The Microphysiological Systems Laboratory aims to develop colorectal cancer tumor models under a hypoxic environment to assess model response to pharmaceutical compounds in vitro. To perform relevant studies, researchers have attempted to use different hypoxic inducing strategies such as a nitrogen pod and hypoxic incubator to recreate in vivo physiological responses to hypoxia. However, studies would be interrupted due to incubator functionality failure. To ensure successful and physiologically relevant studies, I improved and verified the robustness and reliability of a hypoxic incubator previously designed and manufactured in the lab. The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. Electrostatic artificial muscles benefit from a microfluidic approach due to the non-linearity of electrostatic attraction creating disproportionate benefits when miniaturized. When designed …

Implantable drug delivery devices have many benefits over traditional drug administration techniques and have attracted a lot of attention in recent years. By delivering the medication directly to the tissue, they enable the use of larger localized concentrations, enhancing the efficacy of the treatment. The intricate communication between different cell types within pancreatic islets plays a crucial role in regulating glucose homeostasis. Among these cellular interactions, the interplay between α cells (responsible for glucagon secretion) and δ cells (secreting somatostatin) is particularly intriguing due to the inhibitory effects of somatostatin on glucagon release. The extracellular matrix is a complex system surrounding cells that is crucial to all of the cardiovascular system’s functions. Fibrosis is an excessive accumulation (scarring) of the extracellular matrix. Fibrosis is typically a negative event, and the scarring must be reduced in the heart in order to maintain its functionality. Due to this, simulations are run using the software “Netflux” in order to determine the effects of different heart failure drugs on cardiac fibrosis. The high prevalence of dental caries among children and adolescents, especially those from lower socio-economic backgrounds, is a significant nationwide health concern. Early prevention, such as dental sealants and fluoride varnish (FV), is essential, but access to this care remains limited and disparate. In this research, a national dataset is utilized to assess sealants' reach and effectiveness in preventing tooth decay, particularly focusing on 2nd molars that emerge during early adolescence, a current gap in the knowledge base. This research explores the design and development of a forearm-based wearable bioimpedance device for non-invasive measurement of heart rate and respiratory rate at an accuracy level comparable to medical-grade monitors. Dental implantation is an increasingly common procedure used to treat missing teeth. However, surgical drilling to place the implant poses a high risk of injury to critical anatomy, such as inferior alveolar nerve injury or maxillary sinus perforation. A real-time surgical feedback system sensing proximity of these critical anatomy could reduce injury risks. This dissertation investigates the development of such a system that incorporates an electrical impedance sensor into the tip of a surgical drill.

Many patients struggle with disabilities that hinder their ability to walk. This project aimed to create a leg assembly capable of variable gravity so that it could be combined with a Robotic Walking Training Device, and lead to better rehabilitation options for patients. This was accomplished by deriving equations of joint torque, creating circuit diagrams for Arduino systems, modeling leg assemblies in CAD, and finally combining it to create a working small-scale prototype. Composite Performance Measures (CPMs) of human performance are widely employed in applications that range from mundane to life-changing, yet they are frequently presented without explanation or reference to theoretical underpinnings. The conceptual soundness of a widely used CPM approach, based on additive combination of constituent components, has been called into question by Kondraske. He argues the relevance of General Systems Performance Theory (GSPT) and performance capacity envelope concepts when applied to multidimensional CPM measures.

This thesis presents the development, calibration, and initial implementation of transcranial focused ultrasound (tFUS) for neuromodulation in a human study. The primary objectives were to design a system capable of delivering precise and safe ultrasound stimulation to targeted brain regions and to conduct preliminary trials to assess its effects on brain activity. A custom calibration device was developed to ensure accurate measurement of acoustic pressure, addressing challenges such as voltage alterations caused by changes in system impedance. Additionally, a neuro-navigation tool was created to facilitate precise targeting of the right subgenual anterior cingulate cortex (right sgACC), enabling accurate placement of …
