This document outlines a project sponsored by Elizabeth Mentel and Michael Clausson that addresses the shortcomings in current splinting options, particularly for wrist fractures in elderly patients. Stakeholders include clinicians, patients, and emergency medical technicians. Multiple improvements can be made to current splinting options to help alleviate inflammation along with splint versatility, affordability, accessibility, and ease of use. After determining the critical customer requirements, the primary objective of this project was to find a way to manage inflammation. Bone’s ability to resist fracture is often ignored until a low-energy fracture occurs. Patients with Chronic Kidney Disease (CKD) or osteoporosis are at an increased risk of low-energy fracture. Generally, fracture risk is evaluated by using a bone mineral density (BMD) test. BMD values; however, do not fully predict bone’s ability to resist fracture. This suggests that other parameters may be involved. Bone quality is the term used to describe these parameters, which are categorized into three groups: structural, material, and microdamage. Numerous molecular factors active in bone tissue direct fracture repair and remodeling which can be altered by disease conditions such as Peripheral Arterial Disease (PAD) and Osteoporosis. Methods of molecular biology are commonly applied to investigate the expression and role of these molecular factors. This project presents a robust three-step protocol for examining gene expression in the mouse tibia. The protocol begins with isolating RNA from a flash frozen tibia sample. The isolated RNA is reverse transcribed into cDNA. Finally, PCR is performed to indentify expressed genes. The onset of osteoporosis caused by aging, disease states, and post-menopausal conditions significantly impacts patient quality of life, required healthcare funding, personal autonomy losses from increased fracture risk and the subsequent corrective surgery. Research has indicated that osteocyte apoptosis may be a key parameter in bone remodeling, raising the possibility of remodeling rate modulation for the mitigation of bone mass resorption. By developing therapies that target osteocyte apoptosis, it may be possible to prevent undesired bone remodeling activity while maintaining a healthy balance between damage formation in the form of microcracks induced by the strain environment and the removal of …
