Computational Methods For Identification Of Molecular Signatures, Weijun Yi

This work develops computational methods for identifying molecular signatures from high-throughput genomic data and for modeling long non-coding RNA (lncRNA) sub-cellular localization. The response of multiple myeloma to CB-6644, a selective RUVBL1/2 complex inhibitor with potential anti-tumor activity, is analyzed to identify drug-responsive pathways and molecular signatures. Conventional gene set enrichment analysis (GSEA) often excludes low-expression genes. Here, phenotype comparison is reformulated as a supervised machine learning problem: genes most informative for discrimination are first selected using a machine learning approach, and GSEA is then applied to these machine-learning derived gene sets. Quantitative Systems Pharmacology (QSP) refers to a mechanistic modeling approach used for assessing potential therapeutics by linking the molecular and cellular level mechanisms of the disease and therapeutic to the system-wide dynamics and clinical endpoints relevant to the disease. Although the field of QSP is still in its relative infancy, the mathematical methodology employed to predict the kinetics of these systems are not new and can be applied to a wide variety of systems. The increase in human activity in space has revealed several kinds of physiological challenges brought on by the absence of Earth's gravitational pull. NASA observed that during spaceflight, astronauts experience a 1-1.5% monthly loss in the mineral density of weight-bearing bones, with rehabilitation efforts on Earth often proving ineffective. This issue extends to muscles, the neuro vestibular system, the cardiovascular system, and vision. Cancer has emerged as a significant cause of mortality among astronauts, with studies up to 2017 showing that 41.3% of natural cause deaths among U.S. astronauts were due to cancer, primarily due to space radiation exposure. Metal-organic frameworks (MOFs) are organic crystalline hybrids that have been used for a variety of applications from catalysis to gas storage and drug delivery. Studies have shown that generally, while such frameworks are more biocompatible than similar aspect ratio nanostructures, they are also more prone to transformation and disintegration due to their flexible and fragile nature as resulted from the structure assembly and implemented structure-function relationships. Limb loss poses a significant challenge to mobility and the performance of daily activities. Currently, nearly half of the amputees using powered prosthetics abandon their device due to inadequate limb coordination and movement control abilities. In the healthy system, passive and neural dynamics facilitate the synchronized activation of muscles necessary for coordinated movement. However, in the amputee, the system is disrupted. The goal of this research is to use neural and mechanical dynamics to inform the design of prosthetic devices in order to restore intuitive control and coordinated limb movement.

Odor detection and identification are complex processes, and tasks that currently only animals do well. There is a pressing need for an electronic nose, or eNose, with good sensitivity, selectivity, and speed that mimics that ability. Food quality control operations, environmental sensing, occupational safety, and the defense sectors all require systems that can rapidly and reliably detect trace levels of volatile organic compounds. The goal of this work is to create a biologically inspired device which can accurately detect and identify odors at concentrations consistent with the most sensitive biological systems.

The operating temperature is one of the most important variables for gasifier operation. A higher temperature can shorten the service life of the refractory lining. A lower temperature can reduce the extent of carbon conversion and disrupt the flow of molten slag in a slagging gasifier. Therefore temperature is one of the most important variable in a gasifier. In slagging gasifiers, molten slag can penetrate into the refractory lining leading to refractory spallation, undesired downtime, and costly replacement.
