacterial growth. One method which has been used successfully to maintain the system at acceptable growth conditions is to intermittently flush out the spent media and metabolic products and replace with new fermentation media. Our goal was to design and build an automated system that will automatically flush the spent media from the growing culture and resupply new media without manual intervention. An automated control …

A Novel Synthetic Yeast For Enzymatic Biodigester Pretreatment, Tianyu Tan, Mark S. Aronson, Arren Liu, Jill H. Osterhus, Melissa Robins, Suraj Mohan, Erich Leazer, Bowman Clark, Alexa Petrucciani, Katherine Lowery, James Welch, Casey Martin, Helena Lysandrou, Michael E. Scharf, Jenna Rickus

The Summer Undergraduate Research Fellowship (SURF) Symposium

Lignin, a complex organic polymer, is a major roadblock to the efficiency of biofuel conversion as it both physically blocks carbohydrate substrates and poisons biomass degrading enzymes, even if broken down to monomer units. A pretreatment process is often applied to separate the lignin from biomass prior to biofuel conversion. However, contemporary methods of pretreatment require large amounts of energy, which may be economically uncompelling or unfeasible. Taking inspiration from several genes that have been isolated from termites and fungi which translate to enzymes that degrade lignin, we want to establish a novel “enzymatic pretreatment” system where microbes secrete these …

Development Of A Novel Enzymatic Pre-Treatment For Lignocellulosic Biomass, Melissa Robins, Jenna Rickus Aug 2015

Development Of A Novel Enzymatic Pre-Treatment For Lignocellulosic Biomass, Melissa Robins, Jenna Rickus

The Summer Undergraduate Research Fellowship (SURF) Symposium

Biofuels, fuels derived directly from living matter, present a renewable and environmentally friendly alternative to petroleum based fuels. Bioethanol produced from low input energy crops or agricultural waste is a promising fuel source because it does not interfere with the human food supply chain and the ethanol produced can be blended with gasoline. These potential sources of bioethanol are not yet commercially viable due to a polymer called lignin present in the plant’s cell wall which impedes the conversion of cellulose to glucose and the eventual fermentation of glucose to ethanol. Developing new methods for the pretreatment of lignocellulosic biomass …

A Pump Monitoring Approach To Irrigation Pumping Plant Performance Testing, William Merritt Mcdougall May 2015

A Pump Monitoring Approach To Irrigation Pumping Plant Performance Testing, William Merritt Mcdougall

Graduate Theses and Dissertations

Traditionally, irrigation pumping plants have been tested using an instantaneous appraoch, which tests performance parameters over a very short time interval. Using this method, the tester measures the necessary work and energy use parameters to calculate the desired pumping plant performance values. The primary limitation of this approach is its inability to determine the season long efficiency of an irrigation pumping plant.

A new approach to evaluating irrigation pumping plant performance is the use of pump monitoring systems which use high frequency, real-time data collection and telemetry to relay information directly from the pump to the user. This method of …

Transformation Of Swine Manure And Algal Consortia To Value-Added Products, Mahmoud A. Sharara May 2015

Transformation Of Swine Manure And Algal Consortia To Value-Added Products, Mahmoud A. Sharara

Graduate Theses and Dissertations

The swine production sector is projected to grow globally. In the past, this growth manifested itself in increased herd sizes and geographically concentrated production. Although economically sound, these trends had negative consequences on surrounding ecosystems. Over-application of manure resulted in water quality degradation, while long-term storage of manure slurries was found to promote release of potent GHG emissions. There is a need for innovative approaches for swine manure management that are compatible with current scales of production, and increasingly strict environmental regulations.

This study aims to investigate the potential for incorporating gasification as part of a novel swine manure management …

Investigation Of Phanerochaete Chrysosporium And Clostridium Thermocellum For Improved Saccharification Of Lignocellulose Under Nonsterile Conditions, William E. Simon

Theses and Dissertations--Biosystems and Agricultural Engineering

Current research efforts are directed at developing competitive processes that can utilize lignocellulose as a feedstock for biorefineries. The purpose of this study was to investigate methods of processing lignocellulosic material so that its monosacharides can be more easily accessed for fermentation, the lack of which is hindering the economics and widescale adoption of lignocellulosic biorefining. The monosaccharides are of interest because they can be used by Clostridium beijerinckii downstream of P. chrysosporium and C. thermocellum in a sequential bioprocess to produce butanol. Butanol is an attractive biofuel because it can be utilized without modifying current transportation infrastructure. Butanol is …

Characterization Of Extraction Methods To Recover Phenolic-Rich Antioxidants From Blue Green Algae (Spirulina) Using Response Surface Approaches, Ahmad Salamatullah

Department of Food Science and Technology: Dissertations, Theses, and Student Research

Blue green algae (spirulina) is a dietary system that is widely consumed as a whole food or as a supplement in many Asian countries where populations are mostly unaffected by many of the diseases currently afflicting western societies, such as cancer, heart disease and arthiritis. Indeed, spirulina is a rich source of antioxidants with the phenolic compounds playing a significant role. As components of a complex dietary system, phenolic compounds can act alone or through synergistic mechanisms to impart a greater biologic effect than can be elicited by a sum of the individual parts. Therefore, an understanding of the antioxidative …

Pretreatment Of Scenedesmus Sp. Biomass As A Potential Anaerobic Digestion Substrate, Xinyi E, Czarena Crofcheck

Biosystems and Agricultural Engineering Faculty Publications

Algal biomass can be a potential substrate for anaerobic digestion. However, raw algae cells show a resistance to biological degradation, resulting in a slower methane production rate. Varying thermal and chemical pretreatments of algal biomass were investigated in an attempt to increase soluble organic matter (SOM) yield, which would result in enhanced methane production during subsequent anaerobic digestion. Scenedesmus sp. was harvested using three different procedures: with flocculation, with flocculation and drying, and without flocculation or drying. For all pretreatments and algae types, fluorescence micrographs were obtained to visually confirm the degradation of the algal cell walls. A complete 2 …

Techno-Economic And Fluid Dynamics Analysis For Growing Microalgae With The Intent Of Producing Biofuel Using A System Model, Leah R. Raffaeli

Electronic Theses and Dissertations

Techno-economic and systems studies on microalgal growth scenarios to date are abbreviated and missing a number of important variables. By including these variables in a detailed model integrating biology, chemistry, engineering, and financial aspects, a more defined systems analysis is possible. Through optimizing the model productivity based on the resulting net profit, the system analysis results in a more accurate assessment of environmental and economic sustainability of specific algal growth scenarios. Photobioreactor algal growth scenario optimization in the system model has resulted in realistic engineering design requirements based on algal growth requirements and fluid dynamics analysis. Results show feasibility for …

Evaluation Of Endogenous Allergens For The Safety Evaluation Of Genetically Engineered Food Crops: Review Of Potential Risks, Test Methods, Examples And Relevance, Richard E. Goodman, Rakhi Panda, Harsha Ariyarathna Jan 2013

Evaluation Of Endogenous Allergens For The Safety Evaluation Of Genetically Engineered Food Crops: Review Of Potential Risks, Test Methods, Examples And Relevance, Richard E. Goodman, Rakhi Panda, Harsha Ariyarathna

Department of Food Science and Technology: Faculty Publications

The safety of food produced from genetically engineered (GE) crops is assessed for potential risks of food allergy on the basis of an international consensus guideline outlined by the Codex Alimentarius Commission (2003). The assessment focuses on evaluation of the potential allergenicity of the newly expressed protein(s) as the primary potential risk using a process that markedly limits risks to allergic consumers. However, Codex also recommended evaluating a second concern, potential increases in endogenous allergens of commonly allergenic food crops that might occur due to insertion of the gene. Unfortunately, potential risks and natural variation of endogenous allergens in non-GE …

Surface- And Hydrogel-Mediated Delivery Of Nucleic Acid Nanoparticles, Angela K. Pannier, Tatiana Segura

Department of Agricultural and Biological Systems Engineering: Faculty Publications

Gene expression within a cell population can be directly altered through gene delivery approaches. Traditionally for nonviral delivery, plasmids or siRNA molecules, encoding or targeting the gene of interest, are packaged within nanoparticles. These nanoparticles are then delivered to the media surrounding cells seeded onto tissue culture plastic; this technique is termed bolus delivery. Although bolus delivery is widely utilized to screen for efficient delivery vehicles and to study gene function in vitro, this delivery strategy may not result in efficient gene transfer for all cell types or may not identify those delivery vehicles that will be efficient in vivo. …

Dynamics Of Depression Storage During Sprinkler Irrigation And Precipitation Events, Michael B. Mckinney Jul 2012

Dynamics Of Depression Storage During Sprinkler Irrigation And Precipitation Events, Michael B. Mckinney

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Surface depression storage (Ds) is the volume of precipitation excess which is stored by surface microrelief features of soils. The volume of water stored in surface depressions when precipitation rate exceeds infiltration rate reduces the amount of runoff generated. Because Ds is dependent on soil microrelief, land slope, and crop residue, tillage and management practices can have a considerable impact on the magnitude of this value.

When modeling irrigation systems and surface hydrology, depression storage is often treated as a static abstraction, meaning that maximum storage volume must be filled before runoff occurs. However, several researchers have documented …

Photosynthethically active radiation (PAR) is a principle environmental variable used by horticultural specialists, agronomists and ecosystem modelers to characterize the quantity and quality of light conducive to plant growth and development. Spatial distribution of PAR in a greenhouse can be quite variable and diffuse throughout the day time photoperiod, especially at low sun angles in northern regions of the United States. Four colors of reflective plastic mulches (white, red, olive, and black) were evaluated for winter-time strawberry (Fragaria × ananassa Duch.) production based on their reflectance and transmittance properties in a double-polyethylene, plastic-glazed Quonset greenhouse inNebraska. The spectral properties …

The Economics And Logistics Of The Dual Harvest Of Grain And Biomass In A Single-Pass, Matthew T. Wold Dec 2011

The Economics And Logistics Of The Dual Harvest Of Grain And Biomass In A Single-Pass, Matthew T. Wold

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Significant interest has developed in using cellulosic resources, especially crop residues, to create biofuels. Collecting these residues in a single-pass of the harvester across the field has the potential to be a low cost option. Two models have been developed; the first characterizes the in-field logistics of single-pass crop residue collection, the second the economics. These models allow the user to easily examine a wide variety of both grain-only and single-pass residue collection harvest cases. A variety of possible residue collection cases have been examined, and their effects both on harvester field capacity and harvest cost compared to grain-only harvest …

Extrusion And Alkali Extrusion Of Corn Stover To Improve Enzyme Saccharification, Shujing Zhang

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Biomass pretreatment constitutes an important part in the whole process of bioethanol production. The main goals of pretreatment include improving cellulose accessibility to enzyme, enhancing sugar yields from hemicellulose, achieving low capital and energy costs, as well as low sugar degradation and low chemical requirements. Extrusion, as a continuous and high throughput pretreatment method, requires much less chemical compared to traditional alkali pretreatment. In this research, twin-screw extrusion was evaluated for its effect with regard to promoting sugar yields from corn stover. Also, the underlying factors contributing to this improvement were examined. It was found that extrusion with no chemicals …
