Studies suggest that P. gingivalis functions as a keystone pathogen and interacts with primary colonizers in the supragingival biofilm such as S. gordonii. This interaction contributes to the initial colonization of the oral cavity by P. gingivalis and thus represents a potential target for therapeutic intervention. We have identified a peptide (BAR) derived from the streptococcal SspB protein that functions to inhibit P. gingivalis adherence to S. gordonii. In addition, we showed that nanoparticles (NPs) functionalized with BAR inhibit this interaction more potently than free soluble peptide, possibly by promoting interaction with P. gingivalis at higher valency than …

The most frequent clinical presentation of peripheral arterial occlusive disease (PAOD) is intermittent claudication, which may be caused by impaired vasodilation. Patients demonstrate both local and systemic impairments in vasodilation, but as the collateral circulation is the primary site of resistance to the ischemic zone, impaired collateral vasodilation would have the greatest potential to induce claudication. Collateral function following arterial occlusion is not well defined, but immature collaterals may demonstrate impaired vasodilation in animal models, although this is potentially improved with exercise training. In the fight against drug-resistant bacteria, accurate and high-throughput detection is essential. Here, a bimaterial microcantilever with an embedded microfluidic channel with internal surfaces chemically or physically functionalized with receptors selectively captures the bacteria passing through the channel. Bacterial adsorption inside the cantilever results in changes in the resonance frequency (mass) and cantilever deflection (adsorption stress). The excitation of trapped bacteria using infrared radiation (IR) causes the cantilever to deflect in proportion to the infrared absorption of the bacteria, providing a nanomechanical infrared spectrum for selective identification. Analysis of individual cells at the subcellular level is important for understanding diseases and accelerating drug discovery. Nanoscale endoscopes allow minimally invasive probing of individual cell interiors. Several such instruments have been presented previously, but they are either too complex to fabricate or require sophisticated external detectors because of low signal collection efficiency. Here we present a nanoendoscope that can locally excite fluorescence in labelled cell organelles and collect the emitted signal for spectral analysis. Finite Difference Time Domain (FDTD) simulations have shown that with an optimized nanoendoscope taper profile, the light emission and collection was localized within ~100 nm. …

The prevention of social anxiety, performance anxiety, and social phobia via the combination of two generic drugs, diphenoxylate HC1 (opioid) plus atropine sulfate (anticholinergic) and propranolol HCl (beta blocker) was evaluated in mice through behavioral studies. A patent published on a September 8, 2011 by Benjamin D. Holly, US 2011/0218215 Al, prompted the research. The drug combination of diphenoxylate and atropine plus propranolol could be an immediate treatment for patients suffering from acute phobic and social anxiety disorders. 3-D paper-based microfluidic devices (micoPADs) are small and portable devices made out of paper that offer a promising platform for diagnostic applications outside of a laboratory. These devices are easy to use, low cost, require no power source, and capable of detecting multiple targets simultaneously. The work in this thesis demonstrated the ability of a 3-D paper-microfluidic platform to simultaneously detect 5 targets. Rubber cord stock was used in conjunction with an acrylic housing unit to apply pressure along the edge of the channel. Transcatheter aortic heart valve replacement (TAVR) is a procedure to replace a failing aortic valve and is becoming the new standard of care for patients that are not candidates for open-heart surgery [2]. However, this minimally invasive technique has shown to cause ischemic brain lesions, or “silent infarcts”, in 90% of TAVR patients, which can increase the patient’s risk for stroke by two to four times in future years [3]. Peripheral arterial occlusive disease (PAOD) occurs when there is narrowing or blockage of the peripheral arteries that carry blood to the extremities, most commonly the legs. However, traditional prosthetic fitting practices take a lot of time and cost a lot of money, making them inaccessible to millions of people around the world. StandUP Worldwide is an interdisciplinary project team devoted to creating low-cost prosthetic technologies for use around the world, especially in resource poor areas. They are currently developing a low cost, below-the-knee prosthetic kit that can be easily deployed in a resource-poor area. Immune checkpoint blockade has demonstrated great potential in activating antitumor immunity. Ipilimumab is a monoclonal antibody which targets cytotoxic T-lymphocyte antigen-4. CTLA-4 belongs to the CD28 class of receptors and is found on the surface of CD4+ and CD8+ T cells. CTLA-4 acts to suppress the immune system when bound to CD80 and CD86 receptors on antigen presenting cells. The objective of this study was to design citrate-coated gold nanoparticles conjugated with FITC-IgG, a fluorescent antibody, and to qualitatively and quantitatively measure the resulting fluorescent emission. Optical properties of the gold nanoparticles were measured at various stages to provide evidence of successful conjugation. The absorbance spectrum of the citrate gold nanoparticles was compared to that of the reaction mixture containing the gold nanoparticles and the FITC-IgG. A noticeable broadening of the absorption peak was observed at 519 nm indicating a surface modification of the gold nanoparticles. Cerebrovascular reactivity (CVR) is a key factor in regulating blood flow into the brain, and a marker for vascular disease. If the brain's regulatory system is not working, a patient may be in serious trouble. Testing of CVR is one method of assessing the brain's regulatory capabilities. Transcranial Doppler ultrasound (TCD) is one tool to measure CVR. In this method, carbon dioxide in the blood is transiently increased (such as with the holding of breath), and the resulting blood flow in the brain is measured. Transcranial Doppler (TCD) ultrasound is a technique involving the use of high frequency transmitters to measure intracranial blood flow. The brain is supplied by blood in an arterial anastomosis called the Circle of Willis. Using TCD ultrasound on the circle is difficult and requires practice and teaching. A functional model of the Circle of Willis could prove to be a valuable teaching tool. Through the use of AutoCAD and 3D printing software, an anatomically accurate model was created and set in gelatin phantom inside of a plastic skull. Three subject librarians and a data librarian, representing three departments and two libraries at a university, were awarded an NLM Informationist Supplement to support an interdisciplinary Research Group with an NIH grant. The Informationist Team developed a model to utilize the skills of multiple librarians to support the increasing number of interdisciplinary and interprofessional research groups at the university. Librarians routinely attended lab meetings and shared notes with each other to monitor researcher needs.

T-wave Alternans (TWA) in an electrocardiogram (ECG) has received considerable interest as a potential predictor of sudden cardiac death (SCD). However, large clinical trials have shown that while TWA has a very high negative predictive value, its positive predictive value is poor. Results of previous studies suggest that arrhythmia onset can be affected by the phase relationship of alternans of the depolarization and repolarization phase of the action potentials of the ventricles. A series of half-sandwich ruthenium complexes, two containing an arene face-cap and the other a thiacrown ether face-cap were synthesized to investigate the necessity of the arene for anticancer activity in this class of compounds. The complexes are formulated as [(h6-p-cymene)Ru(dmabTSC)Cl]PF6, [(h6-benzene)Ru(dmabTSC)Cl]PF6 (arene complexes), and [([9]aneS3(dmabTSC)Cl]PF6 (dmabTSC = dimethylaminobenzaldehye thiosemicarbazone). It was observed that none of the complexes showed good anticancer activity in vitro against HCT-116 and Caco-2 (colon adenocarcinoma) cells. Yet little is known about the effects of mechanical forces on cell behavior in the context of an organized tissue structure in three-dimensional cell-culture. The majority of patients requiring mechanical ventilation are over the age of 65 and advanced age is known to increase the severity of ventilator-induced lung injury (VILI) and mortality. However, the mechanisms which predispose aging ventilator patients to increased mortality rates are not fully understood.

Pulmonary edema is a hallmark of VILI and the severity of edema increases with age. Ventilation with conservative fluid management decreases mortality rates in acute respiratory distress (ARDS) patients, but has not been investigated in VILI. Selective Serotonin Reuptake Inhibitors (SSRIs) are the most frequently prescribed class of drugs worldwide and are implemented in the treatment of depression and other psychiatric disorders. SSRIs relieve depressive symptoms by modulating levels of the neurotransmitter serotonin in the brain. SSRIs block the function of the serotonin transporter, thereby increasing concentrations of extracellular serotonin. However, serotonin levels in the neurons of the brain only account for 5% while the remaining 95% is present outside the brain. Serotonin receptors and transporter are located on bone resident cells (mesenchymal stem cells (MSCs)), osteoblasts and osteoclasts, and serotonergic activity is believed to affect …
