Perfusion cell cultures typically achieve higher cell densities and generate higher volumetric productivities than fed-batch cultures due to continuous nutrient supply and spent media removal. Despite these advantages, perfusion cell cultures remain underutilized for licensed product manufacturing, primarily due to the lack of small-scale perfusion models for high-throughput process development and the complexities involved in media formulation capable of supporting high cell densities during long-term continuous operation. Abstract

This research explores the development of bio resin from linseed oil by the process of epoxidation and acrylation to produce acrylated epoxidized linseed oil (AELO). Lignin and cellulose biopolymers were added as reinforcement at varying weight fractions of 1 wt.%, 2.5 wt.%, and 5 wt.%, and their mechanical performance was compared against pure AELO. The formulations were printed using an Elegoo SLA 3D printer to create standardized test specimens like tensile bars, compression cylinders, and hardness blocks, which were all prepared according to the ASTM testing standard.

This study explored the impact of microwave pretreatment on the grinding efficiency of bastnaesite ore using a stirred mill. Bastnaesite ore was prepared using a staged crushing and sieving process, followed by microwave pretreatment in a specially designed microwave furnace system. Representative samples of the crushed ore underwent stirred mill grinding, with power draw measurements recorded and adjusted to reflect only the specific energy input required for grinding. Particle size distribution was analyzed periodically using laser particle size analysis. In 2022, ~288,000 American women were diagnosed with metastatic breast cancer which was the leading cause of patient mortality with ~43,000 American women succumbing to the disease. Women can be diagnosed with four different molecular subtypes of breast cancer: luminal A, luminal B, human epithelial growth receptor type 2 enriched (HER2+), and triple-negative breast cancer (TNBC). TNBC accounts for ~12% of all breast cancers, yet it has the worst outcomes. TNBC is highly invasive, with ~46% of TNBC patients developing metastatic disease. NASA observed that during spaceflight, astronauts experience a 1-1.5% monthly loss in the mineral density of weight-bearing bones, with rehabilitation efforts on Earth often proving ineffective. This issue extends to muscles, the neuro vestibular system, the cardiovascular system, and vision. Taxol, a formulation of paclitaxel (PTX), is one of the most widely used anticancer drugs, particularly for treating recurring ovarian carcinomas following surgery. Clinically, PTX is used in combination with other drugs such as lapatinib (LAP) to increase treatment efficacy. Delivering drug combinations with nanoparticles has the potential to improve chemotherapy outcomes. In this study, we use Flash NanoPrecipitation, a rapid, scalable process to encapsulate weakly hydrophobic drugs (logP in vitro. The tested hypothesis was that features on polydimethylsiloxane (PDMS) surfaces coated with a poly(N-isopropylacrylamide)/aminopropyltriethoxysilane or pNIPAAM/APTES thin film would accelerate cell detachment than the film coated on a unfeatured surface. This project tested samples with features generated by molds, wrinkling, and sandpaper roughened substrates. Surface feature generation methods were limited to mechanical means, and characterized by microscopy and strain rates. 50/50 mixtures of 1.5 wt.% pNIPAAM/ APTES were used to coat thin films (30-40 nm) on PDMS membranes by spin-coating, and the coated membranes were thermally annealed to chemically graft pNIPAAm/APTES on the membrane and their thermo-responsive property was assessed …

These animals have inspired synthetic biomimetic adhesives for wet systems, presenting potential for biomedical applications. However, most current commercial adhesives tend to be brittle, not resisting repetitive movements. Modification of the carbohydrate cyclodextrin increases the ability to encapsulate hydrophobic pharmaceutical molecules by forming a carrier with a hydrophobic core and hydrophilic exterior. Analysis of individual cells at the subcellular level is important for understanding diseases and accelerating drug discovery. Nanoscale endoscopes allow minimally invasive probing of individual cell interiors. Several such instruments have been presented previously, but they are either too complex to fabricate or require sophisticated external detectors because of low signal collection efficiency. Here we present a nanoendoscope that can locally excite fluorescence in labelled cell organelles and collect the emitted signal for spectral analysis. Immune checkpoint blockade has demonstrated great potential in activating antitumor immunity. Ipilimumab is a monoclonal antibody which targets cytotoxic T-lymphocyte antigen-4. CTLA-4 belongs to the CD28 class of receptors and is found on the surface of CD4+ and CD8+ T cells. CTLA-4 acts to suppress the immune system when bound to CD80 and CD86 receptors on antigen presenting cells. When levels of heavy metal ions such as nickel (Ni), copper (Cu), lead (Pb), Zinc (Zn), and Cobalt (Co) exceed the tolerance values for industrial wastewater, it can devastate the environment as well as the health of the individuals consuming the water. Heavy metal ions are not biodegradable and will accumulate in organic matter.(1) This accumulation leads to toxic levels in the blood and hence damage to nerve tissue, kidneys, the liver, and other vital organs. Chitosan is a derivative of chitin, a biopolymer found in the exoskeletons of crustaceans. Chitosan is under heavy investigation in the field of drug delivery due to its wide versatility. One such delivery mechanism is the use of hydrogels in cancer immunotherapy delivery. The viscosity of chitosan in its use as a hydrogel depends on the length of chitosan’s N-acetyl D-glucosamine chains, which in turn determines the molecular weight of chitosan. Controlled and localized delivery of Interleukin 12 (IL-12) is critical to effective and non-toxic cancer immunotherapy. Current delivery protocols for pre-clinical anti-tumor trials utilize solutions of chitosan, a mucoadhesive and bioeliminable biopolymer, to slow the release of IL- 12 into the tumor microenvironment. In order to further increase IL-12 retention, we have developed and characterized two chitosan-based in situ gelling hydrogels for IL-12 immunotherapy. One hydrogel, composed of heparin and chitosan, is herein described from initial conception through multiple stages of characterization including isolation of a range of suitable hydrogel formulations and observational physical and chemical properties.
