The Pet Mobility Project, sponsored by Terri Smith and Jasper Jackson, is an initiative aimed at providing a custom mobility aid to a pet in the San Luis Obispo community. Amber Rappozo’s young German Shepherd, LeDoux, was selected as the project’s recipient. LeDoux suffers from hindleg weakness and, as a result, has limited mobility. This document provides a comprehensive overview of the Pet Mobility Project, detailing the motivation, background, and design approach behind the development of a LeDoux’s custom mobility aid.

This thesis evaluates whether accessible and affordable technologies can provide clinically significant, quantifiable gait analysis comparable to traditional marker-based motion capture systems. Modern high-end systems offer excellent accuracy but remain financially and logistically inaccessible to many clinicians and researchers. To address this gap, a framework was developed using markerless pose estimation using the model, MediaPipe, inertial measurement units (IMUs) processed through an Extended Kalman Filter (EKF), and surface electromyography (sEMG) to measure joint kinematics and neuromuscular activity during gait. Cardiovascular diseases (CVDs) are the leading cause of death worldwide in males (XY) and females (XX). Prior to menopause, females have a relative protection against serious cardiac pathologies compared to age-matched males. This phenomenon is widely attributed to the ovarian hormone estrogen. Unfortunately, hormone replacement therapy to maintain estrogen levels in postmenopausal females has overall adverse effects, and it is not recommended for long-term use or as a preventative measure for eCVDs. Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. By utilizing the photothermal behavior of graphene-hydrogel composites confined within micron-sized fluidic channels, patterned light illumination creates the parallel and independent actuation of chemical release in a group of fluidic ports. For this Honors Thesis project, I designed and fabricated a ventilation and recirculation airway sub-system prototype for a hydroponic demonstration unit to be used as an educational demonstration for prospective students, illustrating the capabilities of biological engineers. This thesis focuses on the design of an autonomous air system that toggles between ventilation and recirculation modes based upon sensor output. In the literature review I discuss how ventilation can be used to achieve a targeted climate, the benefits of hands-on projects for education, and the optimum parameters for growing herbs in a hydroponic system.

Mitral valve surgery is incredibly prevalent in the United States with more than 40,000 mitral valve surgical procedures annually. Improving the imaging techniques used to diagnose these cases requires validation of 3D models through experimental data such as mechanical properties of the tissue. This thesis proposes an algorithm that determine the geometry of 3D-printed, custom-designed spring element bands made of thermoplastic elastomer (TPE) for use in a wearable orthotic device to aid in the physical therapy of a human hand exhibiting spasticity after stroke. Each finger of the hand is modeled as a mechanical system consisting of a triple-rod pendulum with nonlinear stiffness at each joint and forces applied at the attachment point of each flexor muscle. Accurate tracking of hand grip force is an important consideration needed for a robust understanding in the study of human biomechanics. One aspect where it proves useful is in hand-arm vibration, such as from gripping a power tool. Depending on how firmly the user is gripping the tool, they may change their exposure levels to the tool vibration, which can lead to potential disorders such as Hand-Arm Vibration Syndrome (HAVS).

This thesis successfully integrates laminate microfluidic devices with an analytic Surface Plasmon Resonance (SPR) instrument. Integration was accomplished at low-cost using materials such as polydimethylsiloxane (PDMS), Poly(methyl methacrylate) (PMMA), Tygon tubing, and a 3-way stopcock. The main components of this thesis are the design and fabrication of the low-cost, in-house fluidics that can integrate with upstream microfluidics and the validation of the in-house fluidics using the Biosensing Instruments BI-2000 SPR instrument.
