A Wearable Fiber-Free Optical Sensor For Continuous Measurements Of Cerebral Blood Flow And Oxygenation, Xuhui Liu Jan 2023

A Wearable Fiber-Free Optical Sensor For Continuous Measurements Of Cerebral Blood Flow And Oxygenation, Xuhui Liu

Theses and Dissertations--Biomedical Engineering

Wearable technologies for continuous monitoring of cerebral hemodynamics in freely behaving subjects not only advance our understanding of cognitive processing and adaptive behavior, but also provide vital information for diagnosis and therapeutic assessment of cerebral diseases associated with hypoxia/ischemia. Wearable near-infrared diffuse optical techniques have been used at the bedside to probe deep cerebral hemodynamics, including near-infrared spectroscopy (NIRS) for cerebral oxygenation measurement and diffuse correlation spectroscopy (DCS) for cerebral blood flow (CBF) measurement, respectively. However, most systems are relatively large and expensive, and use rigid fiber-optic probes that significantly constrain subject’s movement. A novel, wearable, fiber-free diffuse speckle contrast …

Small-Separation Speckle Contrast Optical Spectroscopy For Intraoperative Assessment Of Parathyroid Glands Viability During Thyroid Surgery, Connor Berger Dec 2022

Small-Separation Speckle Contrast Optical Spectroscopy For Intraoperative Assessment Of Parathyroid Glands Viability During Thyroid Surgery, Connor Berger

Symposium of Student Scholars

The parathyroid glands (PTGs) are often damaged during thyroid surgeries due to a lack of methods identifying PTGs and assessing their viability. Damage to PTGs can cause hypocalcemia, a deficiency of calcium in the body. This complication can lead to detrimental consequences with economic burden. The surgeon’s current method of viability assessment is qualitative and subjective. Our technical solution is to employ an optical technique called speckle contrast optical spectroscopy (SCOS) that noninvasively quantifies the blood flow index (Db) of biological tissues at deep tissue levels (>1cm). The goal of this project is to verify SCOS at small source-detector-separation …

Seeing The Big Picture: System Architecture Trends In Endoscopy And Led-Based Hyperspectral Subsystem Intergration, Craig M. Browning Dec 2022

Seeing The Big Picture: System Architecture Trends In Endoscopy And Led-Based Hyperspectral Subsystem Intergration, Craig M. Browning

Graduate Theses and Dissertations (2019 - present)

Early-stage colorectal lesions remain difficult to detect. Early development of neoplasia tends to be small (less than 10 mm) and flat and difficult to distinguish from surrounding mucosa. Additionally, optical diagnosis of neoplasia as benign or malignant is problematic. Low rates of detection of these lesions allow for continued growth in the colorectum and increased risk of cancer formation. Therefore, it is crucial to detect neoplasia and other non-neoplastic lesions to determine risk and guide future treatment. Technology for detection needs to enhance contrast of subtle tissue differences in the colorectum and track multiple biomarkers simultaneously. This work implements one …

Surface Laser Texturing Of Cocrmo Alloys, Gabriel Dharwadker May 2022

Surface Laser Texturing Of Cocrmo Alloys, Gabriel Dharwadker

Biomedical Engineering Undergraduate Honors Theses

Artificial orthopedic implants such as hip joints suffer from excessive wear due to excess friction between the polyethylene and cobalt-chromium-molybdenum surfaces of the device. This increase in friction causes a fraction of them to give out within ten years. Therefore, to minimize the probability of this happening, recent research has been conducted regarding laser surface texturing of the cobalt-chromium-molybdenum alloy components of artificial joints. These surface textures serve to mimic the native lubrication mechanisms of cartilage in natural joints, which, in a natural joint, lowers the friction. However, the production of these laser surface textures is still a field of …

Sequential Illumination In A Tomographic Microendoscopic Probe For Imaging Tumor Microvasculature, Zachariah Neumeier May 2022

Sequential Illumination In A Tomographic Microendoscopic Probe For Imaging Tumor Microvasculature, Zachariah Neumeier

Biomedical Engineering Undergraduate Honors Theses

Knowledge of colorectal cancer biology is improving how we approach cancer treatment. Specifically, the tumor microenvironment and abnormal angiogenesis are of particular interest. Optical methods are a prime candidate for research of the tumor microenvironment due to their ability to quantitively assess tissue structure and perfusion in real time. Particularly, the “transport scattering regime” has been identified as a method of obtaining high-resolution images and reflectance spectroscopy data; this light scattering regime has been demonstrated compatible with endoscopic imaging systems. In this study, a proof-of-concept optical imaging system is presented, capable of resolving absorbers within scattering turbid media using a …

The Effects Of Microgravity On Human Endothelial Cells In Relation To Deep Vein Thrombosis, Allie Burgess May 2022

The Effects Of Microgravity On Human Endothelial Cells In Relation To Deep Vein Thrombosis, Allie Burgess

Biomedical Engineering Undergraduate Honors Theses

Deep vein thrombosis (DVT) is a condition where blood clots form in one or more of the deep veins of the body, causing leg pain and/or swelling, potentially leading to pulmonary embolism and/or stroke. The incidence of DVT among astronauts serving on the International Space Station has been shown to be greater than that of the general population, calling into question the effects of microgravity on human thrombolytic processes. Additionally, microgravity conditions have been shown to cause muscular atrophy and a decrease in muscle strength and mobility. Because collagen composes a large portion of muscle tissue, this study also assessed …

3d Volumetric Mapping Of Tissue Properties Via A Catheter-Scale Microendoscope Imaging System, Andrew Stark May 2022

3d Volumetric Mapping Of Tissue Properties Via A Catheter-Scale Microendoscope Imaging System, Andrew Stark

Biomedical Engineering Undergraduate Honors Theses

The study of tumor microvasculature is an important area of interest for research and clinical communities; however, there are significant limitations in the clinical characterization of the tumor microenvironment. Optical methods offer quantitative real-time measurement of tissue structure and perfusion and can be miniaturized for deployment endoscopically into previously inaccessible locations. However, conventional optical methods (i.e., optical coherence tomography, white light endoscopy, etc.) gives limited information about tissue perfusion while the diameter of various optical probes along with the complexity of scanning mechanisms make it difficult to access certain areas. In this study, a new technique for characterizing the tumor …

Clot Analogs For The Development Of Improved Treatment Methods In Ischemic Stroke, Charles A. Rieth May 2022

Clot Analogs For The Development Of Improved Treatment Methods In Ischemic Stroke, Charles A. Rieth

Biomedical Engineering Undergraduate Honors Theses

According to the CDC, nearly 800,000 individuals experience stroke each year in the United States [1]. Greater than 70% of strokes are of ischemic etiology and involve the occlusion of key arteries in the cardiovascular system [2]. Tissue plasminogen activator (IV-tPA) is the current gold standard for thrombolytic approaches [3]; however, this therapeutic is only effective between 3 and 4.5 hours from the patient’s last know well [4]. As a result, less than 5% of acute ischemic stroke patients receive IV-tPA. In order to increase the viability of AIS treatment as a whole, it is essential that thrombolytic techniques are …

Paris, Alexis P. Applequist, Neil B. Ingels, Hanna K. Jensen, Drew E. Rodgers, Charles R. Cole, Jonathan F. Wenk, Morten O. Jensen

Biomedical Engineering Faculty Publications and Presentations

Current in vitro models of the left heart establish the pressure difference required to close the mitral valve by sealing and pressurizing the ventricular side of the valve, limiting important access to the subvalvular apparatus. This paper describes and evaluates a system that establishes physiological pressure differences across the valve using vacuum on the atrial side. The subvalvular apparatus is open to atmospheric pressure and accessible by tools and sensors, establishing a novel technique for experimentation on atrioventricular valves. Porcine mitral valves were excised and closed by vacuum within the atrial chamber. Images were used to document and analyze closure …

Nano-, Encapsulated Manganese Oxide (Nemo) Particles For Mri Enhancement In Microfluidic Tumor Models, Kasey Alexis Freshwater

Graduate Theses, Dissertations, and Problem Reports (ETD)

Breast cancer is among the deadliest cancers in women, with approximately 2.3 million new cases and 685,000 deaths reported globally in 2020 alone. Magnetic Resonance Imaging (MRI) is commonly used to diagnose, stage, and develop treatment plans for patients predisposed to breast cancer. However, current MRI-based screening of breast cancer has high false-positive rates up to 25%, leading to unnecessary invasive follow-up screening and testing. The gold-standard intravenous gadolinium (Gd)-based contrast agents (GBCAs) account for false-positive rates because they are always “ON”, providing a constant MRI signal that highlights both benign and malignant tumors. The work detailed in this thesis …

Time-Lapse Observation Of Sarcomeric Addition In Hypertrophic Models Based On A Tissue Like Cardiomyocyte Culture, Ailin Wei Dec 2021

Time-Lapse Observation Of Sarcomeric Addition In Hypertrophic Models Based On A Tissue Like Cardiomyocyte Culture, Ailin Wei

All Dissertations

Cardiac hypertrophy is the response of the heart to increased mechanical stress exerted on the heart. In a hypertrophic heart, the muscles elongate or/and thicken with cardiomyocytes growing sizes. It is assumed that cardiomyocytes elongate by adding sarcomeres in series; the cardiomyocytes thicken with the addition of sarcomeres in the parallel position.

In this study, a tissue-like culture model was achieved with step-like ICDs. Uniaxial static stretch has been applied to our unique culture model for studying mechanical-load-induced cardiac hypertrophy. Hypertrophic responses of these cardiomyocytes have been confirmed on transcriptional and translational levels in our tissue-like model. The two-photon excitation …

Diagnosis Of Melanoma Disease State From Patient Blood Samples Using Photoacoustic Flow Cytometry, Jacob Salvatore May 2021

Diagnosis Of Melanoma Disease State From Patient Blood Samples Using Photoacoustic Flow Cytometry, Jacob Salvatore

Electronic Theses and Dissertations

Approximately 1 in 50 Americans develop melanoma in their lifetime. Early detection of melanoma is pivotal to the survival of the patient, with a 99% survival rate for 5 or more years after an early diagnosis. Metastasis, or the spread of cancer, increases the chances of cancerous growth in other parts of the body. In this study, we propose the use of photoacoustic flow cytometry as a diagnostic of patient disease state, by closely monitoring the amount of circulating melanoma cells (CMCs) in a patient’s blood before and after chemotherapy treatments. Using this patient data, we were able to make …

Biomedical Engineering Undergraduate Honors Theses

Ulcerative colitis (UC) is a gastrointestinal, autoimmune disease that causes ulceration and inflammation of the colon with an incidence 10 out of every 100,000 people in North America and Western Europe. Though the exact etiology is uncertain, a number of studies have shown that inflammatory cells along with environmental factors, genetics, and lifestyle habits can contribute to the sustained inflammatory response. In order to determine the cellular mechanism behind relapse and remission of UC, researchers have frequently employed immunohistochemistry, western blotting and gene sequencing, but these destructive analysis methods require the removal of a sample, necessarily limiting these methods to …

High resolution, long-term monitoring of key biological analytes would improve patient outcomes by providing earlier detection of disease states and improved efficacy of treatment. One class of biosensors that have gained much attention in recent years are optical biosensors. Optical probes are attractive biosensors due to their noninvasive nature of detection, as certain light can pass through tissue, water, and blood. Single walled carbon nanotubes (SWNT) are a specific type of optical biosensor that fluoresce in the near infrared range of the electromagnetic spectrum and offer unparalleled spatial and temporal resolution. SWNT have been applied as biosensors in vitro, …

Dissertations and Theses

Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. By utilizing the photothermal behavior of graphene-hydrogel composites confined within micron-sized fluidic channels, patterned light illumination creates the parallel and independent actuation of chemical release in a group of fluidic ports. The …

Multiphoton Microscopy And Deep Learning Neural Networks For The Automated Quantification Of In Vivo, Label-Free Optical Biomarkers Of Skin Wound Healing, Jake D. Jones

Graduate Theses and Dissertations

Non-healing ulcerative wounds that occur frequently in diseases such as diabetes are challenging to diagnose and treat due to numerous possible etiologies and the variable efficacy of wound care products. With advanced age, skin wound healing is often delayed, leaving elderly patients at high risk for developing these chronic injuries. As it is challenging to discriminate age-related delays from disease-related chronicity, there is a critical need to develop new quantitative biomarkers that are sensitive to wound status. Multiphoton microscopy (MPM) techniques are well-suited for 3D imaging of epithelia and are capable of non-invasively detecting metabolic cofactors (NADH and FAD) without …

Computational Imaging Methods For Analysis Of Datscan Spect Images, Hae Sol Moon

McKelvey School of Engineering Graduate Student Theses & Dissertations

There is an important need to develop biomarkers to improve the diagnosis and assess the severity of Parkinson’s disease (PD). The potential to derive such biomarkers from quantitative dopamine transporter scan (DaT-Scan) single-photon emission computed tomography (SPECT) imaging, in particular the uptake of DaT in the caudate, putamen, and globus pallidus regions, is highly appealing as imaging is non-invasive and DaTScan is already used in the management of patients with PD. However, reliable quantification requires reliable segmentation of these regions in these images. Reliable segmentation is challenging due to the limited spatial resolution and high image noise in SPECT images …

Blood flow (BF) impacts the delivery of oxygen and nutrients to tissues and the removal of metabolic byproducts from tissues. Imaging of BF distributions helps characterize many diseases associated with tissue hypoxia/ischemia. The purpose of this study was to develop and validate a novel, noninvasive, noncontact, high-density camera-based speckle contrast diffuse correlation tomography (scDCT) device for use in both animal and human studies. The scDCT uses a galvo-mirror to remotely deliver the focused point near-infrared light to source positions and a sensitive 2D camera to quantify spatial diffuse speckle fluctuations, resulting from the movement of red blood cells in deep …

Quantitative Analysis Techniques For Assessing Organelle Organization And Dynamics In Individual Cells, Isaac Vargas Dec 2019

Quantitative Analysis Techniques For Assessing Organelle Organization And Dynamics In Individual Cells, Isaac Vargas

Graduate Theses and Dissertations

In biomedical optics and microscopy, the organization and morphology of organelles have been widely studied. In spite of novel imaging techniques, there is still a lack of quantitative tools to easily measure cellular characteristics from image data. Previous studies have explored multiple approaches to assess organelle organization and alignment, resulting in complicated and extensive algorithms that are both subject to multiple steps of image processing and influenced by non-cellular artifacts. In this thesis, a technique called the Modified Blanket Method (MBM) is introduced to quantify organelle organization through measurements of fractal dimension (FD) on a pixel-by-pixel basis. With the use …

Vector Flow Imaging In Pediatric Cardiology - Extracting And Validating Data, Mason Belue May 2019

Vector Flow Imaging In Pediatric Cardiology - Extracting And Validating Data, Mason Belue

Biomedical Engineering Undergraduate Honors Theses

In the field of bedside cardiac diagnostic imaging, Doppler Ultrasound (DU) is the gold standard for diagnosing heart conditions. The largest benefit of DU is its ability to noninvasively image cardiac flow and allow the estimation of blood velocity and quantification of anatomical disease. However, to get correct velocity estimation, the position of the transducer in relation to the flow field needs to be known. This is the problem of angle/direction dependency and limits DUs accuracy when imaging in areas where perfect alignment or exact position of the transducer in relation to flow field is not possible or known, such …

Seeing Eye To Eye: A Machine Learning Approach To Automated Saccade Analysis, Maigh Attre May 2019

Seeing Eye To Eye: A Machine Learning Approach To Automated Saccade Analysis, Maigh Attre

Honors Scholar Theses

Abnormal ocular motility is a common manifestation of many underlying pathologies particularly those that are neurological. Dynamics of saccades, when the eye rapidly changes its point of fixation, have been characterized for many neurological disorders including concussions, traumatic brain injuries (TBI), and Parkinson’s disease. However, widespread saccade analysis for diagnostic and research purposes requires the recognition of certain eye movement parameters. Key information such as velocity and duration must be determined from data based on a wide set of patients’ characteristics that may range in eye shapes and iris, hair and skin pigmentation [36]. Previous work on saccade analysis has …

Dual-Display Laparoscopic Laser Speckle Contrast Imaging For Real-Time Surgical Assistance, Jaepyeong Cha, Corey Zheng, Lung Wai Lau

Pediatrics Faculty Publications

Laser speckle contrast imaging (LSCI) utilizes the speckle pattern of a laser to determine the blood flow in tissues. The current approaches for its use in a clinical setting require a camera system with a laser source on a separate optical axis making it unsuitable for minimally invasive surgery (MIS). With blood flow visualization, bowel viability, for example, can be determined. Thus, LSCI can be a valuable tool in gastrointestinal surgery. In this work, we develop the first-of-its-kind dual-display laparoscopic vision system integrating LSCI with a commercially available 10mm rigid laparoscope where the laser has the same optical axis as …
