Enhancing Wound Healing With Synergistic Dual-Drug Electrospun Roflumilast And L-Arginine Loaded Pla/Pva Nanofibers Through Fabrication, Optimization, And In Vivo Assessment, Samar A. Salim

Current trends in improved wound management emphasize the development of dual-drug-loaded electrospun nanofibrous scaffolds. Herein, electrospun dual-drug loaded PLA/PVA nanofibrous scaffolds, incorporating roflumilast, a selective phosphodiesterase-4 inhibitor with anti-inflammatory activity, and L-arginine, a precursor to nitric oxide with stimulating activity towards wound healing and tissue regeneration, were developed. A dual-spinneret electrospinning process enabled the coloading of drugs in PLA and PVA phases. Scaffolds were characterized by SEM, FTIR, and XRD, showing consistent fiber morphology, and amorphous drug formation. FTIR analysis was performed to confirm drug–polymer compatibility and successful incorporation of roflumilast and L-arginine within the PLA/ PVA nanofibrous matrix. Swelling …

Bioceramics are important biomaterials in biomedical engineering because of their biocompatibility, bioactivity, osteoconductivity, and structural resemblance to actual bone tissue. In recent years, materials science and nanotechnology have enabled the use of bioceramics in bone regeneration, dental restoration, tissue engineering, drug delivery systems, and implantable medical devices. This comprehensive review covers advances in bioceramics, including calcium phosphates, hydroxyapatite (HAp), tricalcium phosphate, bioactive glasses, zirconia, alumina, and multifunctional ceramic nanocomposites. Priority is given to techniques such as additive manufacturing, 3D printing, sol-gel processing, electrospinning, and nanostructuring to improve mechanical strength, porosity, bioactivity, and cell interactions. Recent advances in the field of biomaterials have suggested that cells cultured on substrates resembling the native tissue mechanical properties, matrix and growth factor composition, and topography can adopt phenotypes that more closely resemble the in vivo tissue compared to cells cultured on non-mimetic constructs. Understanding the effect of culture substrate on in vitro tissue formation is important for bioengineering applications that include mechanistic studies of healthy tissue function and development of disease models. After injury, wound healing is a complex sequential cascade of events essential for the proper recovery of the wound without the scar formation. Nitric oxide (NO) is a small, endogenous free-radical gas with antimicrobial, vasodilating and growth factor stimulating properties. NO has wide biomedical application especially in wound healing however, its usability is hindered due its administration problem as it is highly unstable.

Hydrogels are useful in wound healing, drug delivery, and tissue engineering applications, but the available methods of injecting them quickly and noninvasively are limited. The medical industry does not yet have access to an all-purpose device that can quickly synthesize hydrogels of different shapes and sizes. Many synthesis procedures that have been developed result in the formation of amorphous hydrogels. While generally useful, amorphous hydrogels exhibit limited capability in tissue engineering applications, especially due to their viscous properties. The purpose of this study was to determine the in vitro response of cells critical to the wound healing process in culture media supplemented with a lyophilized preparation rich in growth factors (PRGF) and Manuka honey.
