Peripheral neuropathies are a set of common diseases that affect the peripheral nervous system, causing damage to vital connections between various parts of the body and the brain and spinal cord. Different clinical conditions are known to selectively impact various size nerve fibers, which often makes it difficult to diagnose which peripheral neuropathy a patient might have. The nerve conduction velocity diagnostic test provides clinically useful information in the diagnosis of some peripheral neuropathies. A greater understanding of bone materials would be beneficial in creating more accurate computer models and in the making of biomedical products involving bone. This study set out to determine whether cortical and trabecular bone are two separate materials, or whether they are the same material with a variance in porosity. To answer this question, samples were taken from different sections of the equine metacarpus, underwent densitometry analysis and were statistically analyzed. In the human genome, the SOST gene codes for a protein sclerostin. Sclerostin is an osteocyte-expressed negative regulator of bone formation. When the SOST gene is not coded, bone formation is reduced in individuals during skeletal maturation. This study utilizes nanoindentation methods to test for the mechanical properties of bones that both express and do not express the SOST gene. 100 transgenic murine femurs were obtained from Lawrence Livermore Labs in the form of 6 and 8 month SOST transgenic mice, 6 and 12 month SOST knockout mice, and wild type control littermates for each of the 4 age groups. …

The goal of this project is to eliminate electroosmotic flow (EOF) in a microfluidic chip. EOF is a naturally occurring phenomenon at the fluid-surface interface in microfluidic chips when an electric field is applied across the fluid. When isoelectric focusing (IEF) is carried out to separate proteins based on their surface charge, the analytes must remain in the separation chamber, and not migrate to adjacent features in the microfluidic chip, which happens with EOF.

The purpose of this thesis project was to demonstrate the ability to utilize the capabilities of aptamers so that they may act as capture reagents for paper microfluidic devices. Several characterization experiments were conducted as a precursor before the final experimentation was performed. Paper characterization, manufacturing protocols for printing and heating, as well as 3D chip fabrication were all performed and analyzed. The results confirmed that the control of fluid through a 3D microfluidic device based in nitrocellulose is possible.

Aptamers are ssDNA, RNA or polypeptide constructs that function like active antibodies. In response to an arterial occlusion, arteriogenesis occurs, causing smooth muscle cells to transition from a contractile to synthetic state. Also following an arterial occlusion, functional impairment was seen in the collateral circuit. An immunofluorescence protocol was developed in order to assess the impact of collateral enlargement (arteriogenesis) on smooth muscle phenotype at various time points. The highly porous Mn2O3-Ag nanofibers were fabricated by a facile two-step procedure (electrospinning and calcination) and then employed as the immobilization matrix for glucose oxidase (GOD) to construct an amperometric glucose biosensor. A notable enhancement of direct electron transfer between GOD and the electrode is observed at the Mn2O3-Ag-GOD modified electrode with a fast electron transfer rate constant. The biosensor also shows fast response to glucose, high sensitivity (40.60 μA×mM-1×cm-2), low detection limit (1.73 µM at S/N=3), low Km,app value and excellent selectivity. Ischemia Impairs Vasodilation In Skeletal Muscle Resistance Artery, Kyle Remington Struthers

Functional vasodilation in arterioles is impaired with chronic ischemia. We sought to examine the impact of chronic ischemia and age on skeletal muscle resistance artery function. To examine the impact of chronic ischemia, the femoral artery was resected from young (2-3mo) and adult (6-7mo) mice and the profunda femoris artery diameter was measured at rest and following gracilis muscle contraction 14 days later using intravital microscopy. Functional vasodilation was significantly impaired in ischemic mice (14.4±4.6% vs. 137.8±14.3%, p<0.0001 n=8) and non-ischemic adult mice (103.0±9.4% vs. 137.8±14.3%, p=0.05 n=10). In order to analyze the cellular mechanisms of the impairment, a protocol was developed to apply pharmacological agents to the experimental preparation while maintaining tissue homeostasis. Endothelial and smooth muscle dependent vasodilation were impaired with ischemia, 39.6 ± 13.6% vs. 80.5 ± 11.4% and 43.0 ± 11.7% vs. 85.1 ± 10.5%, respectively. From this data, it can be supported that smooth muscle dysfunction is the reason for the observed impairment in arterial vasodilation.

Hemoglobin can be measured on a variety of devices using different principles of operation. Noninvasive pulse CO-oximetry represents the latest development in hemoglobin measuring technology. The technology uses principles similar to pulse oximetry to measure total hemoglobin, oxyhemoglobin, reduced oxyhemoglobin, carboxyhemoglobin and methemoglobin. Similar to the introduction of pulse oximetry to the medical field, pulse CO-oximetry has been met with skepticism. Mammary epithelial cells are highly efficient secreting cells. With genetic engineering, the uses of these cells could be endless. Research is being conducted on these cells to determine their full potential to the biotech industry.

This paper investigates whether bovine epithelial mammary cells can survive in glutaraldehyde-treated gelatin tubes in an operating bioreactor. Many bioreactors were developed and tested to suit the needs of the cells. Procedures were created and carried out to ensure sterility of the bioreactors. Bovine mammary epithelial cells were implanted in the bioreactors and samples of their growth were taken over time.

The goal of this study was to compare the morphological and hemodynamic characteristics of normal and remodeled vascular networks in the mouse gracilis muscle. To this end, previously developed models to assess flows and vascular branching were used to assess the normal and remodeled microcirculatory networks. The analysis revealed that the average individual vessel flow rates for vessels of similar caliber and total volumetric flow rates in the networks do not change for vessels of the same caliber after remodeling. The humerus of a racing horse Thoroughbred is highly susceptible to stress fractures at a characteristic location as a result of cyclic loading. The propensity of a Thoroughbred to exhibit humeral fracture has made equines useful models in the epidemiology of stress fractures. In this study, a racing Thoroughbred humerus was simulated during training using a 3D finite element model in conjunction with a bone remodeling algorithm.
