Characterization Of The Fatigue Threshold Behavior Of Uhmwpe, Bethany B. Smith, Anurag Roy, Robert O. Ritchie, Lisa A. Pruitt
Faculty Journal Articles
Ultra-high-molecular-weight-polyethylene (UHMWPE) has been the material of choice for bearings in total joint replacements (TJRs) for decades as a result of its excellent wear resistance, chemical inertness, energetic toughness, low friction, and biocompatibility. Utilization of this polymer in orthopedic devices requires oxidation, wear, and fatigue resistance. Balancing these important properties by tailoring processing techniques and modulating microstructural features has been an ongoing endeavor in the field. Research into the clinical applications of UHMWPE has primarily focused on the challenges of wear and oxidation while studies into the realm of fatigue have been more limited. Literature gaps exist in fully understanding …
Crevice Corrosion Mechanisms Of Cocrmo Alloys In Orthopedic Implants: Retrieval Analysis, Nano-Tribocorrosion And Cellular Responses, Hwaran Lee
All Dissertations
Mechanically assisted corrosion and non-mechanically (chemically) driven corrosion in modular junctions of orthopedic implants using cobalt-chromium-molybdenum (CoCrMo) alloys remain clinical concerns and may contribute to implant failure. The underlying corrosion mechanisms are not yet fully understood, leaving a critical research gap. We hypothesized that corrosion in modular junctions may be driven by aggressive local environments, including fretting, metal ion release (cobalt ion, Co2+), low pH and reactive oxygen species (ROS). This dissertation aims to (1) analyze corrosion modes and potential causes in modular junctions; (2) identify chemically driven corrosion modes on CoCrMo alloys using simulated inflammatory modular taper …

Synthesis And Characterization Of Magnetic Nanoparticles To Study Effective Magnetic Anisotropy For Biomedical And Catalytic Applications, Alexander Malaj
All Dissertations
This dissertation focuses on understanding how to tune the magnetic properties of nanoparticles through controlling the effective magnetic anisotropy (Keff), which is a key variable in determining a nanoparticle’s Néel relaxation time, which will dictate its magnetic behavior in various applications. In this work, magnetocrystalline anisotropy is tuned by synthesizing tri-metallic substituted ferrite (Fe3-x-yMnxCoyO4) nanoparticles with specific metallic compositions that were informed by computer simulations using density functional theory (DFT) to target magnetocrystalline anisotropy values. A drip synthesis was used to control the size and composition of the tri-metallic ferrites, which were revealed to be monodisperse and compositionally mixed by …

College of Graduate Studies: Theses & Dissertations
Abstract
This research explores the development of bio resin from linseed oil by the process of epoxidation and acrylation to produce acrylated epoxidized linseed oil (AELO). Lignin and cellulose biopolymers were added as reinforcement at varying weight fractions of 1 wt.%, 2.5 wt.%, and 5 wt.%, and their mechanical performance was compared against pure AELO. The formulations were printed using an Elegoo SLA 3D printer to create standardized test specimens like tensile bars, compression cylinders, and hardness blocks, which were all prepared according to the ASTM testing standard.
The results indicate that reinforcement effects depend on both filler type and …

Ultrasound Shear Wave Elastography: Development Of Tissue Models And Investigation Of Shear Wave Variability, Emily J. Miller
Dissertations, Master's Theses and Master's Reports
Ultrasound shear wave elastography (USWE) is an evolving and promising clinical tool for noninvasively measuring in vivo soft tissue biomechanical properties. Assumptions incorporated into the clinical workflow and technical limitations have created gaps between theoretical and clinically derived solutions. The heterogeneity of the fibrotic liver tissue, composition of the background, such as the presence of fatty liver tissue, and the preferred local orientation of the scarred fibrotic liver tissues embedded into the liver parenchyma, may contribute to the uncertainty in USWE measurements. This study aims to systematically investigate four cofounding factors (i.e., size, volume fraction, orientation of the fibrotic inclusions, …

During the COVID-19 global pandemic, a shortage of personal protective equipment (PPE) occurred. Manufacturers of these critical products were unable to keep up with the growing demands as the situation evolved. Most face shields, a common type of PPE, include a headband component to provide the appropriate fit for the user and curvature for the clear face shield attachment. To mitigate this shortage in the local region, the Portsmouth Naval Shipyard (PNSY) generated a computeraided design (CAD) file for the headband component and asked the University of New Hampshire (UNH) to manufacture it with all available 3D printers, including printers …
Shape Memory Alloy Capsule Micropump For Drug Delivery Applications, Youssef Mohamed Kotb
Shape Memory Alloy Capsule Micropump For Drug Delivery Applications, Youssef Mohamed Kotb
Theses and Dissertations
Implantable drug delivery devices have many benefits over traditional drug administration techniques and have attracted a lot of attention in recent years. By delivering the medication directly to the tissue, they enable the use of larger localized concentrations, enhancing the efficacy of the treatment. Passive-release drug delivery systems, one of the various ways to provide medication, are great inventions. However, they cannot dispense the medication on demand since they are nonprogrammable. Therefore, active actuators are more advantageous in delivery applications. Smart material actuators, however, have greatly increased in popularity for manufacturing wearable and implantable micropumps due to their high energy …

Mechanical Engineering ETDs
Lead zirconate titanate (PZT) has been a material of interest for sensor, actuator, and transducer applications in microelectromechanical systems (MEMS). This is due to their favorable piezoelectric, pyroelectric and ferroelectric properties. While various methods are available to deposit PZT thin films, radio frequency (RF) magnetron sputtering was selected to provide high quality PZT films with the added capability of batch processing. These sputter deposited PZT films were characterized to determine their internal film stress, Young’s modulus, composition, and structure. After characterization, the sputtered PZT samples were poled using corona poling and direct poling methods. As a means of comparison, commercially …

Additive Manufacturing Of Variable Contrast Computed Tomography Anatomical Phantoms Using A Single Feedstock In Fused Filament Fabrication, Cory J. Darling
LSU New Orleans Theses and Dissertations
Anatomical phantoms used in biomedical education and training benefit greatly from Fused filament fabrication’s (FFF) ability to rapidly produce complex and unique models. Current materials and methods used in FFF have limited ability to accurately produce phantoms that can mimic the radiological properties of multiple biological tissues. This research demonstrates that the CT contrast of FFF produced models can be modified by varying the concentration of bismuth oxide in acrylonitrile butadiene styrene (ABS) filaments and a tunable CT contrast that mimics the CT contrast ranging from fatty tissue to cortical bone using a single composite filament without introducing artificial image …

Dissertations and Theses
Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. By utilizing the photothermal behavior of graphene-hydrogel composites confined within micron-sized fluidic channels, patterned light illumination creates the parallel and independent actuation of chemical release in a group of fluidic ports. The …

Urine Volume Reduction During Long-Duration Cave Exploration By A Light-Weight And Portable Forward Osmosis System, Sebastian Engelhardt, Katey E. Bender, Jörg Vogel, Stephen E. Duirk, Francisco B. Moore, Hazel A. Barton
International Journal of Speleology
The preservation of caves is a challenge during long-duration cave expeditions where human waste can add significant nitrogen to the cave ecosystem. Since the removal of urine that accumulates during a multi-day caving trip is not always feasible due to weight and volume constraints, a light-weight and portable filtration system that is capable of reducing urine volume would be desirable. In this study we tested the Aquaporin Inside hollow fiber membrane in a forward osmosis (FO) setup to evaluate its capability to reduce urine volume while rejecting nitrogenous compounds using different draw solution chemistries and water recovery rates. As a …

Background: Reconstruction of bone fractures and defects remains a big challenge in orthopedic surgery. While regenerative engineering has advanced the field greatly using a combination of biomaterial scaffolds and stem cells, one matter of difficulty is inducing osteogenesis in these cells. Recent works have shown electricity’s ability to promote osteogenesis in stem cell lines when seeded in bone scaffolds; however, typical electrical stimulators are either (a) externally housed and require overcomplex percutaneous wires be connected to the implanted scaffold or (b) implanted non-degradable devices which contain toxic batteries and require invasive removal surgeries.
Objective: Here, we establish a biodegradable, piezoelectric …

Methods To Remotely Eliminate Biofilm From Medical Implants Using 2.4 Ghz Microwaves, Brett Glenn
Mechanical Engineering Undergraduate Honors Theses
Infections associated with biofilm growth are usually challenging to eradicate due to their high tolerance toward antibiotics [11, 12]. Biofilms often form on the inert surfaces of medically implanted devices [13]. No matter the sophistication, microbial infections can develop on all medical devices and tissue engineering constructs [12]. Related infections lead to 2 million cases annually in the U.S., costing the healthcare system over $5 billion in additional healthcare expenses [12].
Novel solutions to biofilm’s microbial colonization span the spectrum of engineering and science disciplines. Yet a practical solution still does not exist. The research presented here will explore a …
Effects Of Micro-Features On Cell Detachment From Poly(N-Isopropylacrylamide) Coated Polydimethylsiloxane Membranes, Luke Webel
Williams Honors College, Honors Research Projects
The tested hypothesis was that features on polydimethylsiloxane (PDMS) surfaces coated with a poly(N-isopropylacrylamide)/aminopropyltriethoxysilane or pNIPAAM/APTES thin film would accelerate cell detachment than the film coated on a unfeatured surface. This project tested samples with features generated by molds, wrinkling, and sandpaper roughened substrates. Surface feature generation methods were limited to mechanical means, and characterized by microscopy and strain rates. 50/50 mixtures of 1.5 wt.% pNIPAAM/ APTES were used to coat thin films (30-40 nm) on PDMS membranes by spin-coating, and the coated membranes were thermally annealed to chemically graft pNIPAAm/APTES on the membrane and their thermo-responsive property was assessed …

Developing Strategies To Toughen Bio-Inspired Adhesives, Narelli P. Narciso, Samuel Lee Huntington, Jonathan J. Wilker
The Summer Undergraduate Research Fellowship (SURF) Symposium
Mussels and other marine creatures adhere very well in underwater environments, having the ability to withstand the force of the sea. These animals have inspired synthetic biomimetic adhesives for wet systems, presenting potential for biomedical applications. However, most current commercial adhesives tend to be brittle, not resisting repetitive movements. This study assesses toughening strategies to improve the mussel-inspired adhesives’ ductility while maintaining its strength. The strategies included altering the polymer’s chemical structure by changing the percentage of polyethylene glycol (PEG) in the molecule and by adding fillers, such as calcium carbonate, silica and nacre - a calcium carbonate compound found …

Selective Serotonin Reuptake Inhibitors (SSRIs) are the most frequently prescribed class of drugs worldwide and are implemented in the treatment of depression and other psychiatric disorders. SSRIs relieve depressive symptoms by modulating levels of the neurotransmitter serotonin in the brain. SSRIs block the function of the serotonin transporter, thereby increasing concentrations of extracellular serotonin. However, serotonin levels in the neurons of the brain only account for 5% while the remaining 95% is present outside the brain. Serotonin receptors and transporter are located on bone resident cells (mesenchymal stem cells (MSCs)), osteoblasts and osteoclasts, and serotonergic activity is believed to affect …

Background: Due to their distinctive physicochemical properties, nanoparticles (NPs) have proven to be extremely advantageous for product and application development, but are also capable of inducing detrimental outcomes in biological systems. Standard in vitro methodologies are currently the primary means for evaluating NP safety, as vast quantities of particles exist that require appraisal. However, cell-based models are plagued by the fact that they are not representative of complex physiological systems. The need for a more accurate exposure model is highlighted by the fact that NP behavior and subsequent bioresponses are highly dependent upon their surroundings. Therefore, standard in vitro models …

The American Cancer Society predicts that 1,665,540 people will be diagnosed with cancer, and 585,720 people will die from cancer in 2014. One of the most common types of cancer in the United States is skin cancer. Melanoma alone is predicted to account for 10,000 of the cancer related deaths in 2014. As a highly mobile and aggressive form of cancer, melanoma is difficult to fight once it has metastasized through the body. Early detection in such varieties of cancer is critical in improving survival rates in afflicted patients. Present methods of detection rely on visual examination of suspicious regions …

Investigations into the use of gold nanorods (Au-NRs) for biological applications are growing exponentially due to their distinctive physicochemical properties, which make them advantageous over other nanomaterials. Au-NRs are particularly renowned for their plasmonic characteristics, which generate a robust photothermal response when stimulated with light at a wavelength matching their surface plasmon resonance. Numerous reports have explored this nanophotonic phenomenon for temperature driven therapies; however, to date there is a significant knowledge gap pertaining to the kinetic heating profile of Au-NRs within a controlled physiological setting. In the present study, the impact of environmental composition on Au-NR behavior and degree …

Fluorescence profiles of quantum dots (QDs) were characterized to select the ideal QDs for encapsulation in phospholipids for use as biomarkers to selectively adhere to cancer cells. QDs were synthesized and extracted 0, 30, 60, and 90 seconds after precursor compounds were mixed. These extractions were isolated by extraction time. Portions from each vial were coated in a zinc sulfide shelling procedure, leaving at least half of the QD solution unshelled. These samples were characterized over four days to monitor fluctuations in fluorescence. This was done utilizing an Ocean Optics spectrometer in conjunction with Spectra Suite software. The central wavelength, …

In the modeling of brain mechanics subjected to primary blast waves, there is currently no consensus on how many biological components to be used in the brain–meninges–skull complex, and what type of constitutive models to be adopted. The objective of this study is to determine the role of layered meninges in damping the dynamic response of the brain under primary blast loadings. A composite structures composed of eight solid relevant layers (including the pia, cerebrospinal fluid (CSF), dura maters) with different mechanical properties are constructed to mimic the heterogeneous human head. A hyper-viscoelastic material model is developed to better represent …

Cells sense and respond to chemical and physical stimuli through signal transduction pathways, which mediate cell proliferation, differentiation, migration, and survival. The cytokines interleukin-2 (IL-2) and interleukin-4 (IL-4) are key regulators of the adaptive immune system, particularly influencing the clonal expansion and differentiation of T cells. At least in culture, both synergistic and antagonistic effects of IL-2 and -4 co-stimulation have been reported; the antagonism, when observed, is thought to arise from the utilization of a common subunit shared by IL-2 and IL-4 receptors. We have sought to characterize IL-2 and IL-4 signaling at the level of intracellular pathways activated …
